Abstract

The aim of this research was to know the effect of supplemented Spirulina in diet on nilem fish haematologys and to obtain the best dose. Pre-treated fishes were acclimated to the laboratory environment for 10 days then allocated randomly into 4 treatment groups. Each group consisted of three aquaria containing 100 L of water equipped with recirculatory system, 12 fishes were allocated into each aquarium. Fish in group A served as control given pellet without Spirulina. Fishes in groups B, C, and D were given pellet supplemented with 2, 4, and 6 g.kg-1 Spirulina, respectively, given daily at 07.30 and 16.00 at 5% of body weight. The results showed that haematologys profile on the nilem fish was affected by Spirulina. This was indicated by increasing erythrocyte counts, total leucocyte, and haemoglobin level as well as haematocrite value. Supplement with 4 g.kg-1 Spirulina was the best dose.

Abstract

Kelompok Pembudidaya Ikan (Pokdakan) binaan Balai Benih Ikan (BBI) Majenang Kabupaten Cilacap, belum memiliki pengetahuan dan keterampilan pembenihan ikan dengan teknik pijah induksi, sehingga perlu dilatih agar mampu mengelola pembenihannya secara optimal. Penguatan kapasitas individu dan kelompok dapat dikembangkan dengan penerapan strategi yang tepat dan terencana. Salah satu konsep yang dapat diterapkan adalah penyuluhan yang dikemas dalam bentuk pelatihan. Terkait dengan hal itu, telah dilakukan pelatihan terhadap Pokdakan binaan BBI Majengan dengan tujuan untuk memberikan pengetahuan dan keterampilan dalam pembenihan ikan nilem (Osteochilus hasselti Valencienes, 1842) dengan pemijahan induksi. Metode yang digunakan adalah partisipatif, yang meliputi tahapan persiapan, pelaksanaan pelatihan, evaluasi, dan pendampingan. Kegiatan berlangsung pada periode Maret-Juni 2021. Hasil kegiatan menunjukkan bahwa anggota Pokdakan mampu mengelola dan melakukan pembenihan ikan nilem dengan pijah induksi menggunakan wadah serbaguna. Keberhasilan dalam memproduksi benih ikan nilem tersebut mengindikasikan telah terjadi peningkatan pengetahuan dan keterampilan, yang sekaligus juga menunjukkan telah terjadi penguatan kapasitas individu serta kelompok.

Abstract

Barred loach is wild fish in which there is no information related to its enzymatic digestive capacity yet. Therefore research on this subject needs to be conducted.  The spectrophotometry method was used to study protease, lipase, and amylase activities in barred loach. The study used 89 fish with an average weight of 3.61 ± 0.26g, and 1.68 ± 0.21g.  The results showed that pH 7.0 was the highest protease activity. Similarly, lipase activity found  between pH 7.0 and pH 8.1. Still, the amylase activity found to be high at pH 8.1. Protease activity also found to be high in the posterior intestine than in the hepato-pancreas and anterior intestine. Lipase and amylase activities did not found differences between hepato-pancreas, anterior intestine, and posterior intestine. In conclusion, the protease, lipase, and amylase activities of barred loach found along the gastrointestinal tract in both large and small fish, which shows that barred loach can digest protein, fat, and carbohydrates in diets better.

Abstract

Eel population in nature reaches critical number, so that culture strategy is urgently needed to fulfil the high demand of this fish. A shortcut to get functional male, which proven difficult to be founded from natural catching, is masculinization. This research aimed to induce masculinization of tropical eel, Anguilla bicolor McClelland supplemented with various doses of 17α-methyltestosterone during a month culture in freshwater (0 ppt) or brackish water (10 ppt).  The eel was grouped and fed with supplementation of 17α-methyltestosterone at various doses, depending upon treatments, namely 0 mg Kg-1 diet (control), 40, 80 or 120 mg Kg-1 diet. Eels size were similar, at approximately 16,78 g ± 0,62 in weight and 25,38 cm ± 0,15 in length were either culture in freshwater or brackish during the experiment for eight weeks. Sex gonad, based on anatomical histological structures, Eye Index and Fin Index were measured after time culture treatment achieved, as well as body length, weight, eye diameter and the length of the pectoral fin were measured. Results showed that supplementation 17α-methyltestosterone 80 mg/Kg diet culture in brackish water has the highest number of male (90%).  This study proven that, the hormone was effective for masculinization in eels, It useful for masculinization in eels. Results proved that the 17α-methyltestosterone highly significant (P<0.01) effect on the Eye index (3.63 – 5.14) and Fin Index (3.03 – 4.08) of eels. This study concluded, that 17α-methyltestosterone more effective in improving the number of males in brackish water than in freshwater culture.

