Abstract

Ganoderma sp. Banyumas 1 isolate that reffered as Ganoderma sp. is a new discovered isolate from Banyumas, Central Java, Indonesia expected to have a potential properties of antioxidant of medicinal mushroom. This study aimed to determine the antioxidant potential and the appropriate solvent for it’s extracting from Ganoderma sp. This research result showed that ethyl acetate was able to extract as many as 15.57%, while etanol was only able to extract 3.87% active compounds from dried 28 days old Ganoderma sp. mycelium cultivated in the Mushroom Complete Medium (MCM). Extract of ethyl acetate (non-polar) extraction of mycelium of Ganoderma sp. had a potential character as an antioxidant source and performed a better result than from ethanolic (polar) extraction as shown in the IC50 value. Extract from ethyl acetate extraction had an average IC50 value smaller than  from ethanolic extract (581.80 < 1285.67). Extract from ethyl acetate extraction resulted in a higher amount of phenol than that ethanolic extract 29.23 < 57.67. Inhibition percentage of both extracts at 65% was known to occur at concentration of 1000 ppm for ethyl acetate extract and 2000 ppm for ethanolic extract. An important finding was that ethyl acetate can be used as appropriate solvent for extracting antioxidant compound better than ethanolic. In conclusion, the mycelium extract of Ganoderma sp. extracted with ethyl acetate and ethanol as solvent is potential to be used as a source of natural antioxidants. This research result has benefit in developing potency of local resources as herbal resources.

Abstract

Diabetes mellitus (DM) is a metabolic syndrome which occurs when insulin is insufficiently produced or insulin cannot well serve its function. Diabetes is marked with increase in blood glucose level followed by increase in glycosylated hemoglobin level and decrease in insulin level. This research aims to examining the effect of Ganoderma lucidum extract on the blood glucose, insulin and glycosylated hemoglobin (HbA1c) level of diabetic white rat and determining the most effective dose of extract to be a diabetic agent. This research was experimentally conducted by employing Completely Randomized Design (CRD) with 6 treatments and 4 repetitions. The treatment groups consisted of healthy rats group (K1), rats with diabetes as negative control (K2), diabetic rats with the administration of metformin 45 mg/kg BW as the comparison (K3) and diabetic rats with the administration of G. lucidum extract with dose of 250, 500 and 1000 mg /kg BW (K4, K5 and K6 respectively). Blood glucose level examination was conducted after the alloxan induction with single dose of 125 mg/kg BW by intraperitoneal injection. The results show that mushroom G. lucidum extract administration with dose of 1000 mg/kg BW (K6) is the best dose to be an anti-diabetic agent. The benefit of the research is developing anti-diabetic agent from herbal resources.

Abstract

Button mushrooms (Agaricus bisporus) is an edible mushroom that is most widely cultivated in the world. It contains bioactive compounds that might provide beneficial effects on diabetes mellitus patient. The study aimed to determine the effects of A. bisporus administration on the blood glucose, and malondyaldehyd (MDA) levels as well as superoxide dismutase (SOD) activity of alloxan-induced diabetic rats. This study was also conducted to determine the secondary metabolites produced by A. bisporus. The method used was experimental methods with Completely Randomized Design. A. bisporus extract at the doses of 250, 500 and 750 mg/kg body weight (BW) per day were orally applied to alloxan-induced diabetic rats for a period of 14 days after the rats became diabetes. The results showed that the extract of A. bisporus could decrease blood glucose, and MDA levels as well as increase SOD activity (p < 0.05). A. bisporus extract 500 mg/kg BW is the most effective dose to be used. Based on Thin Layer Chromatography (TLC) test, it was known that secondary metabolites produced by A. bisporus are flavonoids, alkaloids, terpenoids and saponins. A. bisporus has potential as an antidiabetic through the ability to decrease blood glucose, and MDA levels, as well as increase SOD activity in diabetic rats. This research is able to provide information about the antidiabetic potential of A. bisporus extract so that it can be used as an alternative natural antidiabetic agent and can be applied in the community with ease and in a more controlled industrial scale.

Abstract

Intercellular adhesion plays a role in cancer formation and protein has a key potential in maintaining cell adhesion, including syndecan-1. Meanwhile, oral cancer originates from the oral epithelium, which has an invasive and metastatic level. Its treatments involving chemotherapy and radiotherapy commonly leave unfavorable side effects, hence, suitable alternatives are needed. Natural ingredients are widely used as an alternative treatment for cancer, for example, Ganoderma lucidum (G. lucidum) which has anti-cancer and anti-angiogenic properties, induces apoptosis, stimulates an immune response, inhibits the degradation of Extracellular matrix (ECM), reduces inflammation, affects cell cycles, cytotoxic, and acts as an antioxidant.This study aims to determine the effect of ethanol extract from Ganoderma lucidum Cianjur isolate on syndecan-1 expression in KB CCL-17 oral cell cancer. This was an experimental study with a post-test only control group design, the treatment group used G. lucidum ethanol extract with a concentration of 2.12 μg/ml (P1), 4.24 μg/ml (P2), and 8.49 μg/ml (P3), while the positive control group used cisplatin with a concentration of 11.5 μg/ml (K1). In contrast, the negative control used aquadest (K0), while syndecan-1 expression was observed using the immunohistochemical examination.The highest syndecan-1 expansion rate was found in the treatment group with a concentration of 8.49 μg/ml. A significant difference was indicated by one-way ANOVA (p<0.05) between K0 - K1, K0 - P1, K0 - P2, K0 - P3, K1 - P1, K1 - P2, K1 - P3, P1 - P2, as well as P1 and P3. The administration of ethanol extract from G. lucidum Cianjur isolate increases syndecan-1 expression in KB CCL-17 oral cell cancer.

