Abstract

Disturbance on the immune system and deficiency of Zn is two factors which often trigger vaginal candidiasis patient. The aim of the research was to study the effect of virgin coconut oil (VCO) enriched with Zn to the amount of neutrophil and lymphocyte subset cells, and the level of IL-2 and IgG in vaginal candidiasis patient. Thirty women were grouped into three (ten women in each group): A, B and C, and intervened for two months. Women in A group were intervened with two tablespoon/day; those in B group were intervened with one tablespoon/day; while those in C group served as control (placebo). Blood was sampled at baseline time, one and two months after intervention. Hematological test by Micros-OT was done on a part of blood, and the plasma was used for IL-2 and IgG level tests using ELISA. The virgin coconut oil enriched with Zn maintained the number of neutrophil and NK cells, but increased Tc cells from 521 to 649 cells/mm3, increased Th cells from 1.090 to 1.380 cells/mm3. The enriched VCO also increase level of IL-2 from 0.25313 to 0.27337 pg/ml, while the IgG level changed from equivocal to negative. The recommended dosage was one tablespoon each day. Abstract

ABSTRAK Kadmium merupakan salah satu logam berat yang bersifat nefroksik dan membahayakan kesehatan manusia, terutama pekerja bengkel las. Kadmium masuk ke dalam tubuh berikatan dengan metalotionin membentuk ikatan Cd+Mt akan terdeposit di organ ginjal dan menginduksi terbentuknya radikal bebas sehingga mengakibatkan peroksidasi lipid yang dapat merusak organ ginjal, ditandai dengan peningkatan kreatinin dan β2 mikroglobulin. Tujuan penelitian adalah untuk mengetahui efek paparan Cd terhadap fungsi ginjal pekerja bengkel las ditinjau dari parameter kreatinin dan β2 mikroglobulin (β2-M). Rancangan penelitian menggunakan rancangan kasus kontrol. Subyek penelitian terdiri dari 30 orang pekerja bengkel las di Purwokerto dengan masa kerja lebih dari 3 tahun sebagai subyek kasus dan subyek kontrol dari desa Pamijen Baturaden yang mewakili bukan pekerja bengkel las. Data dianalisis menggunakan uji t independent. Hasil penelitian menunjukkan kadar Cd darah, kreatinin darah dan β2 mikroglobulin pada subyek kasus berbeda nyata dibandingkan dengan subyek kontrol. Kadar Cd, kreatinin darah dan β2 mikroglobulin subyek kasus lebih tinggi dibandingkan subyek kontrol. Kesimpulan penelitian paparan Cd berpengaruh terhadap penurunan fungsi ginjal pekerja bengkel las yang ditandai dengan peningkatan kadar Cd darah, kreatinin dan β2-M.

Abstract

Plumbum bersifat toksik terhadap manusia. Teh hijau digunakan untuk mengobati penyakit karena mengandung catechin. Tujuan penelitian untuk mengetahui efek catechin ekstrak teh hijau sebagai kelator Pb. Sebanyak 36 ekor tikus Wistar digunakan dalam penelitian ini. Tikus dibagi 6 kelompok, 6 ekor per kelompok. Kelompok I sebagai kontrol negatif. Kelompok II sampai VI diberi Pb asetat selama penelitian (66hari). Kelompok II sebagai kontrol positif tidak diberi perlakuan. Kelompok III, IV, dan V pada hari ke 35 diberi ekstrak teh hijau masing- masing 0,75 g/kgbb, 1,5 g/kgbb dan 3 g/kgbb. Kelompok VI diberi dimerkaprol dosis 3 g/kgbb sebagai pembanding. Ada tidaknya beda nyata antar perlakuan dianalisis dengan ANOVA diikuti dengan uji Duncan untuk mengetahui letak perbedaan. Parameter yang diukur adalah malondialdehid, aktivitas glutation peroksidase dan nitrit oksida, diukur dengan komersial kit. Hasil penelitian menunjukkan semua dosis ekstrak teh hijau dapat menurunkan MDA, Pb darah dan meningkatkan GPx serum serta NO. Dosis 3 g/kgbb optimum dalam menurunkan kadar MDA dari 2,75–0,07 µmol/L dan Pb darah dari 2,35–0,02 ppm. GPx serum meningkat dari 58,5–177 µmol/L dan NO serum dari 1,27,95 µmol/L. Simpulan hasil penelitian, ekstrak teh hijau dapat digunakan sebagai kelator Pb.

