Abstract

Infectious diseases are estimated to be the cause of half of all deaths worldwide. About 50-75% of deaths in hospital are reported to be due to infection. This number continues to increase rapidly in line with the development of bacterial resistance to certain antibiotics. One of the causes of resistance due to antibiotic abuse. The number of resistance events that occur encourages new discoveries in order to overcome the problem of resistance. Actinomycetes are Gram-positive microscopic bacteria that are widespread in nature. These bacteria can live in various environmental conditions that contain lots of nutrients. Actinomycetes population increases in the presence of organic matter that undergoes decomposition. Acinomycetes have the ability to produce antimicrobial compounds. The need for antibiotic compounds and chemotherapy compounds that are effective with low toxicity to the host, and the resulting waste can be degraded by the environment, is a problem that needs serious attention. The existence of resistance to antibiotics causes the need to develop new types of antibiotics from natural ingredients and microorganisms that can control microbapatogens. Compounds obtained from natural materials, especially plants and microbes, provide promising results in the development of new antibiotic compounds, among the existing types of microorganisms, Actinomycetes are the most potential source of producing antibiotics. Apart from antibiotics, actinomycetes also produce bioactive compounds of high economic value in the health sector as antiviral and anti-cancer, while in agriculture as herbicides, insecticides, and anti-parasitic compounds. This causes the need to explore actinomycetes which have the potential to produce antibiotic compounds against bacteria. This study aims to determine the antibacterial activity of secondary metabolites of actinomycetes isolates against cuts infected with Staphylococcus aureus resistant methicillin (MRSA) in winstar rats (rattus norvegicus). This type of research is carried out in an experimental laboratory. The results showed that the potential for secondary metabolites of actinomysetes isolates against MRSA bacteria was classified as strong with an inhibitory index of 10.1.

Abstract

The degree of smoking habits in the urban community is still high, although carried out health promotion of harmful substances in cigarettes, one of which is cadmium (Cd). The dangers of cadmium in cigarette smoke causes harm to active and passive smokers. Increased exposure of cigarettes will lead to an increase in the burden of disease in groups at risk such as people with Diabetic Type 2 (DMT2). The purpose of this study was to determine the differences in levels of cadmium (Cd) in the blood of active smokers and passive smokers patients with DMT2 in Puskesmas Kedungdoro Surabaya. The type of this research is a comparative with cross-sectional study design conducted in the Toxicology Laboratory of the Department of Health Analyst and Research and Standardization Center of Industry in August 2020-May 2021. The independent variable in this study is active smokers and passive smokers patients with DMT2, and the dependent variable in this research is the content of cadmium (Cd) in the blood. Analysis of the levels of cadmium in the blood using Atomic Absorption Spectrophotometer by the method of acidifying carbon furnaces. The results showed range levels of cadmium in blood in 8 people suffer from Type 2 Diabetic as active smokers are 0.605-1.5535 µg/dL. While the range levels of cadmium in the blood of the 8 people who suffer from Type 2 Diabetic as passive smoking are 0.5648-2.7045 µg/dL. Based on the results of statistical tests using test Independent Sample T-Test, it can be concluded that there is no difference in the levels of cadmium (Cd) in active smokers and passive smokers patients with DMT2 in Puskesmas Kedungdoro Surabaya.

Abstract

Kidney failure is caused by a permanent decline in kidney function and requires renal replacement therapy, one of which is Continuous Ambulatory Peritoneal Dialysis (CAPD). This study aimed to determine the relationship between albumin concentrations and sodium and potassium concentrations in CAPD patients. The study was carried out between January to May 2022. Cross-sectional research methodology is used in this study. It included 60 chronic kidney failure patients who had CAPD as therapy at PHC Surabaya Hospital. Then, patients were divided into three groups with an age interval of 40-50 years, 51-60 years, and 61-70 years in. The results of grouping CAPD patients according to the characteristics based on the age of CAPD patients aged 61-70 years had low albumin concentrations and low sodium concentrations A total of 12 people (66.67%), patients with large CAPD aged 51-60 years, had low albumin and potassium concentrations, namely 11 people (61.11%). CAPD patients aged 51-60 years mostly had low albumin and sodium concentrations in 13 people (72.22%). the conclusion is that in patients with renal failure on CAPD therapy, there is a decrease in albumin and sodium-potassium concentrations due to peritonitis (inflammation) which is associated with a measurable excess of fluid (overhydration) which causes more dilute sodium, causing hyponatremia and hypokalemia.