Abstract

Perkembangan industri protease telah pesat dan menempati posisi penting dalam bidang industri. Protease dapat diperoleh dari semua mahluk hidup, namun saat ini pemakai enzim banyak yang beralih pada enzim mikrobioal. Keunggulan enzim microbial adalah produksi tinggi, kemudahan dan efisiensi dalam pengaturan pertumbuhan, limbah pertanian dapat dimanfaatkan sebagai sumber substrat. Salah satu mikroba adalah Trichoderma spp. Trichoderma sp dapat berasal dari tanah perakaran tanaman (rhizosfer). Jenis tanah perakaran diantarnya adalah jahe, nenas, pisang dan bawang merah. Keempat tanaman ini merupakan tanaman yang banyak dibudidayakan oleh masyarakat. Tujuan penelitian ini adalah menentukan jumLah protease yang terdapat pada Trichoderma spp. Metode penelitian dilakukan dengan kualitatif dan kuantitatif. Uji kualitatif dilakukan dengan mengukur zona bening yang terdapat pada medium proteolitik agar sedangkan uji kuantitatif dilakukan dengan menggunakan spektrofotometer UV-Vis dengan larutan pembanding tirosin. Hasil menunjukkan bahwa jumLah protease yang dihasilkan oleh Trichoderma spp tidak berbeda nyata yaitu. isolat jahe adalah 12,95 U/mL, isolat nenas sebesar 11,86 U/mL, isolat pisang sebesar 11,56 U/mL dan isolat bawang merah sebesar 9,74 U/mL.

Abstract

Desa Wanadadi terletak berbatasan langsung dengan Waduk Mrica. Dampak dari pembangunan waduk adalah terjadinya pergeseran sosial masyarakat dari yang semula mata pencahariannya petani menjadi buruh, dan beternak. Masalah utama yang dihadapi peternak adalah ketersediaan pakan, sedangkan yang dihadapi pengelola Waduk Mrica adalah sedimentasi dan pertumbuhan eceng gondok. Eceng gondok merupakan gulma air yang dapat mengganggu kualitas perairan. Penanggulangan eceng gondok merupakan salah satu upaya dalam manajemen sumber daya dan lingkungan perairan untuk menjaga kelestarian oraganisme parairan. Disisi yang lain tanaman ini memiliki beberapa manfaat dan potensi yang layak untuk dikembangkan, yaitu sebagai bahan pakan ternak karena kandungan proteinnya cukup tinggi, sehingga dapat digunakan sebagai pengganti pakan pabrikan untuk ternak ruminansia maupun unggas. Kegiatan ini bertujuan untuk memanfaatkan eceng gondok menjadi usaha konservasi perairan waduk dan memberikan nilai tambah di masyarakat, yaitu meningkatkan ketrampilan masyarakat dalam pembuatan pakan ternak fermentasi guna mendukung usaha peternakan kambing. Metode yang digunakan dalam kegiatan pemberdayaan masyarakat ini adalah PRA (Partisipatory Rural Appraisal) yaitu peran serta aktif seluruh masyarakat yang terlibat dalam kegiatan ini. Meliputi kegiatan: penyuluhan dan pelatihan pemanfataan eceng gondok sebagai bahan silase pakan kambing, Pembuatan Demplot percobaan ternak kambing, pendampingan dan evaluasi. Diharapkan kegiatan ini dapat memberikan manfaat bagi masyarakat Desa Wanadadi.Kata Kunci: Budidaya Kambing; Eceng Gondok; Silase; Waduk Mrica.AbstractWanadadi Village is located directly adjacent to the Mrica Reservoir. The impact of the reservoir building is a social shift from the people whose livelihoods were originally farmers to become laborers, and livestock. The main problem faced by farmers is the availability of feed, while what is faced by the manager of the Mrica Reservoir is sedimentation and growth of weeds. Eichhornia crasippes is a water weed that can interfere with water quality. Eichhornia crasippes prevention is one of the efforts in the management of resources and the aquatic environment to maintain the sustainability of wter microoganism. On the other hand this plant has several benefits and potential to be developed, namely as animal feed ingredients because the protein content is quite high, so it can be used as a substitute for manufacturing feed for ruminants and poultry. This activity aims to utilize water hyacinth into conservation efforts of reservoir waters and provide added value in the community, namely improving community skills in the manufacture of fermented animal feed to support goat farming. The method used in this community empowerment activity is PRA (Partcipatory Rural Appraisal), namely the active participation of all communities involved in this activity. Includes activities: counseling and training on the use of water hyacinth as goat feed silage material, Making goat experimental demonstration plots, mentoring and evaluation. Abstract

