Abstract

This research aimed to observe quantitatively the effect of temperature and pH on protease activities from thermophilic bacteria collected from Lejja—Hot springs.  Spectrophotometry on the casein substrate was used to test the proteolytic activity of the crude protease.  Mixtures of enzyme and casein were incubated at various temperatures and pH for 20 minutes. The absorbance of tyrosine from protein hydrolysis was determined by spectrophotometry on λ 280 nm. Temperature and pH impacted on protease activity were determined at temperature (60; 65; 70; 75; 80; 85 and 90oC) and pH (6.0; 6.5; 7.0; 7.5; 8, 0; 8,5 and 9,0). Results showed that treatment of temperature variations and pH had a significant effect on protease activity. Crude extract of Bacillus licheniformis protease showed that the highest activity at 80oC and pH 7.5 was 0.1303 unit/ml/minute. Bacillus stearoformis showed the highest enzymatic activity of the protease at 85oC, and pH 7.5 was 0.1226 Unit/ ml/minute. In comparison, Bacillus coagulans reached optimum activity at 75oC, and pH 7.5 was 0.2052 Unit/ml/minute. Isolates of Bacillus licheniformis, B. coagulans and B. stearoformis are bacteria that produce thermostable protease enzymes that can be developed as a source of genes and as a producer of the enzyme itself.

Abstract

Chicken egg shells contain an inorganic element (CaCO3) which has potency to be a source of calcium hydroxide (Ca(OH)2) which is then reacted with diammonium hydrogen phosphate ((NH4)2HPO4) as a phosphate precursor for hydroxyapatite synthesis. This study aims to determine the characterization of hydroxyapatite from eggshells and determine the absorbability of hydroxyapatite-gelatin composites against methylene blue. Synthesis of hydroxyapatite used precipitation method. The calcination results obtained calcium hydroxide (Ca(OH)2) content in the shell of 68.3%. Analysis of hydroxyapatite with Fourier Tranform Infra-Red (FT-IR) there was a wavenumber of 3434 cm-1 perform hydroxyl groups (-OH-), 1421 cm-1 carbonate groups (-CO3-2) and 1035 cm-1, 604 cm-1, 471 cm-1 phosphate groups (-PO4-2), and analysis by x-ray diffraction revealed that pure hydroxyapatite crystals had formed at an angle of 2Ɵ = 34,08 with crystallinity of 100%. The peaks that appear on the XRD diffractogram are adjusted to the Joint Committee on Power Diffraction Standards (JCPDS) No. 09-0432. The synthesis of HAp-gelatin composites obtained typical groups such as, amine groups from gelatin and phosphate groups and carbonates from Hap. The results of methylene blue absorption using hydroxyapatite-gelatin composites have obtained optimum conditions at a ratio of 2: 4 for 240 minutes with an efficiency of 63%. Based on these data, it reveals that the longer the time given, the greater the methylene blue absorbed

Abstract

Cow bone has the potential to be used as a basic ingredient in the synthesis of hydroyapatite because it contains calcium. The addition of High Density Polyethylene (HDPE) to hydroyapatite aims to produce biocomposites that have better properties compared to the previous properties. The purpose of this study is to determine the characteristics of biocomposites with the HDPE:HAp in the ratio of 1: 2, 2: 1 and 3: 2 (%w / w). The method used is the sol-gel method and characterized by FTIR to see the bonding of functional groups and electron microscope to see the morphology of biocomposites. The biocomposites that have the best mechanical properties in this study are biocomposites with a ratio (HDPE: HAp) of 3: 2 with compressive and tensile strength values of 0.66 and 0.1290 N / mm2, density and water absorption values of 0.505 g / cm3 and 6,828% respectively and heat resistance with a temperature of 110 ° C experiencing a mass decrease of 0,675 g.

Abstract

Abstract

The phenomenon of Heatwaves has struck several countries across the globe due to climate change. This climate change has led to an increase in greenhouse gas emissions surpassing the limits set by the IPCC Fourth Assessment Report GWPs. This study utilizes the Support Vector Machine (SVM) classification method to identify and categorize greenhouse gas emission data from 1990 to 2020 using four kernels function such as linear, polynomial, radial basis function (RBF), and sigmoid. The SVM method demonstrates excellent performance in constructing classification models with a polynomial kernel function. This is evidenced by high values of training accuracy, testing accuracy, and F1-score, accompanied by short training and testing analysis times. Successively, these values are 97.39%, 97.69%, 96.82%, 0.59 seconds, and 0.22 seconds.
