Abstract

Plant growth promoting rhizobacteria, known as Plant Growth Promoting Rhizobacteria (PGPR), are bacteria found on the surface of plant roots and are beneficial microorganisms. Fluorescent pseudomonads are a group of bacteria belonging to PGPR and have the ability to dissolve phosphates, produce nitrogen, phosphorus, siderophores, and Indole Acetic Acid (IAA) to stimulate plant growth. PGPR must be immobilized in a certain carrier in the form of a formulation so that it is easy to use, store, commercialize, and apply to the field. Due to the increasingly expensive instant media, the development of media formulations continues to be carried out which can be organic or inorganic.

Abstract

The majority of farmers currently use chemical fertilizers and pesticides in farming. However, continuous use of these materials can cause negative impacts such as environmental pollution, health risks, nutrient cycle imbalances, and reduced crop yields. To overcome this problem, research has been conducted that focuses on the use of microorganisms found in the soil layer surrounding plant roots, called the rhizosphere. These microorganisms such as bacteria, fungi and protozoa form a group known as Plant Growth Promoting Rhizobacteria (PGPR). PGPR can increase plant growth through various mechanisms such as nitrogen fixation, phosphorus dissolution, production of cyanide acid compounds (HCN), and Indole Acetic Acid (IAA) compounds. This research aims to collect and analyze articles related to the role of PGPR in sustainable agriculture. The method used is a literature review by collecting sources from Google Scholar and other internet databases. The results of the articles collected were on topics related to PGPR, including mechanisms of nitrogen fixation and phosphorus solubilization, as well as their role in producing IAA and siderophores. Apart from that, the research also discusses the role of PGPR as a biocontrol agent that can control plant pathogens, and the antagonistic activity of PGPR against pathogens.

Abstract

The xylanase enzyme is an extracellular enzyme that is capable of hydrolyzing hemicellulose into xylooligosaccharides and xylose. One of the benefits of the xylanase enzyme is as a paper and pulp bleach. This enzyme is produced by thermophilic bacteria with the substrate contained in Beechwood Xylan. However, using Beechwood Xylan is expensive, so other alternatives are needed at more affordable prices. Agricultural waste is believed to be able to replace Beechwood Xylan because it contains xylan. The aim of this research is to find the best waste substrate to replace Beechwood Xylan as a medium for a consortium of thermophilic bacteria to produce xylanase enzymes. This research was conducted at the Microbiology Laboratory, Faculty of Mathematics and Natural Sciences, Padang State University in December 2023. The method used was descriptive, using 3 types of agricultural waste, namely straw, rice husks, and corn with samples of thermophilic bacteria SSA2, SSA3, and SSA4. Enzyme activity was measured using a spectrophotometer with a wavelength of 540nm. The data obtained is analyzed and displayed in tabular form. The results of this study show that straw has the highest average xylanse enzyme activity of 9,289 Units/mL, and husks have the lowest average enzyme activity of 6,161 Units/mL.

Abstract

The increasing global energy demand and the negative impact of the use of fossil energy on the environment encourage the use of environmentally friendly renewable energy sources, one of which is through Microbial Fuel Cell technology. This study aims to measure or analyze bioelectrical production by utilizing a consortium of thermophilic bacterial bicultures (SSA 14 and SSA 16) in an MFC system arranged in a series circuit configuration. The research method was carried out experimentally in the laboratory using a dual-chamber reactor arranged in a series of two and three series, with voltage measurements carried out every 2 hours for 24 hours. The results showed that the series three series produced a higher average voltage (0.823 V) than the series two series (0.744 V). This study indicates that the addition of the number of fermenter units in the series series significantly increases the electrical voltage produced.

Abstract

The global urgency for sustainable energy solutions amid climate change and fossil fuel depletion has driven innovation in Microbial Fuel Cells (MFCs). This study investigates the synergistic interactions within a thermophilic microbial consortium (MS9 & MS12) to optimize energy efficiency in MFCs. By varying the inoculum ratios (1:1, 1:2, and 1:3), the research evaluates how microbial population balance impacts bioelectricity production. The MFCs were operated for 24 hours, with voltage measurements recorded every 2 hours. Results revealed that a 1:3 (MS9:MS12) ratio achieved the highest voltage output, peaking at 20 hours before declining, indicating metabolic synergy and efficient substrate utilization. Descriptive analysis highlighted the critical role of cross-feeding mechanisms and functional complementarity between species in enhancing electron transfer. The findings align with modular co-cultivation principles, emphasizing the importance of engineered microbial ratios for stable system performance. This study provides a protocol for designing balanced thermophilic consortia, advancing sustainable bioenergy strategies through optimized microbial interactions.
