Abstract

Isolation of Yeasts from Sugarcane Stems and Their Identification Based on Internal Transcribed Spacer Sequences ABSTRACTFermentative yeasts used in food, health, and energy industries need to be explored to discover their potential. The purpose of this study was to obtain fermentative yeast isolates from sugarcane stems and subsequently to undertake morphological, biochemical, and molecular identification. The isolation of epiphytic and endophytic yeasts was carried out by spread plate method using sugarcane soak water and sugarcane juice on potato dextrose agar (PDA) and yeast-glucose-peptone (YGP) agar media. Morphological identification was based on macroscopic and microscopic observations. Biochemical identification was performed using carbohydrate fermentation and 50%-glucose media tests. Selected isolates were identified molecularly using Internal Transcribed Spacer (ITS). Seven yeast isolates were obtained, of which isolate Ed 1B was selected. Isolate ED 1B was of round colonies, creamy white colour, shiny, embossed, and wavy appearance, ovoid cell shape with a cell diameter of 4.74 µm. It had budding cells, was able to ferment glucose and sucrose (but not lactose), and grew on 50 %-glucose media. Results of BLAST showed that isolates Ed 1B had 99% homology with Kodamaea ohmeri.Keywords: isolation, ITS, molecular identification, Saccharum officinarum L., yeast ABSTRAKKhamir fermentatif yang digunakan dalam industri pangan, kesehatan dan energi perlu dieksplorasi untuk mengetahui potensinya. Tujuan penelitian ini adalah untuk memperoleh isolat khamir fermentatif dari batang tebu dan untuk kemudian diidentifikasi secara morfologi, biokimia dan molekuler. Isolasi khamir epifit dan endofit dilakukan dengan metode cawan sebar dari air rendaman tebu dan jus tebu pada media potato dextrose agar (PDA) dan yeast-glucose-peptone (YGP). Identifikasi morfologi berdasarkan pengamatan makroskopis dan mikroskopis. Identifikasi biokimia menggunakan uji fermentasi karbohidrat dan uji media glukosa 50%. Isolat terpilih diidentifikasi molekuler menggunakan Internal Transcribed Spacer (ITS). Hasil isolasi memperoleh 7 isolat khamir. Satu isolat terpilih (Ed 1B) didapatkan dan memiliki ciri-ciri koloni bulat, putih krem, mengkilap, timbul, bergelombang, bentuk sel ovoid dengan diameter sel 4,74 µm, memiliki budding cell, mampu memfermentasi glukosa dan sukrosa, tidak memfermentasi laktosa, serta tumbuh pada media glukosa 50%. Hasil BLAST menunjukkan bahwa isolat Ed 1B memiliki homologi 99% dengan Kodamaea ohmeri.Kata Kunci: identifikasi molekuler, isolasi, ITS, khamir, Saccharum officinarum L.

Abstract

Lentigo maligna (LM) is a subtype of melanoma in situ, which can evolve into lentigo maligna melanoma (LMM) if treated inadequately. LM and LMM are usually found on chronically sun damaged skin such as the face (cheek and nose) of the elderly on seventh or eight decades. Clinical manifestation of LM may be quite subtle, so early diagnosis is difficult to perform.The treatment of LM and LMM are challenging due to ill-defined clinical margin, predilection on the face with great size, and preponderance of the elderly, which are potential for recurrency and progressiveness from LM into LMM.

Abstract

Background: Ulcus vulvae acutum, also known as Lipschütz ulcer, is a rare condition characterized by multiple, painful, and acute genital ulceration in young women without venereal infections, associated with systemic symptoms like malaise, fever, and inguinal lymphadenopathy. Case Illustration: A 24-year-old woman developed multiple and painful vulvar ulcers that recurred for almost a year. The patient was primarily diagnosed and treated as genital herpes in several hospitals with only slight improvement and no complete healing. The patient came to our hospital for examinations to rule out sexually-transmitted infection (STI) (genital herpes, syphilis, HIV, and chancroid) and possibility of Behçet’s. The patient was treated with a combination of 2.5% lidocaine and 2.5% prilocaine creams for 1 week, 2% fusidic acid ointment for 1 week, with no significant progression. Later, the therapy was changed to 0.05% clobetasol propionate ointment for 10 days. Complete healing occurred in about 10 weeks with no scarring. Discussion: Diagnosis of ulcus vulvae acutum in this patient was made by excluding other differential diagnoses. Conclusion: Ulcus vulvae acutum is a difficult diagnosis, and clinician should be aware of this entity especially when encountering young woman without previous sexual experience problem with ulcer.

