Abstract

Background: Dietary antioxidants are essential to prevent free radical-induced cell damage and maintain the body’s physiological functions. Sembung leaves are rich in polyphenols, which are known to have substantial antioxidant activities. The antioxidant efficacy of Sembung leaves can be enhanced by nanoparticle size. Objectives: To examine the enhancement of antioxidant activity of Sembung leaf (Blumea balsamifera L.) extract modified into nanoparticles. Methods: The extract used was an ethyl acetate fraction extract. The fresh leaves of Sembung ware macerated in 96% ethanol for 24 hours. Ethanol, n-hexan, and ethyl acetate solvents were used to fractionate the ethanolic extract. The extract of the ethyl acetate fraction was modified into nanoparticles through the ionic gelation method. The nanoparticles comprised three formulations, F1 (0.25% chitosan and 10 ml NaTPP solution), F2 (0.50% chitosan and 20 ml NaTPP solution), and F3 (0.75% chitosan and 30 ml NaTPP solution). The antioxidant activity was assessed by lipid peroxidation using ferric thiocyanate (FTC) followed by the thiobarbiturat (TBA) method. A Spectrophotometer UV-Vis was used to analyse the % inhibition values of the samples. Results: The antioxidant activity assay on the sample of ethyl acetate fraction using the FTC method showed 31.25% inhibition, and using the TBA method indicated 17.34% inhibition. Meanwhile, the % inhibition was 23.77% and 40.43% in the nanoparticle sample with the FTC and TBA methods, respectively. Conclusions: This study revealed that nanoparticles of Sembung leaves  can increase antioxidant activities. These findings offer potential applications in the development of existing antioxidant products.

Abstract

Kemumu leaf stalks (Colocasia Gigantea Hook.F) are a plant that has many benefits for society, one of which is anti-inflammatory. Inflammation is process in the body to respond to infection or tissue damage. This study aims to test the anti-inflammatory effectiveness of ethanol extract, n-hexane, and ethyl acetate of kemumu leaf stalks in vitro. Kemumu leaf stalks contain flavonoids, alkaloid and tannin which have anti-inflammatory properties. Anti-inflammatory effectiveness testing was carried out on peritoneal macrophage cells to determine the inhibition of NO production in LPS-stimulated peritoneal macrophage cells by quantifying nitrite accumulation in the culture medium using Griess reagent. This research is an experimental study with 5 dose treatments for each extract with 3 replications. The parameters observed included NO production and the percentage of peritoneal macrophage cell inhibition. The results of statistical tests using One Way Anova showed a significant difference in NO levels between the sample group given the extract and the control sample (p=0.000). From the results of the percentage inhibition of NO production in peritoneal macrophage cells, it shows that ethanol extract of kemumu leaf stalks with concentrations of 50 and 100 µg/ml has the best anti-inflammatory effect, namely with NO production of 4.67 and 1.44 µg/ml respectively, and with respective inhibition 80.32% and 89.01% respectively.

Abstract

This study aimed to evaluate the potential of durian seed flour (Durio zibethinus Murr) as an alternative medium for the growth of Candida albicans and Aspergillus niger. The bleaching process utilized sodium metabisulfite at varying concentrations (600 ppm, 800 ppm, 1000 ppm) and immersion durations (15, 20, 25 minutes). The bleached samples were dried at 70℃ for 3 hours. Durian seed flour media were formulated into three variants and compared with Potato Dextrose Agar (PDA) as the control medium. The activity test for Candida albicans used the pour plate method with incubation for 48 hours at 37℃, while Aspergillus niger was tested using the agar block method and incubated for 72 hours under the same conditions. Immersion duration significantly affected the flour's color, with longer durations producing whiter flour. Higher drying temperatures enhanced water evaporation efficiency, resulting in an average moisture content of 10.56%, compliant with national standards. The best medium formulation was F1 (10 g durian seed flour, 2 g sugar, 1.5 g agar, 0.25 g micronutrients, 100 mL distilled water), yielding 3.6 × 10⁷ CFU/mL of Candida albicans colonies and an Aspergillus niger growth diameter of 28.5 mm after 72 hours of incubation. However, sporulation of Aspergillus niger was better on PDA media. Durian seed flour shows potential as an alternative fungal growth medium.
