Abstract

The research on physiological and reproduction status of Bawean deer (Axis kuhlii) in its habitat has been conducted, to understand and to find out as a basic information on Bawean deer (Axis kuhlii) in its habitats abaseline data for wildlife conservation efforts. The deer is categorized as an endangered animal, therefore, more attention was given toward Bawean deer conservation. Habitat changes, loss of habitat, fragmentation and illegalhunting might caused the wild animals become more marginalized and the populations have been declined. Therefore, it is needed to have research as an effort to save Bawean deer in its natural habitat. This research activity covered monitoring on physiological and reproduction status, and also the examination of deer's feces samples was parasite coprological data that serve as a basic consideration data in conservation and determination of Bawean deers health in the nature and/or conservation. The research was done in Bawean Island, Gresik and East Java, by taking faces of Bawean deers in the nature and conservation. Data gathering of physiological and reproduction status were done by examining deer's condition, either directly in conservation or in its habitat, and also by interviewing people around the forest. The data on physiological status, behavior monitoring, reproduction status, faces samples, blood, food analysis, and interview result were analyzed descriptively. Some worm eggs were found during feces examination, those were Strongyl, Strongyloides sp, Trichuris sp, Fasciola gigantica and Oocysta coccidia. The result showed that the same parasites were found in the cattle and goat raised by people around in the vicinity of the forest, meaning, there was an interaction between wildlife andlivestock. According to the result, it is needed to monitor the physiological status of Bawean deers routinely since Bawean deers is classified as endangered species.

Abstract

       Research aimed to identify the most effective concentration of five extract types (EKBMCP, EKPAS, EDMSP, EBJCP, dan EDMCO) as vermicidal anthelmintic against          H. contortus in-vitro. Research was consisted of seven groups, among them five groups were considered as treatment groups that named based on the extract type (EKBMCP, EKPAS, EDMSP, EBJCP, dan EDMCO). Each treatment group was consisted five subgroups of etnopharmacological concentration : 0,5%, 1,5%, 2,5%, 3,5% dari 0,2 g/mL. Other two group were regarded as a negative control by distilled water treatment, and a positive control by pyrantel pamoat 5%. Every single treatment group was quadruplet and exposure each concentration were performed in quadruplet with variation in exposure times i.e. : 1, 3,             5, and 7 hours. The research focused on several variables that were the percentage of live and dead H. contortus in the treatment groups, extract concentration, and treatment period by using selected etnopharmacological extract. The effective vermicidal anthelmentic concentration of each treatment was analyzed descriptively. The results showed that five etnopharmacological extract types that were EKBMCP, EKPAS, EDMSP, EBJCP, dan EDMCO had a potency as vermicidal anthelmentic. Among those treatment, EKPAS and EBJCP had the lowest concentration (2,5%) to produce optimum vermicidal effect (100%) with period five hours exposure. Other treatment groups with EKBMCP and EDMSP needed 3,5% concentration with exposure period seven hours to produce optimum vermicidal effect, respectively. Another treatment group with EDMCO needed 3,5% concentration with exposure time seven hours to produce optimum vermicidal effect.

Abstract

Gastrointestinal Nematodes are one among cause decrease production efificiency in goat and sheep in Indonesia, especially parasite of abomasum namely Haemonchus contortus. This parasite causes disease of Haemonchosis causing anemia, and causes mortality to young goat or sheep. Loss of economics caused by high morbidity and  mortality this parasite so that can improve the expense of prevention and medication of this disease. This research aim was to evaluate ovicidal and larvicidal apus bamboo leaf infusion (Gigantochloa apus) against Haemonchus contortus by in vitro. Examination of apus bamboo leaves infusion to seven concentrations namely : 1.2; 1.0; 0.8,; 0.6; 0.4; 0.2;0.1; 0.05 and 0 mg / ml as negative control. Positive control uses Albendazole with concentration 0,025 mg / ml. The highest concentration 1.2 mg / ml apus bamboo leaves infusion   inhibited 92.35% egg hatching ( LC 0.49 mg / ml ) and have larvicidal activity equal to 90% ( LC50 0,47mg / ml). The concentration 1.2 mg / ml also have similar effect with positive control which use Albendazole. Phytochemical apus  bamboo leaf analysis of the highest concentration revealed the presence  tanin condensed, saponins, and alkaloids.

