Abstract

Beta glucan is a polysaccharide that can be found in cereals, yeast, bacteria, and algae. Beta glucan produced by Saccharomyces cerevisiae has the ability to enhance the body’s defense system. The study aims to investigate the antioxidant activity of B glucan produced by three strains of Saccharomyces sp BR 1, BR 2,and SC. Antioxidant activity assay was carried out in vitro on t BHP induced red blood cells (REC) by observing the level of catalase activity, SOD activity and MDA levels as lipid peroxidation product. The investigation was performed on 6 groups: normal control, negative control, positive control (RBC+ t-BHP + vitamin E 100 IU), and samples (RBC+t-BHP) treated with 1.4 µg/mL β-glucan from Saccharomyces strain BR1, BR2 and SC respectively. Saccharomyces cerevisiae strain SC produced the highest β-glucan of 88.70% (glucose equivalent), while the BR1 and BR2 of 22.56% and 46.28% respectively. Antioxidant activity assay showed that the highest increase in catalase activity (581.48%) was given by the B glucan from SC strain, the highest increase of SOD activity and decrease of MDA levels were given by the β- glucan from BR1 strain (359.17% and 48.18%) as compared to negative controls. Statistical test (α = 0.05) showed antioxidant activity of β-glucan from Saccharomyces Sp BR2 and SC did not differ significantly from the activity of vitamin E (100 IU).

Abstract

Investigation on formulation of film coated tablets containing combination of three active substances, i.e. extracts of soursop leaf, mangosteen rind and Ling Zhi mushroom has been carried out, which was expected to have high antioxidant and immunomodulator activities. The research consists of formulation of film coated tablets F1 containing extracts of soursop leaf-mangosteen rind-Ling Zhi mushroom (100:100:125 mg) and F2 (125:75:125 mg) in each 840 mg tablet. Full examination showed that film coated tablet F1 and F2 comply with the 4th Indonesian Pharmacopoeia, except for the dissolution test. Determination of antioxidant activity with DPPH method showed IC50 of F1 tablet was 215.93 μg/mL, IC50 of F2 tablet was 363.86 μg/mL, whereas IC50 of soursop leaf extract 602.64 μg/mL, IC50 of mangosteen rind extract 111.80 μg/mL and IC50 of Ling Zhi mushroom extract 2199.50 μg/mL. Determination of immunomodulatory activity by in vivo carbon clearance method showed a moderate phagocytic index of 1.385 for F1 tablet, 1.469 for F2 tablet, and <1 for Echinacea extract as positive control. Stability test performed based on the α-mangostin content showed that the shelf-life of F1 tablet was 1.865 years, and F2 tablet was 1.842 years. The result showed that film coated tablet F1 and F2 have both synergistic antioxidant and immunomodulatory activities as compared to their respective single extract. The result showed that the antioxidant activity and stability of film coated F1 tablet is better than F2 tablet, but the imunomodulatory activity of F2 tablet is better than F1 tablet.

Abstract

White turmeric Curcuma zedoaria (Berg) Rosc. is useful for treating cancer disease. It is very susceptible to microbial, therefore gamma irradiation technique to reduce microbial contamination is necessary in order to extend the storing period. Gamma irradiation on white tumeric was carried out by cobalt-60 source at doses of 5, 7.5, 10, and 15 kGy. Then they gradually were maeerated with n-hexane, ethyl acetate, and ethanol. The purpose of this research was to study the effect of gamma irradiation by observing parameters on the cytotoxic activity of ethyl acetate extract and active fraction of white tumeric rhizome against L1210 leukemia cells, also profiles of thin layer and HPLC chromatograms of active fraction as anticancer agent. Ethyl acetate extract of irradiated and control samples were the most active to inhibit the growth of L1210 leukemia cells with IC50 value of 4.71 ug/mL. Ethyl acetate extracts from irradiated and control samples were fractionated using column chromatography and obtained 7 fractions (Fr), respectively. Fraction 3 of control sample was the most active fraction with IC50 values 1.43 µg/ ml. The IC50 value of Fr 3 decreased with increasing irradiation doses and chromatogram profiles of radiated samples with doses of > 5 kGy were changed. The maximum radiation dose for white turmeric preservation is 7.5 kGy, at this dose its anticancer efficacy was maintained.