Ganoderma spp. are known as both beneficial and harmful fungi to humans. These are distributed worldwide in sufficiently high diversity. To generate a polymorphic profile and a genetic inter-relationship of several isolates of Ganoderma spp., a study was conducted using the Random Amplified Polymorphic DNA (RAPD) markers on Ganoderma spp. from Banyumas Regency, Central Java, Indonesia. The fruiting bodies of the collected Ganoderma spp. were first morphologically characterized, then analyzed using RAPD with four random primers, i.e., OPC-1, OPC-2, OPC-4, and OPC-5. The results revealed that the four primers generated polymorphic bands of the 10 samples with a polymorphism level of 100%, showing high genetic diversity. The level of genetic similarity ranged between 0.48 and 0.82, indicating moderate similarities among samples. The constructed dendrogram resulted in the grouping of the Ganoderma spp. isolates into three clusters at a similarity coefficient of 0.63, but neither according to geographical locations nor growth substrates.

Abstract

Pelatihan budidaya Jamur Tiram Putih (Pleurotus ostreatus) di Rt.01//Rw06 dan Rt. 02/Rw01 Kelurahan Sumampir Purwokerto Utara ini, bertujuan memberikan keterampilan tambahan guna menambah penghasilan keluarga dan menunjang penyediaan menu harian bergizi. Menggunakan metode partisipatif dengan mengkombinasikan kegiatan ceramah, praktek, pembuatan demplot, dan pendampingan. Khalayak sasaran dilatih membuat rumah jamur, rak tempat baglog, memanen jamur, dan mengolah hasil budidaya menjadi jamur krispi, sop dan kerupuk. Baglog media tanam jamur yang digunakan berasal dari hasil produksi mitra pelaksana kegiatan. Jamur hasil budidaya memiliki kualitas yang baik, putih bersih, satu rumpun dapat mencapai berat 100g, ukuran tudung jamur antara 7-18cm. Hasil panen harian bervariasi antara 0,5-12kg, sehingga cara pemasaran yang diterapkan tergantung hasil panennya. Saat hasil panen berjumlah banyak, dijual di pasar tradisional, saat hasil panen sedikit, dikonsumsi sendiri atau dijual kepada tetangga sekitar rumah. Kegiatan pelatihan ini berdampak positif bagi warga di sekitar khalayak sasaran, karena dapat menunjang menu harian mereka.

Abstract

The Biopulping is defined as the biological process of lignin degradation by utilizing microorganisms that produce some enzymes. A microorganism which widely known in the degradation of lignin and cellulose is a group of white-rot fungi. The aims for this research to know the most effective white rot fungi species of G.lucidum, P.tuber-regium, and T.versicolor in the degradation of lignin and cellulose with different incubation time on bagasse substrate. The effectivity of biopulping indicated by the highest degradation of lignin concentration and the lowest degradation of cellulose concentration. This study used an experimental design with Completely Randomized design with a two factorial pattern. The independent variable of this study is white rot fungi species and incubation time while the dependent variable is the concentration of lignin and cellulose. The main parameter was the concentration of lignin and cellulose, supporting parameters were pH, weight loss of substrate and mycelial growth. The result showed the degradation of lignin and cellulose in all treatment. The best degradation of lignin and cellulose showed by species T.versicolor and P.tuber-regium within 30 days of incubation

Abstract

Agaricales is a group of macrofungi which comprises of many edible and poisonous members. Agaricales is characterized by having a fruiting body that is fleshy in some species and few species are not fleshy, lamellae, and stipe. The group usually grows well in quite moist areas and it cannot be found in dry conditions. The Indonesian Agaricales has not been intensively studied and there is still very few information on its diversity. While in fact, the diversity of Indonesian Agaricales is high due suitable environment and humidity for these fungi to grow. However, there are only a few species that have been identified. Under the circumstances above, this research is aimed to identify the Indonesian Agaricales, particularly from Baturraden Botanical Garden, Banyumas, Central Java and to collect the specimens as dry herbarium. The research involved collection, description, identification, preservation, and documentation of specimens, and the method used a survey with purposive random sampling. The collected Agaricales were identified by using both macromorphological and micromorphological characters. The macromorphological characters as shape, size, color, margin, surface, the flesh of the pileus, lamellae, and stipe were examined. The micromorphological characters as spores and the basidium were examined. Nine specimens had been collected in Baturraden Botanical Garden and were identified to 6 genera that belong to 4 families and 2 unidentified species, namely Hydropus sp., Mycena sp., Panellus sp., Psathyrella sp., Simocybe sp., DRRE 002, DRRE 010.