Abstract

AbstractBasil leaves (Ocimum basilicum) is one of the traditional medicinal plants that is used as medicine.This study aims to determine the effect of giving basil leaves on levels of Beta 2 microglobulin, uric acid and kidney histology in white wistar-induced rats in MSG and determine the effective dose of basil leaves in an effort to improve kidney function.This study used 25 male Wistar strain rats with body weight 200-230 g. The test animal group was divided into five groups based on the stage of treatment. The first treatment group was a control group consisting of five mice that were not given basil leaf extract or MSG, the second treatment group was the group that was only given MSG without being given basil leaf extract, the third treatment group was the group given MSG followed by basil leaf extract dose 87, 5 mg / kgBB, the fourth treatment group was the group that was given MSG followed by the administration of basil leaf extract with a dose of 175 mg / kgBW, and the fifth treatment group was the group that was given MSG followed by the administration of basil leaf extract at a dose of 350 mg / kgBB. At the end of the treatment, rats were drawn for blood analysis of B2M levels and uric acid levels and kidney tissue uptake, as well as analyzing kidney tissue histology.The results showed that there were differences between Beta 2 microglobulin, uric acid and kidney histopathology of rats receiving basil leaf extract compared with MSG group without basil leaf extract. This study concludes that consuming basil leaf extract can reduce levels of B2M and uric acid and improve kidney histology induced by MSG.

Abstract

Limbah batik merupakan salah satu penyebab pencemaran di lingkungan perairan karena diketahui mengandung beberapa jenis logam berat. Jenis logam berat tersebut antara lain seperti krom (Cr), seng (Zn), merkuri (Hg), tembaga (Cu), timbal (Pb), Arsen (As), kadmium (Cd), mangan (Mn) dan nikel (Ni). Ikan yang terpapar limbah batik memberikan respon secara fisiologis. Respon yang muncul sebelum ikan mengalami kematian disebut efek subletal. Ikatan antara logam berat atau metal (M) dengan protein metalotionin ini disebut sebagai ikatan M+Mt. Ikatan ini bersifat stabil sehingga tidak mudah lepas. Hal inilah yang akhirnya memicu pembentukan radikal bebas. Ketika radikal bebas dalam tubuh semakin meningkat, maka kadar enzim Superoksida Dismutase atau SOD akan semakin menurun. Pengujian kadar enzim SOD dapat dijadikan sebagai upaya deteksi dini adanya pencemaran perairan yang disebabkan oleh limbah batik. Berdasarkan hal tersebut, maka penelitian ini memiliki tujuan untuk melihat pengaruh toksisitas limbah batik terhadap penurunan enzim SOD pada ikan nila. Berdasarkan hasil penelitian ini, kadar SOD mulai dari kontrol, konsentrasi 17% v/v, 34% v/v dan 51% v/v berturut-turut yaitu 29,48 ± 1,40 U/mL, 22,20 ± 1,18 U/mL, 17,77 ± 1,16 U/mL, 13,68 ± 0,56 U/mL. Hasil analisis data menunjukkan bahwa nilai F hitung (218,570) > F tabel (4,30) yang artinya terdapat pengaruh paparan limbah batik terhadap penurunan aktivitas SOD. Semakin tinggi konsentrasi limbah batik maka semakin rendah aktivitas SOD. Kata kunci: Limbah Batik, Oreochromis niloticus, Superoksida Dismutase (SOD)