Abstract

Motor vehicles emit exhaust gas emissions that produce pollutants, one of the compounds contained is carbon monoxide (CO). CO may cause intoxication in humans if exposed for a long time especially street vendors because the work area is filled with vehicle fumes containing CO gas. High CO levels in the air indicate HbCO levels to increase. The increased of HbCO levels will cause abnormal hemoglobin levels and induce vasoconstriction which makes blood pressure increase. The purpose of this study was to prove the correlation between CO levels in the air with HbCO, hemoglobin, and blood pressure levels to street vendors in the Gading Fajar Sidoarjo Street Vendor Area. This research uses the Conway diffusion cell method and is an analytic observational study with a cross sectional design implemented at the Surabaya Health Laboratory Center and the Hematology Laboratory of Medical Laboratory Technology Department Poltekkes Kemenkes Surabaya in March 2022. Blood tests was carried out on 25 street vendors using random sampling technique. The results showed that average air CO level was 11.0 ppm and 13.3 ppm; mean HbCO levels 0.87%; mean hemoglobin levels 14.4 g/dL; mean systolic blood pressure levels 128 mmHg; and the mean of diastolic blood pressure levels 80 mmHg. The data were analyzed using SPSS. The outcome of normality test showed that the data was not normally distributed. Statistical tests was performed using Spearman correlation. The conclusion showed there was correlation between air CO level with HbCO level, hemoglobin level, and blood pressure level on street vendors in the Gading Fajar Sidoarjo Street Vendor Area. If CO levels in the air increase, then HbCO levels and blood pressure also increase. Street vendors are expected to always use Personal Protective Equipment (PPE) such as mouth masks when working to reduce the risk of exposure to CO gas and doing a healthy lifestyle as a preventive effort to avoid health problems.

Abstract

The prevalence of MRSA in various hospitals in the world ranges from 2-70% with an average of 20% .In recent decades there has been an increasing prevalence of S. aureus and MRSA in the world. Population-based studies in North America and Europe indicate the prevalence of S. aureus is between 18-30%. The overall prevalence of MRSA in Asia has reached 70%, while publications and prevalence of MRSA in Indonesia are still very limited and very difficult to obtain. The prevalence of MRSA in Atmajaya Hospital, Jakarta, reached 47%. Yuwono also reported in 2010 MRSA at Dr. RSUP. Moh. Hoesin Palembang reached 46%. Amplification were carried out under PCR conditions as a result of optimization, namely the results of PCR optimization were carried out for 30 cycles with the following thermal conditions. Denaturation phase at 94oC for 30 seconds, annealing at 52.0oC for 30 seconds, and extension at 72oC for 1 minute. Amplification results showed that of the 30 samples only 90% (27 samples) were detected to have the Mec-A gene, 10% (3 samples) were negative. Keywords: Mec-A; gene; Methicillin Resistant Staphylococcus aureus