Abstract

A. galli infection caused high economic losses and decreased the amount and quality of egg production. The existence of anthelmintic resistance lead to the development of research on alternative treatments for A. galli. This aim of the research was to know LC50 of Areca catechu on infestation of A. galli. In this research A. galli was taken from a chicken slaughterhouse in Terban, Yogyakarta, and was divided into 8 groups for in vitro research. Group I was treated with an 10% infusion of A. catechu; group II was treated with 12.5% infusion of A. catechu; group III was treated with a 15% infusion of A. catechu; group IV was treated with a 17.5% infusion of A. catechu; group V was treated with 20% infusion of A. catechu; group VI was treated with a 22.5% infusion of A. catechu; group VII was treated with 25% A. catechu infusion and group VIII as negative control (0.9% NaCl). A. galli mortality is recorded every one hour until worm mortality is 100%. The results were then analyzed using the Reed and Muench method. Based on the in vitro test the infusion of Areca catechu against Ascaridia galli can cause death in worms with an LC 50 calculation of 21. 18%.

Abstract

Abstract Surra's disease is caused by Trypanosoma evansi parasite has been established as one of the strategic infectious animal diseases. Drug resistance in this case is one of the major challenges in handle and control them. The aim of this study is to evaluate the provision drug resistance diminazene aceturate (Tryponil®) on Trypanosoma evansi isolate from Pemalang and Brebes Central Java province with in vivo test in mice. Total 50 mice, BALB / c strain, male, 2 months, body weight ± 30 gram are obtained from LPPT-UGM, adapted for one week. Mice were divided into 10 groups consist of 5 each. Each mouse was infected with Trypanosoma evansi by intraperitonial route. Treatment was given when mice had reached the level of parasitemia 108 – 109 trypanosoma / mL of blood this was predicted 24 hours post-infection (Eisler et al., 2001). The administration of the drug tryapanosidal was done intraperitonial with doses 1mg/kg, 3mg / kg, 5 mg / kg and 7mg / kg. Observation of parasitemia did every 2 times in one week till 60 days of observation. Parasitemia observation was performed using 3 techniques. The first method was native examination used a microscope, if the negative results would be followed by MHCT (Microhaematocrit centrifugation Technique) and BCT (Buffy Coat Technique) according to OIE (2012). Data obtained from the treatment group were the level of parasitemia, the number of deaths and the number of live mice from each test dose. The results are analysed by standard logit or probit. The results of this study showed the effects of the drug Dimianzene aceturate on both isolates varied. On Brebes Isolate was effective at doses of 7 mg / kg BW (100%) and 5mg / kg BW (80%), whereas in the effective dose Pemalang isolate at 3 mg dose / kg BW (80%), 5 and 7 mg / kg BW (100%). While at the lowest dose of 1 mg / kg obtained a level of effectiveness of 0% in both isolates. It could be concluded that both isolates have different pathogens and indicate resistance subpopulation to diminazene aceturate.Keywords : diminazene aceturate, in vivo, resistance, Trypanosoma evansi.