Abstract

Welding was one of the industry in Purwokerto City. One of the exhaust particulate material from the welding process, this will be the potential occurrence of exposure to workers and caused renal dysfunction. Several risk factors have contributed in caused kidney difunction of workers. This study aims to determine the levels of cadmium metal (Cd), chromium (Cr), lead (Pb) with creatinine and Glomerulus Filtration Rate (GFR). The correlation of risk factors for renal dysfunction with creatinine and GFR. The risk factors that play a major role in caused kidney dysfunction. The method used is cross sectional. Sampling using purposive sampling with giving inform consent to the respondent. Provision of questionnaires and blood sampling as much as 3 ml according to ethical clearens. Measurement of metal content of Cd, Cr and Pb; serum creatinine and LFG levels. Number of respondents 30 people with 95% confidence level. The results showed that levels of Cr, Cd and Pb were 0.049 + 0.12 ppm; 1,029 + 2.38 ppm; 4,933+ 11,66 ppm. Cr levels are still within normal limits with levels but Cd and Pb exceeding the normal threshold. Creatinine levels of 1.867 + 0.446 mg / dl showed higher than normal. Minimum of GFR decrease in welding workers was 44 mL / min / 1.73 m2. Risk factors significantly correlated with creatinine and GFR was levels of Pb (p <0.05). Risk factors that play a major role in caused kidney dysfunction in welding workers in Purwokerto city is Pb content with 12.9% percentage effect on serum creatinine level and 9% effect on Glomerulus Filtration Rate.

Abstract

The research about rhizofiltration of Pb in several species of aquatic plant had been conducted in a greenhouse of Biology Faculty, Jenderal Soedirman University. The aims of this research were to evaluate the aquatic plant species which has Pb accumulative characteristic and to determine Pb concentration tolerated by those plants. This research was used experimental method with split plot design. The main plot was kind of aquatic plant of Hydrilla verticillata (L.f), Pistia stratiotes L, and Salvinia molesta D.S.Mitchell. The Pb (NO3)2 concentrations of 0, 5, 10, 15 mg/l were as sub plot.   Each treatment was replicated for 3 times. The parameter observed were biomass (raw weight and dry weight), the Pb absorbed whether in root and shoot, and chlorophyll content. Data collected were analyzed using F test continued by LSD. The result of this research showed that both Pistia and Salvinia planted in 10 mg/l media were able to accumulate high Pb concentration. However, Pistia were able to accumulate Pb concentration mostly in root and only part be translocated in shoot. Whereas Salvinia was able to translocate Pb concentration in shoot higher than those in of Pistia.  The implications of these results for rhizofiltration are discussed.

Abstract

Cadmium is a heavy metal pollutant sourced from various industries and toxic to the kidneys. Cadmium exposure can be used natural chelator of ethanol extract of the fruiting body of Ganoderma lucidum. The aim of the study was to determine the effect and effective dose of the ethanolic extract of the fruiting body of G. lucidum on reducing the toxicity effect of cadmium in male albino rats (Rattus norvegicus) Wistar strain. The research design was experimental research, which consisted of 5 treatment levels. The research parameters were blood cadmium, β2-microglobulin, malondialdehyde, and superoxide dismutase levels. The ethanolic extract of fruiting body of G. lucidum was tested by gas chromatography-mass spectrometry (GCMS) to determine linalool as active compounds. The results of each parameter had a significant effect, which decreased blood cadmium, β2M and MDA levels, and increased SOD level that linear according to the addition of the dose of ethanol extract of the fruiting body of G. lucidum. Blood cadmium levels with β2M and MDA levels had a positive correlation, while blood cadmium levels with SOD levels had a negative correlation. The dominant compound detected was linalool which has potential as a radical scavenger. The dose of 750 mg.kgBW-1 is the effective dose of the ethanolic extract of fruiting body of G. lucidum based on a decrease in blood cadmium levels (54.10%), β2M (63.94%) and MDA (20.31%), as well as an increase in SOD levels (14.20%) compared to sick control.