Abstract

Lead is one of the heavy metals that can cause health problems and is widely found in the environment. Lead level examination can use specimens of hair, urine, and blood. Hair can be used as an indicator of examination for lead poisoning that is internal and external. While urine is a screening test on lead poisoning. And the blood specimens is an overview of the level of lead absorbed by the body. The purpose of this research was to look at the content of lead levels in specimens of hair, urine, and blood of TPS 3R Sutorejo garbage officers. This research is a descriptive research with cross sectional design conducted at Balai Besar Laboratoriun Kesehatan Surabaya in October 2020 – June 2021. The samples used were 10 garbage officers who were taken using purposive sampling techniques. The results showed that lead levels in the hair were between 0.108 μg/g – 0.239 μg/g, in urine 0.000095 mg/L – 0.00121 mg/L, and in blood 1,665 μg/dL – 4,785 μg/dL. All respondents (100%) have lead levels in specimens of hair, urine, and blood in the normal category. Examination using blood and urine specimens can be used to determine the level of lead that is collected in a short period of time. While in the long term can use hair specimens. Timbal merupakan salah satu logam berat yang dapat menyebabkan permasalahan kesehatan dan banyakditemukan di lingkungan. Pemeriksaan kadar timbal dapat menggunakan spesimen rambut, urin, dan darah.Rambut dapat dijadikan indikator pemeriksaan untuk keracunan timbal yang bersifat internal dan eksternal.Sedangkan urin merupakan screening test pada keracunan timbal. Dan darah menunjukkan gambaran kadartimbal yang terserap oleh tubuh. Tujuan dari penelitian ini adalah untuk melihat kandungan kadar timbal dalamspesimen rambut, urin, dan darah petugas sampah TPS 3R Sutorejo menggunakan Spektrofotometri SerapanAtom. Penelitian ini merupakan penelitian deskriptif dengan rancangan cross sectional yang dilaksanakan diBalai Besar Laboratorium Kesehatan Surabaya pada bulan Oktober 2020 – Juni 2021. Sampel yang digunakanadalah 10 orang petugas sampah yang diambil menggunakan teknik purposive sampling. Hasil penelitianmenunjukkan bahwa nilai kadar timbal dalam rambut antara 0,108 µg/g – 0,239 µg/g, dalam urin 0,000095mg/L – 0,00121 mg/L, dan dalam darah 1,665 µg/dL – 4,785 µg/dL. Semua responden (100%) memiliki kadartimbal dalam spesimen rambut, urin, dan darah dengan kategori normal. Pemeriksaan menggunakan spesimendarah dan urin dapat digunakan untuk mengetahui kadar timbal yang terabsorbsi dalam jangka waktu pendek.Sedangkan pada jangka waktu lama dapat menggunakan spesimen rambut.Kata kunci : Timbal; Petugas sampah; Rambut; Urin; Darah; Spektrofotometri Serapan Atom

Abstract

Lead (Pb) is a heavy metal that can cause poisoning and accumulate in the human body. Determination of lead levels by using AAS the condition is that the sample must be a solution, then it is necessary to destruct first. There are two types of destruction that can be done, namely wet destruction using acid reagents to decompose samples and dry destruction by heating or destruction using very high temperatures. HNO3 oxidizing agent and HNO3 mixture with HClO4 are the main oxidizing agents that can provide the best destructive results compared to other oxidizing agents. This study aims to determine the best oxidizing and destructive substances present in the hair using Atomic Absorption Spectrophotometry (AAS). The research was conducted at surabaya regional health laboratory in April - May 2021. This research is comparative research. The sample used is the hair of gelam gas station operator officer, Candi. There are two ways of preparing samples that can be done in pb analysis on hair, namely proses wet and dry destruction. The result of destruction is determined by the level of lead using Atomic Absorption Spectrophotometry (AAS). The results of the analysis pno dry destructionHNO3 as a whole has rentang lead levels 0.1281-0.1958 mg / L. Dry destruction HNO3 : HClO4 secara overall has rentang lead levels 0.1457-0.2159 mg /L. HNO3 closed wet construction has lead content range 0.1431-0.2034 mg/L and wet destructive closed HNO3 : HClO4 has lead content range 0.1080-0.5371 mg/L. Based on these results, it can be known that all hair samples tested in this study contain lead levels that are still within normal limits or within the tolerance limit of < 12 μg/g.

