Abstract

The red calliandra is a fuel wood crops that gave benefit for people. Recently, it is getting wide attention due to its important use as biofuel in industrial scale. The massive propagation is needed to meet industrial needs. Beside the use of conventional seedling production technique, the use of tissue culture for mass propagation is one of the alternatives to address this problem. This research was conducted to develop standard protocol for micropropagation of red calliandra through tissue culture. Here, the first step for that purpose was reported. The MS medium was used for basal media treatment. The seed source was stored for eight months under 5oC. This research was carried out in two stages i.e. 1) seed germination test and 2) shoot initiation and multiplication treatment. Each stage was arranged in experimental method. The result showed that seed were pre-treated with warm water (40oC) gave 62.5% germination under red light when compared to control (37.5%, pre-treated with water at 25oC under white light) three days after planting. Conversely, there is no striking differences of germinated seed percentages between light treatment at 25oC. Furthermore, it was found that addition of 2 mg/L BAP gave better result in shoot initiation and number of shoots formed from nodal explants after four weeks in treatment media. These results indicated that mass propagation of red calliandra can be done through tissue culture where multiplication stage is one of the important steps to produce multiple shoots only from single nodal explant.

Abstract

Menenun sudah dilakukan sejak dulu kala untuk kebutuhan sandang manusia. Orang Minangkabau menyebut diri mereka sebagai orang beradat, kain tenun tidak bisa dipisahkan dari upacara-upacara adat istiadat.Kain tenun ditinjau dari bahan, cara pembuatan, dan harganya, menunjukkan kemuliaan derajat dan martabat pemakainya. Kondisi ini menjadikan kerajinan tenun sebagai salah satu industri rumah tangga dan memunculkan kelompok usaha kecil dan menengah (UKM) di kalangan masyarakat khususnya di Nagari Kubang Kabupaten Limapuluh Kota. Kelompok usaha kecil dan menengah (UKM) Marni Geneng dan CV Rangkiang adalah mitra dari IbPE Kerajinan Tenun Minangkabau di Nagari Kubang Kabupaten Limapuluh Kota. Kedua UKM ini sepakat untuk bermitra dalam kegiatan IbPE yang diselenggarakan oleh Tim Pengabdian Politeknik Pertanian Negeri Payakumbuh dalam rangka menuju pasar ekspor luar negeri. Tujuan dalam program IbPE pada kedua UKM ini adalah memperbaiki pengelolaan usaha berupa perbaikan administrasi, jumlah dan mutu produk serta promosi untuk memperluas pasar produk. Hasil yang dicapai dalam program IbPE tahun I ini adalah perbaikan adminsitrasi pengelolaan UKM berupa penataan buku kas, buku stok, buku penjualan, buku pembelian, buku tamu tersedia dengan rapi. Pelatihan pewarna alami menghasilkan produk tenun yang berwarna natural. Pelatihan pengembangan motif meningkatkan kualitas produk tenun yang dihasilkan lebih menarik. Pemasangan arah penunjukan jalan pada kedua UKM sehingga lokasi UKM mudah ditemui konsumen. Pemberian kemasan pada produk kedua UKM meningkatkan nilai jual produk tenun UKM.

Abstract

Red calliandra is one of woody plants that has been extensively developed as biofuel. Tissue culture techniques are employed to produce a large and uniform quantity of seedlings in a relatively short period. Previous stages of in vitro initiation and multiplication of shoot from calliandra explants have shown promising results. The shoots that were multiplied by in vitro technique need to be induced for rooting process to produce plantlets. These plantlets are then prepared for the subsequent acclimatization process. This research was conducted in two stages; root induction of shoots using a modification of MS half-strength media (MS ½) supplemented with 1-4 mg/L 1-naphthaleneacetic acid (NAA), and acclimatization of the plantlets into soil media. The results shown that MS ½ medium and media supplemented with 1 mg/L NAA were suitable for root induction, particularly to induce the root length. Furthermore, MS ½ media supplemented with 1-2 mg/L NAA stimulates the increment of plantlet height and leaves numbers. Plantlets which were transferred into soil exhibit robust growth during 3-weeks of acclimatization process, up to 100% plantlets survived during this stage. These results underscore the potential of tissue culture techniques in providing large scale calliandra seedlings production for future biofuel development.