Abstract

The study evaluates in vitro effects of Areca catechu crude aqueous extract  as anthelmintics on Ascaridia galli adult mortality rate and morphometry. Naturally infected chickens were collected from slaughtered chickens at local slaughterhouses in Yogyakarta. Chicken’s intestines were carefully examined and transported to the Parasitological Laboratory of Gadjah Mada University, Yogyakarta. Ascaridia galli were collected from intestines (including duodenum, jejunum, and ileum) and put into a petri dish containing 0.62% saline water. There were 60 A. galli in each concentration of Areca catechu crude aqueous extract. Investigation on mortality rate of adult worms at various concentration and observation as well as differences in A. galli morphometry and mortality rate were analyzed using analysis Anova. The figures of the parasites or the parts of the parasites were captured using camera Lucida and then they were measured using both micrometer and curvimeter. The morphology of the parasites was identified to find out their morphometric characteristics.The results of morphometric observation of the A. galli showed that there was a significant difference in the body width and the vulva length of the females and in the body width, the esophageal length and the esophageal width of the males.This research disclosed that Areca catechu crude aqueous extract significantly affected the mortality rate of adult A.galli at various concentration. This research observed that the best concentration to kill the adult worms was 25% Areca catechu crude aqueous extract.

Abstract

Infestasi parasit pada ikan dalam jumlah besar dapat menyebabkan kerugian ekonomi yang cukup signifikan. Parasit dari jenis Trematoda monogenean dan protozoa mendominasi infestasi tersebut.  Penelitian ini dilakukan untuk mendapatkan status kejadian infestasi parasit pada ikan mas biasa (Cyprinus carpio). Sebanyak 200 ikan mas (Cyprinus carpio) dikumpulkan dari kolam-kolam di Sleman dan Magelang, Jawa Tengah Indonesia. Metode pemeriksaan menggunakan metode wet mount. Parasit yang ditemukan sebagai berikut: Ichthyopthirius multifilis 41% Dactylogyrus  spp. 44,5% dan Gyrodactylus spp. 25%. Kejadian insidensi parasit sebagai berikut: Ichthyopthirius multifilis 1496/82, Dactylogyrus  spp. 119/37 dan Gyrodactylus spp. 2/2. Kondisi kolam yang buruk dan pergantian musim di Indonesia merupakan faktor yang sangat berpengaruh terhadap kejadian infestasi parasit pada ikan mas (Cyprinus carpio).

Abstract

Culex tritaeniorhynchus and Culex vishnui are medically essential mosquitoes that transmit the Japanese encephalitis (JE) virus. There is less information about the recording data and research due to genetic character differences among them. The objective of this study was to examine the genetic variation of Cx. tritaeniorhynchus and Cx. vishnui in 3 sites of Central Java using polymerase chain reaction randomly amplified polymorphic DNA (PCR-RAPD). The study was done in January to November 2017 in Pekalongan city, Pekalongan regency, and Semarang regency. Adult female mosquitoes collected by human bite method. DNA of ten Cx. tritaeniorhynchus samples and fifteen samples of Cx. vishnui purified using DNA extraction kit. Furthermore, PCR amplification was conducted with 5 RAPD primers (OPA 11, 12, 15, 16, and 20) and would run into 2% gel electrophoresis for 45 minutes. Cluster analysis was using MVSPTM software (version 3.1). The results showed 213 genetic characters of Cx. vishnui, while 142 characters shown by Cx. tritaeniorhynchus. The dendrograms showed three distinct groups of Cx. vishnui from 2 sites of Pekalongan and one site of Semarang, while Cx. tritaeniorhynchus showed two distinct groups, which were 1 group from Pekalongan and 1 group from Semarang. Low genetic similarity (<10%) shown Cx. vishnui from Pekalongan city and Pekalongan district, and there was no genetic similarity in Cx. tritaeniorhynchus from Pekalongan and Semarang. It concluded that the polymorphism of Cx. tritaeniorhynchus and Cx. vishnui reached 100%. ANALISIS FILOGENETIK CULEX TRITAENIORHYNCHUS DAN CULEX VISHNUI VEKTOR VIRUS JAPANESE ENCEPHALITISNyamuk Culex tritaeniorhynchus dan Culex vishnui memiliki peran penting di bidang medis terutama dalam penularan virus Japanese  encephalitis (JE). Sampai saat ini data dan riset tentang karakter genetik vektor JE masih sangat terbatas. Penelitian ini bertujuan menjelaskan variasi genetik Cx. tritaeniorhynchus dan Cx. vishnui di 3 lokasi di Jawa Tengah berdasar polymerase chain reaction randomly amplified polymorphic DNA (PCR-RAPD). Studi ini dilakukan dari bulan Januari sampai November 2017 di Kota Pekalongan, Kabupaten Pekalongan, dan Kabupaten Semarang. Metode human bite digunakan untuk koleksi nyamuk. Ekstraksi DNA nyamuk dilakukan pada 10 ekor Cx. tritaeniorhynchus dan 15 ekor Cx. vishnui menggunakan kit ekstraksi DNA. Selanjutnya, diamplifikasi dengan 5 macam primer RAPD (OPA 11, 12, 15, 16, dan 20), serta dielektroforesis pada 2% agar selama 45 menit. Analisis klaster dilakukan menggunakan program MVSPTM (versi 3.1). Ditemukan 213 dan 142 karakter genetik masing-masing pada Cx. vishnui dan Cx. tritaeniorhynchus. Analisis dendogram menunjukkan 3 grup yang berbeda untuk Cx. vishnui, sedangkan untuk Cx. tritaeniorhynchus terdapat 2 grup yang berbeda, yaitu 1 grup dari Pekalongan dan 1 grup dari Semarang. Similaritas genetik yang rendah (<10%) ditunjukkan Cx. vishnui dari Kota Pekalongan dan Kabupaten Pekalongan, bahkan tidak ada persamaan genetik pada Cx. tritaeniorhynchus dari Pekalongan dengan Semarang. Disimpulkan bahwa polimorfisme Cx. tritaeniorhynchus dan Cx. vishnui mencapai 100%.