Abstract

The taxonomic status of Anguilla species' in river watershed that empties into Segara Anakan Cilacap is uncertain, thereby making it difficult for further studies to be carried out to determine its genetic in that area. Therefore, this study evaluates Anguilla's taxonomic status and population genetic in Cibeureum and Sapuregel River watersheds. Data were obtained from molecular characterization study using cytochrome c oxidase 1, with fourteen Anguilla specimens collected from two sequenced watersheds. Taxonomic status was determined based on homology and divergence values and monophyly of the samples to the reference species. Meanwhile, genetic divergences among samples to the reference species were calculated based on the Juke-Cantor substitution model in DnaSP6. A homology test was performed using a basic local alignment search tool, with monophyly inferred from the cladogram, which was developed using neighbor-joining and maximum likelihood algorithms in MEGAX with 1000 pseudoreplicates and out-group comparison. Furthermore, population genetic was analyzed through polymorphism, haplotype, nucleotide diversity within the population, divergence, and genetic differences. All calculations conducted in Arlequin 3.5 had Anguilla samples comprising of high (99.23% to 99.84%) to low genetic divergences (0.224% to 1.127%). The result shows that cladogram with all Anguilla samples formed a monophyletic clade with A. bicolor, separated from their taxa. Furthermore, Anguilla samples from both watersheds have low genetic polymorphisms with medium to high haplotype and nucleotide diversity. The population's comparison proved that both populations have low genetic divergence, and no genetic difference based on variance analysis (p=0761). Therefore, Anguilla resources in river watersheds that empty into Segara Anakan are a single genetic conservation unit.

Abstract

In general, female eel dominates the results of catching eel in the river. Male fish dare rarely found in nature, therefore masculinization is necessary for obtaining in males.  The administration of 17α-methyltestosterone  to masculinize Anguilla bicolor McClelland.   It is a synthetic anabolic-androgenic steroid which has potential to endocrine disrupter that disturbed function of normal reproduction in human or animal. It urgently needed that a masculinization technique needs to study the use of an environmental factor. Population density is one of the environmental factors that influence gender determination (ESD-environmental dependent sex determination). This will result in increased cortisol secretion, which will further stimulate the synthesis of 11-KT steroids that affect male gonad differentiation. This study aims to induce masculinization in tropical eel Anguilla bicolor McClelland in different density. Three treatments and three replicates conducted the research. The treatments were one fish.48 L-1, two fish.48 L-1   and three fish.48 L-1.  Eels size was approximately similar, at 16,78 g±0,62 in weight, and 25,38 cm±0,15 in length were either culture in brackish water for eight weeks. The results showed that density population treatment significantly increased the Fin Index (P<0.05), but no sign for Eye Index, GSI, HSI, and testosterone level. The highest male population (77,8%) achieves at the highest density treatment, three fish.48 L-1 population density. It can be concluded, based on reproductive observations, population density does not affect gonad maturity/puberty, but the high population density (3 fish.48 L-1)stimulates Anguilla bicolor McClelland masculinization.

Abstract

Ikan Sidat, Anguilla bicolor McClelland, pada kondisi alami atau budidaya jarang ditemukan pada fase matang gonad, sehingga upaya untuk melakukan pembenihan buatan sulit dilakukan. Oleh karena itu, diperlukan informasi yang berkaitan dengan aspek reproduksinya, karena masih terbatasnya informasi tersebut. Penelitian ini bertujuan mengetahui profil hormon dan kinerja reproduksi Ikan Sidat. Penelitian ini dilakukan dengan metode survey. Ikan Sidat diperoleh dari perairan segara Anakan Cilacap. Bobot gonad ikan ditimbang untuk mengamati tingkat kematangan gonad (GSI), sedangkan serum hormon steroid diukur menggunakan metode ELISA. Hasil penelitian menunjukkan bahwa nilai GSI (Gonado Somatic Index) berkisar antara 0,1–2,88%, sedangkan kadar hormon estradiol berkisar antara 9–458,94 (pg/ml); Progesteron 0,25–0,57 (ng/ml) dan Testosteron < 0,1 (ng/ml). Dengan demikian dapat disimpulkan bahwa perkembangan gonad ikan Sidat mengalami peningkatan seiring dengan peningkatan serum hormon steroid, khususnya estradiol dan progesteron.