Abstract

Background: Being the most commonly found sexually-transmitted infection (STI), anogenital warts are mainly caused by human papilloma virus (HPV) type 6 and 11. These infections are often recurrent, leading to loss of time and medical expenses. Various therapeutic modalities can be chosen singularly or in combination with varying results. Treatments with less responsive results usually occur when acquired comorbidities are present, such as HIV infection. Case Illustration: A 28-year-old homosexual male was reported to have a recurrent perianal wart for the duration of eight months. He was also diagnosed with AIDS and pulmonary tuberculosis. Initially, patient was treated with 25% podophyllin tincture, but there was no improvement. Patient was known to still practicing high-risk sexual behavior. After six treatments of TCA, the warts shrank gradually and no new warts had grown. Discussion: It was found that there is a strong relationship between high incidences of genital warts with reduced immune function due to by HIV infection. Patients infected with HIV will often experience recurrence and needs longer treatment. In this case ARV treatment is given and followed by increase in CD4 count, thereby increasing the effectiveness of the treatment of perianal warts. Conclusion: As recommended by clinical guideline in Indonesia, 90% TCA is proven to be effective in treating recurrent anogenital warts in this case. It is important to counsel the patient regarding the sexual behavior for him and his sexual partner, as it will affect the patient’s prognosis significantly.

ABSTRACT This study means to decide how huge the impact of Professional Ethics and Gender on Auditor Professionalism. This study utilizes quantitative techniques. Terlebih lagi, 38 penelitian ini menggunakan persamaan alasan pemeriksaan. Pengujian spekulasi eksplorasi menggabungkan beberapa tes kekambuhan yang dapat diukur: uji-t, uji f atau tidak lengkap, dan koefisien kepastian (R2). Mengingat apa yang terjadi, konsekuensi dari pemeriksaan menunjukkan bahwa tingkat moral ahli mempengaruhi keterampilan evaluator yang mengesankan. Abstract

Penelitian ini berjudul: Penerapan Konseling Kelompok Dengan Teknik Stress Inoculation Training Dalam meningkatkan Resiliensi Menjelang Masa Akhir Warga Binaan Di Lapas Kelas IIB Kayu Agung. Tujuan penelitian ini adalah Untuk mengetahui penerapan konseling kelompok dengan teknik stress inoculation training dalam meningkatkan resiliensi menjelang masa akhir warga binaan di lapas kelas III kayuagung. Untuk mengetahui gambaran resiliensi dalam konseling kelompok dengan teknik stress inoculation training, Untuk mengetahui faktor yang mempengaruhi resiliensi. Penelitian ini merupakan penelitian lapangan (Field research), alat pengumpulan data yang digunakan yaitu: wawancara, observasi, dan dokumentasi. Teknik analisi data yang digunakan yaitu reduksi data, pengajian data, dan penarikan kesimpulan. Hasil dari penelitian ini: Konseling kelompok dengan teknik Stress Inoculation Training (SIT) bertujuan untuk meningkatkan resiliensi individu, khususnya menjelang pembebasan dari lembaga pemasyarakatan. Responden menunjukkan berbagai emosi seperti marah, sedih, dan takut, tetapi juga berusaha untuk berpikir positif dan mencari kesibukan yang positif. Resiliensi diperkuat melalui penyesuaian diri, pemecahan masalah, dan pemahaman terhadap konsekuensi masa depan. Temuan ini sejalan dengan penelitian sebelumnya yang menunjukkan efektivitas konseling kelompok dalam meningkatkan resiliensi. Penerapan teknik SIT membantu individu mengembangkan keterampilan dalam menghadapi stresor tertentu dan mempersiapkan mereka untuk menghadapi tantangan masa depan. Meskipun membutuhkan waktu dan kesabaran, konseling kelompok dengan teknik SIT dapat menjadi strategi efektif untuk meningkatkan resiliensi warga binaan menjelang pembebasan.

Abstract

Yeast is a single-celled fungus that acts as epiphytes, endophytes or parasites. Yeast is divided into fermentative yeast and oxidative yeast. Fermentative yeast can produce primary and secondary metabolites. The role of fermentative yeast is widely used in the food industry, health and energy, so necessary to be explore fermentative yeast from sugarcane stems. The purpose of this study was to isolate fermentative yeast from sugarcane stems and identify molecular yeast based on the Internal Transcribed Spacer sequences (ITS). Isolation of epiphytic and endophytic yeast was carried out by spread plate of water soak sugarcane and sugar cane juice. Yeast isolation using 2 media, PDA and YGP with chloramphenicol. Morphological characterization was carried out by observing macroscopic and microscopic characteristics. Biochemical characterization was carried out by carbohydrate fermentation test and 50% glucose media growth test. Selected isolates were molecularly identified using Internal Transcribed Spacer (ITS) sequences. Primers used are ITS 1 and ITS 4. Phylogenetic analysis using Neighbor Joining from MEGA-6 program. The results of isolation obtained 7 yeast isolates characterized morphologically and biochemically. The based result of morphology and biochemical characterization were found 1 selected isolate with name Ed 1B. Selected yeast isolate have characteristics are round colonies, creamy white, shiny surface, raised elevation, wavy edges, ovoid cell shape, cell diameter 4,74µm, budding, glucose fermentation and sucrose fermentation, but not for lactose and grow well of 50% glucose media. The results of the Basic Alignment Search Tools (BLAST) are Ed 1B isolates had 99% homology with Kodamaea ohmeri species.
