Abstract

Sari. Tujuan penelitian adalah mengevaluasi pengaruh ethyl methanesulphonate (EMS) terhadap pertumbuhan tanaman kentang Granola dari biji botani dan memperoleh konsentrasi efektif untuk mendapatkan mutan berdaya hasil tinggi. Percobaan dilakukan dalam dua tahap. Tahap pertama dilakukan di laboratorium kultur jaringan BALITSA, Lembang, yang terdiri dari tahap perlakuan EMS pada biji dengan konsentrasi  0,01%; 0,03%; 0,05%; 0,07%; 0,10%; 0,13%; 0,15%; 0,17%; dan 0,20%; selama 3 dan 6 jam, penanaman biji pada media kultur MS, perbanyakan planlet dan pengamatan planlet. Tahap kedua dilakukan di rumah kasa di Pangalengan yang terdiri dari tahapan aklimatisasi menggunakan Rancangan Acak Kelompok dengan 14 perlakuan EMS yang diulang 3 kali dan pengamatan pertumbuhan tanaman. Hasil menunjukkan bahwa EMS menyebabkan penurunan pada daya kecambah. Pengamatan pertumbuhan di rumah kasa menunjukkan tinggi tanaman, jumlah daun, indeks kandungan klorofil, dan berat ubi pertanaman hasil perlakuan memiliki hasil yang lebih rendah dibandingkan kontrol. Konsentrasi EMS 0,07% dengan perendaman 3 jam dan konsentrasi 0,01% dengan perendaman 6 jam menghasilkan genotipe 3D12 dan 6A8 yang memiliki hasil panen tinggi. Kata kunci: kentang, EMS, mutasi, pertumbuhan. Abstract. The aim of the study was to evaluate the effect of ethyl methanesulphonate (EMS) on true potato seed germination and growth of potato Granola also obtained effective concentrations of EMS to produce mutants with high yield. Experiment was conducted in two stages. The first stage was carried out in BALITSA tissue culture laboratory, Lembang which consisted of EMS treatment steps in seeds with concentrations of 0.01%, 0.03%, 0.05%, 0.07%, 0.10%, 0.13%, 0.15%, 0.17%, and 0.20% for 3 and 6 hour; seeds planting on MS culture media; planlet propagation and plantlets observations. The second stage was carried out in screen house in Pangalengan which consisted of acclimatization stages using a randomized block design with 14 EMS treatment repeated 3 times and observations of plant growth, and yield. The results showed that EMS caused a decrease in germination. Growth observation results at screen house showed plant height, number of leaves, chlorophyll content index, and weight of tubers from treatment had lower than controls. Treatment with 0.07% EMS concentration for 3 hours and 0.01% consentration for 6 hours produced 3D12 and 6A8 mutan genotypes which had high yields.  Keywords: potato, EMS, mutation, growth.

Abstract

Abstract The object that conducted in this study is farmer's preferences of rice plant charac-teristics. This study was taken in 3 villages which are village Ciganjeng, village Pamotan, and village Rawaapu. The method used in this study was quantitative design with descriptive analysis. The respondents were 64 rice plant farmers. Measuring instrument used was questionnaire and interview. The result of this study showed that farmers has its own preference for the character of rice plants. Conclution of this study is farmers preferred rice plants which has an average height of plants, wide leaves, leave's surface, leave's angle, and rice straw's type; rice straw that has many branches; strong braches; has a lot rice grains; maturity of rice plants; lenght and wide grains size; non-aromatic; and tender texture. The farmers thought that leave's surface isn't an important character of a rice plants;  high of plants, leave's lenght and wide, leave's angle, rice straw's type, grain's lenght and wide, and rice's scent are quite important characteristics; branch's strenght, grain's quantity, and plant's maturity are an important characteristics; resistance to pests, disease resistance, drought resistance, resistance to inundation, salinity resistance, productivity, and rice texture are the most important characteristics. Keywords : preferences, farmer, superior varietiesObjek yang dikaji dalam penilitian ini adalah preferensi petani terhadap karakter tanaman padi. Tempat penelitian dilaksanakan di 3 desa (Desa Ciganjeng, Desa Pamotan dan Desa Rawaapu). Desain penelitian menggunakan desain kuantitatif dengan pendekatan analisis data secara deskriptif. Metode yang diperunakan adalah survey untuk mengumpulkan data dengan mengedarkan kuisioner dan melakukan wawancara. Jumlah petani yang diwawancarai yaitu 64 orang. Hasil dari penelitian ini adalah petani lebih menyukai tanaman padi yang memiliki tinggi tanaman, panjang daun, lebar daun, permukaan daun, sudut daun dan tioe malai yang sedang, cabang malai sekunder yang banyak, ketegaran batang yang kuat, jumlah anakan yang banyak, umur yang genjah, ukuran gabah yang panjang dan lebar, aroma yang tidak wangi, serta tekstur nasi yang pulen. Petani beranggapan bahwa karakter permukaan daun merupakan karakter yang tidak penting; karakter tinggi tanaman, panjang daun, lebar daun, sudut daun, tipe malai, panjnag gabah, lebar gabah, dan aroma tergolong karakter cukup penting; karakter ketegaran batang, jumlah anakan, dan umur tanaman merupakan karekter yang penting; karakter ketahanan terhadap hama, ketahanan terhadap penyakit, ketahanan terhadap kekeringan, ketahanan terhadap genangan, ketahanan terhadap salinitas, produktivitas, dan karakter tekstur nasi merupakan karakter yang sangat penting.Kata kunci : preferensi, petani, varietas unggul