Abstract

Eel, Anguilla bicolor McClelland was hard to mature in captivity, due to low levels of pituitary gonadotropin. In order to stimulate gonadal maturation in captivity, exogenous gonadotropin is needed. The purpose of this study was to assess the effect of GnRH-analog induction on gonadal maturation of tropical eel base on the value of the eye index, gonado-somatic index, and estradiol levels. The study was carried out experimentally with a completely randomized design with three treatments and eight replications. The treatments were the induction of hormone GnRH-analogues with doses of 0.0; 0.5 and 1.0 ml kg-1 body weight. Eel with an average total length of 67 cm and an average weight of 500 g were injected with different doses of GnRH-analog (0.0; 0.5 and 1.0 ml kg-1 body weight), kept in the fiberglass aquarium for two months. The eels were fed with discard fish daily in ad libitum at 16.00 pm. The observed variables were eye index, Gonadosomatic Index (GSI), and estradiol levels. The results showed that injection of three different doses of GnRH-analog did not affect on the increasing of GSI, eye index (P> 0.05), and estradiol (P <0.05). Induction of GnRH-analog of 0.0; 0.5 and 1.0 ml kg-1 body weight can not stimulate the gonadal maturity of tropical eel. Abstrak Ikan sidat Anguilla bicolor McClelland sulit matang gonad dalam kondisi budi daya, karena rendahnya kadar gonado-tropin pituitari. Oleh karena itu untuk memacu pematangan gonad ikan sidat dalam kondisi budi daya perlu ditambah-kan gonadotropin secara eksogen. Tujuan penelitian ini adalah mengkaji pengaruh induksi GnRH-analog terhadap pe-matangan gonad ikan sidat berdasarkan perubahan nilai indeks mata, indeks kematangan gonad (IKG), dan kadar estradiol. Penelitian dilakukan secara eksperimental menggunakan rancangan acak lengkap dengan tiga perlakuan dan delapan ulangan. Perlakuan percobaan terdiri atas induksi hormon GnRH-analog dengan dosis 0,0; 0,5 dan 1,0 ml kg-1 bobot badan. Ikan sidat dengan rata-rata ukuran panjang 67 cm dan rata-rata bobot 500 g sebanyak 18 ekor yang telah diinduksi dengan berbagai dosis GnRH analog (0, 0,5 dan 1 ml kg-1 bobot badan) dipelihara dalam akuarium fiberglass selama dua bulan. Pakan berupa ikan rucah diberikan satu kali sehari pada pukul 16.00 secara ad libitum. Variabel yang diamati adalah indeks mata, IKG, dan kadar estradiol. Hasil penelitian menunjukkan bahwa induksi hormon GnRH-analog tidak berpengaruh terhadap peningkatan nilai indeks mata, nilai IKG (p>0,05), dan kadar estradiol ikan sidat (p<0,05). Induksi hormon GnRH-analog dengan dosis 0,5 dan 1 ml kg-1 bobot badan tidak efektif memacu pematangan gonad ikan sidat.

Abstract

The aim of this research was to know the histology of eel ovary, Anguilla bicolor McClelland, at various gonad developmental stage, which caught at Segara Anakan Cilacap. This research was conducted with survey method, there are 69 samples, about 47.1–76.5 cm in length and 143.6–982.5 g in weight. The parameters are gonad weight, histological structure and gonad maturation level. The result show that all of eels is female, which in yellow eel, and presilver stage. As the result, it can be concluded that Anguilla bicolor McClelland which caught at Segara Anakan Cilacap is immature, whereas mature stage of eel is not found.

Abstract

Hasil budidaya ikan nila pada pembudidaya ikan di Desa Karangnangka kurang optimal karena dilakukan dengan cara polikultur dan mixed sex. Solusi yang diambil untuk mengatasi masalah tersebut adalah dengan pelatihan pembuatan benih nila jantan untuk budidaya satu jenis. Metode pelaksanaan kegiatan bersifat partisipatif dengan tahapan kegiatan yaitu 1) persiapan, 2) pelaksanaan pelatihan, 3) evaluasi, dan 4) keberlanjutan. Kegiatan praktek dan pembuatan demplot dilakukan dengan melatih khalayak sasaran untuk memilih benih jantan dan betina, kemudian membesarkan secara terpisah untuk benih jantan dan betina dan membesarkannya secara seksual di kolam tanah selama enam minggu dengan menggunakan pakan pelet komersial. Seleksi kelamin dilakukan setelah biji memiliki panjang total 6 cm dan sebelum mencapai 12 cm. Hasil demplot menunjukkan bahwa benih ikan nila yang dipelihara dengan satu kelamin jantan memberikan hasil terbaik.
