Abstract

(Article History: Received 23 October 2020; Revised 9 January 2021; Accepted 18 January 2021) ABSTRAKSelama beberapa dekade terakhir, teknik PCR memberikan manfaat yang begitu besar dalam kegiatan penelitian di bidang biologi molekuler. Digital droplet PCR (ddPCR) merupakan salah satu teknologi PCR terbaru yang diklaim memiliki keunggulan dibanding teknik qPCR. Prinsip kerja teknik ini yaitu membagi sampel menjadi molekul-molekul kecil yang dipisahkan oleh emulsi minyak, air, dan senyawa penstabil sehingga membentuk droplets. Teknik ini memiliki kelebihan mampu melakukan kuantifikasi absolut maupun relatif pada DNA dengan konsentrasi sangat rendah, tidak memerlukan kurva standar, serta tidak sensitif terhadap kehadiran senyawa inhibitor. Teknik ini telah diaplikasikan pada kegiatan analisis molekuler tanaman di antaranya kegiatan pengukuran konsentrasi DNA dengan sangat akurat, deteksi kehadiran patogen pada jaringan tanaman, dan estimasi jumlah salinan T-DNA pada proses transformasi genetik.Kata kunci: PCR; droplet digital PCR; DNA; biologi molekuler; alat deteksi ABSTRACTOver the past decades, PCR technique has provided enormous benefits in molecular biology research activities. Digital droplet PCR (ddPCR) is one of the latest PCR technologies that is claimed to have advantages over the qPCR technique. The working principle of this technique is to divide the sample into small molecules, which separated by emulsions of oil, water, and stabilizing compounds to form droplets. This technique has the advantage of being able to perform absolute and relative quantification with very low DNA concentrations, does not require a standard curve, and less sensitive to the presence of inhibitor compounds. This technique has been applied to a number of plant molecular analysis, such as for measuring DNA concentrations very accurately, detecting the presence of pathogens in plant tissue, and estimating the copy number of T-DNA in the genetic transformation process.Keywords: PCR; droplet digital PCR; DNA; molecular biology; diagnostic tool.

Abstract

This study analyzes the reality of religiosity of students of the Religious Studies Study Program class of 2020 and the role of the Religious Studies Study Program, Faculty of Ushuluddin UIN Syarif Hidayatullah Jakarta in maintaining student religiosity. The main focus of this study is how the quality of religiosity of students of the Religious Studies Study Program and the implementation of the efforts and role of the Religious Studies Study Program in maintaining and improving the religiosity of its students. The research method used in this study is to use a descriptive qualitative method with a sociological approach and religious psychology. The results of this study show that in reality students do not experience a decrease in the quality of their religious beliefs as a Muslim. And the Religious Studies Study Program is able to implement its role and efforts in maintaining and improving student religiosity. The conclusion of this study confirms that a religious education institution which in this study is the Religious Studies Study Program, this is indeed one of the most important factors that has a very large and significant role in maintaining student religiosity.

Abstract

Kabupaten Kepulauan Meranti di Provinsi Riau merupakan salah satu wilayah penghasil sagu terbesar di Indonesia, dengan potensi produksi yang diperkirakan mencapai hampir 3,92 juta ton beras sagu pada tahun 2040. Selain sebagai sumber pangan, sagu juga menghasilkan limbah yang dapat dimanfaatkan untuk berbagai produk bernilai seperti pakan ternak, pupuk organik, dan bioenergi, yang mendukung keberlanjutan dan pengurangan dampak negatif terhadap lingkungan. Namun, pemanfaatan sagu masih dihadapkan pada kendala rendahnya nilai tambah produk, keterbatasan teknologi pengolahan, serta akses pasar yang terbatas. Artikel ini bertujuan untuk mengeksplorasi potensi agronomis, sosial-ekonomi, dan lingkungan dari sagu di Kabupaten Kepulauan Meranti, serta mengidentifikasi tantangan dan peluang dalam pengembangannya. Metode yang digunakan adalah studi kepustakaan dengan menganalisis berbagai literatur relevan antara tahun 2010 hingga 2024. Hasil penelitian menunjukkan bahwa sagu memiliki potensi besar sebagai komoditas unggulan dengan dampak positif terhadap ketahanan pangan dan perekonomian lokal. Namun, pengembangannya memerlukan peningkatan teknologi pengolahan, dukungan infrastruktur, dan penguatan akses pasar. Sagu juga memiliki peluang sebagai bahan baku bioenergi yang ramah lingkungan, khususnya bioetanol. Untuk memaksimalkan potensi ini, diperlukan sinergi antara pemerintah, lembaga penelitian, pelaku usaha, dan masyarakat lokal dalam meningkatkan nilai tambah produk dan pengembangan industri berbasis sagu.

Abstract

Marigold (Tagetes erecta) from the Asteraceae family is well-known for its various uses as cut flowers, potted plants, ornamental plants, medicines, dyes, and biopesticides. Micropropagation and optimization of in vitro regeneration of T.erecta species provide opportunities for plant propagation and utilization of biotechnology in plant breeding. This study evaluated the regeneration efficiency of in vitro plantlet leaf explants with different combinations of IAA and BAP concentrations in organogenesis induction media. IAA concentration affects the growth of adventitious shoots on explants. Decreasing IAA concentration affects the percentage of shoot growth, roots, and average number of roots in organogenesis induction media. Meanwhile, adding cytokinins to the induction media will modulate auxin movement during organogenesis and plant cell regeneration. Leaf explants on Murashige and Skoog (MS) media supplemented with 2.8 µM IAA + 31.0 µM BAP showed shoot growth reaching 58%, with root growth reaching 32%. Meanwhile, using lower BAP 2.8 µM IAA + 13.3 µM BAP resulted in 63% of explants forming shoots with a lower percentage of root growth. Increasing the concentration of IAA (5.7 µM + 13.3 µM BAP) showed a root formation response reaching 61% with a smaller number of shoots (48%). The explant response began with the explant part differentiating to form a callus with a growing point that would form yellowish-green shoots (RHS / Fan3-N144-C) at 28 days of induction
