Abstract

Asupan kalori tinggi meningkatkan profil lipid yang menyebabkan dislipidemia. Tujuan penelitian ini adalah untuk mengetahui perbedaan efektivitas pemberian simvastatin, ekstrak bawang putih, ekstrak buah naga merah dan kombinasinya terhadap kadar LDL dan kolesterol total. Penelitian ini adalah eksperimental murni dengan desain pretest posttest control group pada 30 ekor tikus yang dibagi menjadi 6 kelompok secara acak. Lima kelompok diberikan diet tinggi kolesterol dan asam kolat sampai dislipidemia selama 14 hari dan satu kelompok tidak. Lima kelompok tikus dislipidemia mendapat perlakuan tertentu selama 14 hari. Semua kelompok diperiksa kadar LDL dan kolesterol total. Hasil uji Kruskal Wallismenunjukkan nilai p<0,05 artinya terdapat perbedaan penurunan kadar LDL yang signifikan di antara keenam kelompok. Hasil uji post hoc Mann Whitney menunjukkan bahwa kelompok P2 (ekstrak bawang putih 400 mg) memiliki efektivitas paling baik dalam menurunkan kadar LDL. Hasil uji Kruskal Wallis menunjukkan nilai p<0,05 artinya terdapat perbedaan penurunan kadar kolesterol total yang signifikan di antara keenam kelompok. Hasil uji post hoc Mann Whitney menunjukkan bahwa kelompok P1 (ekstrak buah naga merah 65 mg) dan P3 (ekstrak buah naga merah 32,5 mg dan bawang putih 200 mg) memiliki efektivitas paling baik dalam menurunkan kadar Kolesterol Total.

Abstract

Introduction: The administration of antioxidant combination has been shown to be more effective than that of individual antioxidant in male infertility.Objective: to determine the effect of different combination of antioxidants (vitamin/vit C, vit E, glutathione and zinc) on the number of Sertoli and Leydig cells, sperm quality and texpression of caspase-3 in rats exposed to cigarette smoke.Methods: This was an experimental study using a post test only control group design. Thirty six rats were randomly divided into 6 groups to receive one of the following combinations of antioxidants: vit C and E, glutathione, zinc (T4-G); vit C and E, glutathione (T3-G); vit C and vit E (T2-G); Glutathione, zinc (T1-G); control positive (CP-G) cigarette smoke exposed only, normal (CN-G). The dose of vit C, vit E, glutathion and zinc was 9 mg/day, 1.8 IU/day, 1.8 mg/day, 0.2 mg/day respectively. All groups were exposed to cigarette smoke (3 cigarettes/day for 21 days), except for the CN-G group. On day 22, sperm samples were taken. Testicular tissue was taken for measurements of sperm quality, sertoli and leydig cell.numbers.Results: There was a significant difference in mean number of Leydig cells, sperm motility, sperm count and caspase-3 between groups (p<0.05). Post hoc LSD showed that the administration of combination of four antioxidant agent increasing the number of Sertoli cells and sperm morphology. The results also showed that group treated with 4 antioxidants combination had the highest number of Leydig cells, sperm motility, sperm counts and the lowest expression of caspase-3.Conclusion:The combination of vitC, vit E, glutathione and zinc affects the number of Sertoli and Leydig cells, sperm quality, expression of caspase-3 in rats exposed to cigarette smoke .

Abstract

Background: Hypercholesterolemia is a state of total blood cholesterol levels transported by LDL exceeding normal limits. As a result, it can clog blood vessels, and this condition is a risk factor for CHD. Alternative medicine includes flavonoids in Averrhoa bilimbi (belimbing wuluh) known to reduce cholesterol and LDL levels, and curcumin in turmeric known to reduce fat absorption and increase fat excretion. Objectives: The aim of this study was to analyze the combined effect of bilimbi fruit and turmeric extracts on LDL levels. Methods: This experimental research used post-test only with a control group design involving male Wistar rats divided into 6 (six) random groups. Group KN (standard feed, aquadest for 28 days), K- (high-fat feed for 14 days, followed by standard feed and aquadest), K+ (high-fat feed for 14 days, followed by standard feed, aquadest, and simvastatin 0.18mg/200 g BW/day), P1 (high-fat feed for 14 days, followed by standard feed, aquadest and extracts of bilimbi fruit and turmeric extract at a dose of 240: 270 mg/200 g BW/day), P2 (high-fat diet for 14 days, followed by standard feed, distilled water and bilimbi fruit and turmeric extract at a dose of 360: 175 mg/200 g BW/day), P3 (high-fat diet for 14 days, followed by standard feed, distilled water and extracts of bilimbi fruit and turmeric at a dose of 120: 405 mg/200 g BW/day). The determination of LDL levels was done through a direct method using a spectrophotometer with a wavelength of 550 nm. Results: The mean LDL levels were KN (53.60 ± 10.69) mg/dL, K- (193.40 ± 16.86) mg/dL, K+ (100.20 ± 42.48) mg/dL, P1 (118.00 ± 64.71) mg/dL, P2 (104.00 ± 45.28) mg/dL, P3 (78.00 ± 4.69) mg/dL. The data received was analyzed using the Kruskal-Wallis test. Then, the results obtained differences in LDL levels between various groups (p<0.001). Subsequently, it was continued with the Mann-Whitney test on KN compared to K-, K+, P1, P2, P3 indicating p=0.009. As a result, it was found a significant difference in LDL levels between KN and K-, K+, P1, P2, and P3. Conclusions: This research concludes that providing a mixture of bilimbi fruit extract and turmeric extract can reduce LDL levels.

Abstract

Background: The use of probiotics to treat colitis is unsatisfactory, leading to a need for the combination with purple sweet potatoes (Ipomoea Batatas) and zinc. Objectives: This study aims to determine the effect of the combination of probiotics, purple sweet potatoes, and zinc on epithelial thickness.Methods: This was a post-test-only randomized controlled group experimental study using 30 mice divided into six groups, including KN, K-, K+, P1, P2, and P3. All groups except KN were induced with 4% acetic acid on day 1, then each received treatment for five days. KN, K+, P1, and P2 groups were given distilled water, sulfasalazine 1.3 mg/20 g BW, probiotics 0.078 mg/20 gBW + purple sweet potatoes 0.00169 g/20 gBW, and probiotics 0.078 mg/20 gBW + zinc 0.052 mg/20 gBW, respectively. Meanwhile, P3 received a combination of probiotics 0.078 mg/20 gBW + purple sweet potatoes 0.00169 g/20 gBW + zinc 0.0052 mg/20 gBW. Histological preparations were stained with Hematoxylin-Eosin and examined under 400× magnification across 10 fields of view. Results: The average epithelial thicknesses were KN (75.48±5.09), K− (28.02±5.60), K+ (69.1±13.82), P1 (47.05±9.41), P2 (48.6±9.73), and P3 (71.57±14.31). One-way analysis of variance (ANOVA) showed a significant difference among the groups with a p-value <0.001. Post Hoc LSD test identified significant differences (p-value<0.05) among all group pairs except between K+ and P3 (p-value=0.134) as well as P1 and P2 (p-value=0.349), but P3 had the highest average thickness value (71.57±14.31).Conclusions: The combination of probiotics, purple sweet potatoes, and zinc had an effect on intestinal epithelial thickness in male BALB/c mice with ulcerative colitis.