Abstract

Penelitian ini bertujuan mengetahui tingkat prevalensi Toxoplasma pada kucing dan menyusun basis epidemiologis pada kucing yang sangat penting dalam peranannya sebagai hospes definitif toksoplasmosis. Sampel serum darah dan feses kucing digunakan dalam penelitian ini. Metode pemeriksaan menggunakan card agglutination Toxoplasma test (CATT) Pastorex™ Toxo kit dan metode sentrifus. Data yang dip eroleh dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa tingkat prevalensi Toxoplasma dengan pemeriksaan serum darah kucing sebesar 6,8% dan pemeriksaan feses sebesar 9,4%. Infeksi toksoplasmosis yang terjadi pada kucing secara umum dari pemeriksaan klinis tidak mempunyai gejala yang spesifik.

Abstract

This present project was carried out to determine the effect of apus bamboo (Gigantochloaapus )leaves infusion to mortality rate and morphometry of Haemonchus contortus adult worms in vitro. The formulated tools were collected through camera, 3 ml syringe, object glass, deck glass, mortar, drip dropper, waterbath, microscope, stopwatch, counter check, oven, erlenmeyer flask, electric scale, surgical scissor, and pipette. The allocated materials used in this studywere bamboo leaves infusion, Haemonchus contortus adult worms, aquadestilata, ethanol and NaCl 0.62%. The research was firstly concernedwith making 0.1% and 1% apus bamboo leaves infusion. The samples were obtained from Haemonchus contortus collection. Adult worms were obtained directly from abomasum of naturally infected goats that were cut at Animal Slaughter House (RPH). To investigate the matter, mortality rates of adult worms at various doses and times of observation as well as differences in Haemonchus contortus morphometry were analyzed using ANOVA. The findings disclosed that apus bamboo leaves infusion demonstrated a significant difference in the mortality rate of adult worms Haemonchus contortusat various doses and times of observation. This report presented the findings of research that the best dose to increase the mortality of worms was 1% bamboo leaves infusion. Moreover, apus bamboo leaves infusion affected the morphometry of Haemonchus contortus adult worms, especially in body length, cervical papilla width, and spicules length in males, and body length, cervical papilla width, and vulvar length in females.