Abstract

Obesity is a health problem in the community because it can cause a risk of degenerative diseases such as type 2 diabetes mellitus, high blood pressure, heart disease, cancer, and atherosclerosis. Unhealthy lifestyles such as lack of physical activity by exercising, a diet high in carbohydrates and fats, can cause fat deposits in the body, especially in the abdomen. The use of chemical drugs such as Orlistat as a weight loss or to help reduce the risk of regaining lost weight, is less effective because it requires a long consumption time and the presence of side effects. Tin leaf content (Ficus carica Linn) is flavonoids, alkaloids, tannins and steroids, tin leaf extract has IC50 150mg/L antioxidant activity. This study was to determine the anti-obesity activity of tin leaf extract in male Sprague-Dawley rats given a high fat diet. 30 Sprague-Dawley rats were classified into 6 groups. The results obtained, in the group dose of 100 mg / kg obtained an average body weight of 381.8 grams, the test group 200 mg / kg obtained an average body weight of 414.5 grams, in the test group 400 mg / kg obtained 387 grams. The conclusion of this study is that the activity of Tin leaf extract (Ficus carica Linn) can be used as an anti-obesity.

Abstract

Exclusive breastfeeding is defined as giving breast milk to a baby without any additional drink, food or beverages during the first 6 months after birth. Today there is a decline in exclusive breastfeeding, and among others, one reason is the increase in the participation of mothers in the workforce, especially in urban areas. Many factors that affect the regularity of exclusive breastfeeding by working mothers, one of which, is the demographic factor. This study aims to obtain profile data of exclusive breastfeeding by working mothers and statistical modelling of exclusive breastfeeding by working mothers based on demographic factor, by conducting a survey on 384 respondents. This research uses descriptive analysis and logistic regression analysis with SPSS software version 15.0. Descriptive analysis showed that most mothers working in Jakarta (61.2%) did not give exclusive breastfeeding. Logistic regression statistical analysis showed that age, occupation, distance to work, and travel time to the workplace of working mothers are dominant factors that influence exclusive breastfeedings, with the p-value is smaller than the p value (Sig) = 0.05.

Abstract

The leaves of tea benalu (Scurrula oortiana (Korth) Danser) were extracted with nhexane, ethyl acetate, methanol, and water respectively. The antioxidant activities were measured using 1,1-diphenyl-2-picrylhidrazyl (DPPH) methods. The result showed that the isolated compound has scavenging effect with IC50 92.22 ppm. Identification by interpretation of ultraviolet, infrared, and nuclear magnetic resonance spectra showed that the isolated compound was cathecin.

Abstract

Formulation of lozenges containing: A) green tea extract; B) green tea extract with sodium ascorbate; and C) green tea extract with grape seed extract have been carried out. Antioxidant activities were compared to the activity of raw active materials. Lozenges were tested for physical characteristics, hedonic values, dissolution rate, and stability during storage, Physical tests indicated that the lozenges comply with the Farmakope Indonesia IV requirements. Determination of IC50 antioxidant activity using DPPI-I free radical scavenger showed that IC50 of formula A, B, and C were 844.04 µg/mL, 221.80 µg/ mL, and C 57.4 µg/mL, respectively. Meanwhile, IC50 value of green tea extract, sodium ascorbate, and grape seed extract were 4.29 µg/mL, 12.95 µg/mL, and 1.96 µg/ml., respectively. Accelerated stability test were carried out based on the EGCG concentration observed by using HPLC and then calculated by Arrhenius equation showed that the shelf life of lozenges of formula A, B, and C Were 2,11 years; 1.87 years; and 2,73 years, respectively. The result showed that the lozenge containing green tea extract and grape seed extract (formula C) is the best formula based on FI IV requirements, antioxidant activity and lozenges stability.