Abstract

Exopolysaccharide (EPS) produced by nitrogen-fixing bacteria Azotobacter protect nitrogenase from oxygen. In legume,EPS plays a role in the immobilization of rhizobia to the roots. The objective of this experiment was to study the effect of EPSAzotobacter and organic matter on increasing number of nodules and biomass of soybeans grown in Inceptisols and Ultisols;as well as nitrogen-fixing bacteria population in soybean rhizosphere. The experiment was set up in a completely randomizedblock design with five replications to test combined treatments of two doses of crude EPS and organic matter. Nodule number,shoot dry weight and nitrogen uptake, as well as Azotobacter and Rhizobium population in soybean grown in Inceptisolsfollowing crude EPS and compost application, were higher than those grown in Ultisols. The application of EPS and compostIn Ultisols did not affect the number of nodule and other traits, but in Inceptisols, adding 6.25 g of compost and 20 mL of EPSto each plant increased the number of nodules and shoot weight at 42 days after planting. However, the highest N uptake wasdemonstrated by soybean received 10 mL and 20 mL EPS along with 12.5 g compost.

Abstract

Keragaman genetik ubi jalar dapat dinilai berdasarkan analisis terhadap karakter-karakter tertentu. Tujuan penelitian ini adalah untuk melakukan pendugaan keragaman klon ubi jalar ungu berdasarkan karakter agronomi dan morfologi tanaman. Percobaan dilaksanakan selama musim hujan di Kebun Percobaan Ciparanje, Fakultas Pertanian Universitas Padjadjaran, Sumedang, Jawa Barat mulai November 2018 sampai April 2019. Perlakuan adalah 11 klon ubi jalar ungu yang berasal dari Indonesia, Peru, dan Jepang. Pengamatan terdiri atas karakter agronomi dan morfologi, serta komponen hasil. Pendugaan keragaman genetik menggunakan analisis klaster, analisis komponen utama, uji mantel, dan perhitungan ragam genotipik dan fenotipik. Hasil penelitian menunjukkan terdapat perbedaan keragaman genetik berdasarkan karakter agronomi dan karakter morfologi. Jarak genetik berdasarkan analisis klaster dan analisis komponen utama pada karakter agronomi dan morfologi masing- masing adalah 0,31–5,18 Euclidean dan 1,73–4,55 Euclidean. Korelasi keragaman agronomi dengan keragaman morfologi sangat lemah (r=0,096). Perhitungan ragam genotipik dan fenotipik juga menunjukkan bahwa keragaman karakter agronomi cenderung luas.

Abstract

This study was aimed at determining the appearance of phenotypic characters in five cayenne genotypes in Sukamantri, Ciamis Regency. The research method used was the experimental method with Randomized Block Design (RBD) with five treatments repeated five times. Data were analyzed by variance and followed by Least Significant Difference (LSD) Test. The testing material consisted of five genotypes of cayenne pepper, namely one of the new superior varieties (Ratuni Unpad), and four varieties (CR8873, Dewata, Taruna, and Rabani). The tests were carried out in Sukamantri, Ciamis Regency in 983 m asl in August 2017 to February 2018. The observations were made on the character of the results and components of results which included the seed length, the seed diameter, the seed weight per unit, the seed weight per plant, the seed weight per plot, and the seed weight per hectare. The test results showed that there were differences in phenotypic characters between the five genotypes tested. The Dewata, Ratuni Unpad, and CR8873 genotypes show the highest potential yield (8.34 tons/ha, 7.71 tons/ha, and 7.24), with the character of fruit weights per fruit of 1.382 grams, 0.911 grams respectively. and 1,311 grams.PENAMPILAN KARAKTER FENOTIPIK HASIL PADA LIMA GENOTIP CABAI RAWITPenelitian ini bertujuan untuk mengetahui penampilan karakter fenotipik pada lima genotip cabai rawit di Sukamantri, Kabupaten Ciamis. Metode penelitian yang digunakan adalah metode eksperimen dengan Rancangan  Acak  Kelompok  (RAK)  dengan  lima  perlakuan  diulang  lima  kali. Data dianalisis dengan sidik ragam dan dilanjutkan dengan Uji Least Significant Difference (LSD). Bahan pengujian terdiri atas lima genotip cabai rawit, yaitu salah satu varietas unggul baru (Ratuni Unpad), dan empat varietas (CR8873, Dewata, Taruna, dan Rabani). Pengujian dilakukan di Sukamantri, Kabupaten Ciamis dengan ketinggian tempat 983 m di atas permukaan laut (dpl.) pada Agustus 2017 sampai Februari 2018. Pengamatan dilakukan terhadap karakter hasil dan komponen hasil yang meliputi panjang buah, diameter buah, bobot buah per buah, bobot buah per tanaman, bobot buah per plot, dan bobot buah per hektar.Hasil pengujian menunjukkan bahwa terdapat perbedaan karakter fenotipik antarlima genotip yang diuji. Genotip Dewata, Ratuni Unpad, dan CR8873 menampilkan potensi hasil yang tertinggi (8,34 ton/ha; 7,71 ton/ha; dan 7,24); dengan karakter bobot buah per buah masing-masing sebesar 1,382 gram; 0,911 gram; dan 1,311 gram.
