Abstract

Actinomycetesis one of the microorganisms which have the ability of enzymes hydrolases such as cellulose, amylase, protease, and manannase. Manufacturer of enzymes of the hydrolase can be used as a candidate probiotic. A candidate probiotic should have a good resistance in different environmental conditions including pH and salinity.  This research is conducted to filter the candidate’s probiotics from bacteria Actinomycetes that tolerance salinity and has the activity of the enzyme hydrolase. Testing the activity of the enzyme was preformed qualitatively on the variation of pH 4, 7, and 9.8.  Theability of Atinomycetesproduce enzymes hydrolases is indicated by the colony on media containing the substrate.From the test results, cellulase enzyme activity with the largest enzymatic index value is 3.5 AF2 isolates pH 9.8. Amylase enzyme activity with the largest enzymatic index value is 3.66 RH1 isolates pH 7. Protease enzyme activity with the largest enzyme index value is 0.66, AF2 isolates at pH 7. Mananase enzyme activity with the largest enzymatic index value is 0.85, namely AF2 isolates pH 4. In testing the effect of salinity test isolates were able to grow at variations in the concentration of NaCl 0, 3, and 6%.

Abstract

Salah satu cara untuk mengenal dan memahami suatu organisme dapat dilakukan dengan mengoleksi organisme tersebut untuk dapat memudahkan pengamatan ciri-cirinya. Hal ini k disebabkan karena hewan bergerak aktif. Oleh karena itu perlu proses koleksi untuk memudahkan pengamatan, baik koleksi kering maupun koleksi basah. Pengetahuan untuk mengoleksi dan mengamati organisme hidup sangat penting bagi guru-guru Ilmu Pengetahuan Alam (IPA) tingkat SMP, untuk membantu pengadaan media praktikum. Untuk itu diperlukan pengetahuan bagaimana cara/ metode mengoleksi hewan, hrwan apa saja yang apa saja yang dapat dikoleksi dan dibuat preparatnya. Dari hasil survei awal, sebagian besar guru IPA –SMP di Lampung Barat belum memiliki ketrampilan dalam koleksi hewan, khususnya serangga. Untuk meningkatkan pengetahuan tentang hal tersebut maka diperlukan bimbingan teknis cara koleksi dan pembuatan insektarium bagi guru –guru yang tergabung dalam MGMP IPA SMP Kabupaten Lampung Barat. Metode bimbingan teknis dilaksanakan dengan pendekatan teori dan praktek pembuatan insektarium. Hasil bimbingan menunjukkan terjadi peningkatan pengetahuan peserta sebesar 48,4% yang ditunjukkan dari persentase sebelum bimbingan dilaksanakan , pengetahuan peserta tentang pembuatan insektarium sebesar 40%. Setelah pelatihan pengetahuan peserta meningkat menjadi 88,4%. Selain itu hasil bimbingan, pembuatan insektarium dapat digunakan berulang kali sebagai media pembelajaran bidang IPA Biologi, khususnya media praktikum keanekaragaman hewan.

Abstract

Fusarium sp is a type of fungus that is pathogenic especially on plants. This fungus attacks the pith tissue of the stem through the injured root. Yellow leaf color is an early indication of this fungal infection. An alternative in controlling this fungus could be to use Bacillus sp because these bacteria have growth inhibitory compounds. This study aims to determine the effect of Bacillus sp from Liwa Botanical Garden on Fusarium sp. Antagonist testing using dual culture method on media Mueller Hinton Agar (MHA) with 3 bacterial isolates was tested. The results of the antagonist test showed that the isolate Bacillus sp.TSR 6 had an inhibitory capacity of 19.7 mm with a strong inhibitory category against the fungus Fusarium sp.

Abstract

This study aims to isolate Bacillus that can fight the growth of Vibrio harveyi . Based on the results of the inter-Bacillus competition test show that Bacillus isolates was able to compete and grow with each other on the SWCA media. The challenge test Bacillus bacterial to against Vibrio harveyi bacteria, that Bacillus did not yet produce anti-bacteria on the second day. In the joint culture test method between Bacillus and Vibrio harveyi that Bacillus were able to inhibit the growth of Vibrio harveyi bacteria on the 4th day.