Abstract

 Lipemic serum can interfere with examination parameters that require light transmission and may lead to inaccurate analysis results. One of the tests carried out in the clinical laboratory is total protein. Handling of lipemic serum can be done by several methods including centrifugation and precipitation. Precipitation is carried out with cyclodextrins which are water soluble and can form complexes with water molecules, and are effective in overcoming disturbances of lipid particles in lipemic serum. This study aimed to compare and analyze total protein levels in lipemic serum treated with alpha cyclodextrin, gamma cyclodextrin, and high-speed centrifugation. This study was conducted at Surabaya Jemursari Islamic Hospital Laboratory in October 2021-May 2022 and aimed to analyze the difference in total protein levels in lipemic serum that has been treated with 20% alpha cyclodextrin, 20% gamma cyclodextrin, and with high-speed centrifugation treatment. The method used in this study was a pretest-posttest design, involving 9 lipemic serums in total that were visually visible. Determination of this study was performed by statistical analysis using the SPSS application. The asymptotic value obtained was 0.248 for alpha cyclodextrin treatment; 0,125 for gamma cyclodextrin treatment; and 0,229 for high-speed centrifuge treatment. Since the obtained asymptotic value was >0.05. It can be concluded that there was no significant differences in the treatment of lipemic serum with the treatment of alpha cyclodextrin, gamma cyclodextrin, and high-speed centrifugation.Keywords: cyclodextrin; high-speed; centrifugation; lipemic serum; total protein; precipitation

Abstract

The Hb meter is a tool for checking hemoglobin levels using the Point of Care Testing (POCT) method using a small volume of capillary blood samples. Quality control of the Hb meter is needed to see the accuracy and precision values so that the suitability of the tool can be determined. Accuracy is the accuracy of the test results close to the true value and precision is the accuracy of repeated measurement results. The purpose of this study was to determine the accuracy and precision of hemoglobin examinations using two brands of POCT methods on a Hematology Analyzer. The type of research used was a comparative analytical study with a cross-sectional approach. This study was conducted at a community health center, using 30 test materials from outpatients. The measurement results were analyzed descriptively, then continued with ANOVA tests. The results showed that the accuracy value of the POCT method of the HemoCue Hb 301 brand obtained a bias (d%) of 1.8% with a recovery of 101.2%. While the Onemed Hb Check brand obtained a bias (d%) of 8.3% with a recovery of 108.3%. The precision value of the hemoglobin level using the HemoCue Hb 301 POCT method achieved a CV of 1.95%, while the Onemed Hb Check achieved a CV of 2.96%. Furthermore, it can be concluded that the accuracy of the HemoCue Hb 301 and Onemed Hb Check POCT methods on the Hematology Analyzer is good.Keywords: hemoglobin; Hb meter; accuracy; precision; hematology analyzer ABSTRAK Hb meter merupakan salah satu alat pemeriksaan kadar hemoglobin metode Point of Care Testing (POCT) menggunakan sampel darah kapiler dengan volume yang sedikit. Diperlukan pengendalian mutu Hb meter untuk melihat nilai akurasi dan presisi sehingga dapat diketahui kelayakan alat tersebut. Akurasi adalah ketepatan hasil uji mendekati dengan nilai benar dan presisi merupakan ketelitian hasil pengukuran yang berulang. Tujuan penelitian ini adalh untuk mengetahui akurasi dan presisi pemeriksaan hemoglobin menggunakan dua merek metode POCT terhadap alat Hematology Analyzer. Jenis penelitian yang digunakan adalah studi analitik komparatif dengan pendekatan cross-sectional. Penelitian ini dilaksanakan di puskesmas, menggunakan 30 bahan uji dari pasien rawat jalan. Hasil pengukuran dianalisis secara deskriptif, lalu dilanjutkan dengan uji ANOVA. Hasil penelitian menunjukkan bahwa nilai akurasi metode POCT merek HemoCue Hb 301 memperoleh bias (d%) sebesar 1,8% dengan recovery sebesar 101,2%. Sedangkan merek Onemed Hb Check memperoleh bias (d%) sebesar 8,3% dengan recovery sebesar 108,3%. Hasil nilai presisi kadar hemoglobin metode POCT merek HemoCue Hb 301 memperoleh CV sebesar 1,95% sedangkan merek Onemed Hb Check memperoleh CV sebesar 2,96%.
