Abstract

Induction of Strawberry callus (Fragaria sp.) Through Application of 2,4 dichlorophenoxy acetic acid in vitroThe study aims to know the concentration of 2,4 Dichlorophenoxy acetic acid for the growth of callus the strawberries. This study was conducted at laboratorium of tissue culture of agriculture faculty, Udayana on January until May 2015. The compiled using a Randomized Complete Block Design (RCBD) with five repetition and five level of treatment combination that is (D0,D1,D2,D3,D4,D5). If in the result found significant effect then followed up with duncan test 5%. Data that can not be analyzed tabulated and discussed descriptively.The result shows that treatment of D2 give best growth that it showed at percentage of highest deflection (54,765), fastest swelling (12,70 hst), highest swelling procentage (49,34%) and lowest browning procentage (43,05%). Beside that eksplan able to produce callus at D2 treatment with combination of MS media and by giving of 2,4 D 1 ppm.

Abstract

The Influence of Processing Time Delay To Rice (Oryza Sativa L.) Seed Quality of Membramo Variety This research was performed with the purpose to obtain tolerance of processing time delay after appropriate crop so it doesn’t affect the physical and physiological quality of rice (Oryza Sativa L.) seed.This experiment used the Complete Random Design (RAL) by using 8 treatments and 3 repetitions. Therefore, there are 24 experiment units. Research result data were tabulated and analyzed mode investigation according to design that was used, and if the treatment has significant influence then it is proceeded with Duncan’s multiple distance test (5%).Observation was carried out to water content variable during the crop (%), water content during the processing (%), water content during the test (%), the viability test/power of germination (%) and electrical conduction power test (? mhos cm-2 g1).Research results shows that the longer processing time delay the more increase clear water content, the lowest water content at the direct harvest processing and the highest at processing time delay at the seventh day. So thus the longer processing time delay the viability processing will decrease. The processing time delay until 2 days after the crop the rice seed viability is more than 80% (above quality standard, ISTA), the next delay the seed viability is already under standard. The electrical conduction power more increase by the longer seed processing time delay.

Abstract

Mikropropagation Plant Strawberries (Fragaria sp.) Through Induction of Organogenesis Problems for farmers strawberry in Bali are the difficulty of get healthy strawberries  seeds a with prices are relatively cheap. Micropropagation method is ways to multiplay seeds for solving the problem. Research aims to understand the influence of concentration ZPT NAA + BAP to organogenesis on explant leaves strawberries in vitro. Experiment arranged using a Randomized Block Design (RBD). Medium the base used is MS medium with adding of 2 gL- 1 of activated charcoal, and 0.1 ppm NAA. As the treatment is BAP with concentration of 5 ppm, 10 ppm and 15 ppm with 5 replications. The results showed that the use of 0,1 ppm NAA + 15 ppm BAP most effective in deflection (3,0 DAP) with the percentage of 90,00%. The use of the NAA 0,1 ppm + 15 ppm BAP also most effective in a time of swelling (7,6 DAP) with the percentage of 82,95 % , while only the use of the NAA 0,1 ppm + 5 ppm BAP that will be capable of eliciting shoots at 23 DAP time.

Abstract

Induction can Callus Eksplan the Vine (Vitis vinivera L.) With The Application 2,4-D in In Vitro This research aims to find the influence of the provision of 2,4-D concentration on the growth of the explant callus of grapevine leaves by in vitro treatment and to know how much the concentration of the provision of 2,4-D needed to the shape and growth of explant callus of vine leaves. This research was conducted in April until August 2016 in the Laboratory of Plant Tissue Culture, Program Study Agroekoteknologi, Faculty of Agriculture, Udayana University. The method used was Complete Random Method (CRM). The variable observed in this research was when the explant got curvy, the total percentage of the curving explant, when the explant got swollen, percentage of the swollen up explant and the percentage of brownish explant. Influence of the ZPT 2,4-D provision gives real influence to the observed variable to the variable when the explant got curvy and the percentage of brownish explant, affects really real to the total percentage of the curved explant.MS + 1,5 ppm 2,4-D treatment gives the best growth seen at the variable when the explant curves the fastest (2,4 days), the highest percentage of the curved explant (93,3%) and the highest percentage of the swollen explant (20,0%). This treatment which successfully creates is 1,5 ppm 2,4-D treatment.

Abstract

Study of Grafting Time on the Success of Grafting on Wani Ngumpen Bali (Mangifera caesia Jack. Var. Ngumpen Bali) Wani ngumpen Bali is a mango relative plant that grows in Bali and potential to be widely developed because it has unique and superior properties because 90% of the fruit produced has small seeds and thick flesh that makes it seem like it has no seeds. Considering the wani ngumpen Bali plant is difficult to propagate through generative because the seeds are relatively difficult to get so that propagation is done using vegetative cultivation technology. One of the vegetative propagation is grafting or connecting shoots. This research aims to study the timing of splicing to the success rate of wani ngumpen Bali shoots. The study was conducted from February to April 2021. This study was conducted in Sudaji Village, Sawan Subdistrict, Buleleng Regency, with an average height of 500 mdpl, rainfall of 3000 mm per year with a temperature of 18o-23 o C.This study used a randomized block design (RBD) with 1 factor consisting of 5 levels treatment time grafting. The treatment was repeated 7 times and in each replication there were 2 plants, so that there were 70 experimental units, with treatment W1 (at 09.00-10.00 am), W2 (at 11.00-12.00 pm), W3 (at 13.00-14.00 pm), W4 (at 15.00-16.00 pm), W5 (at 17.00-18.00 pm). The data obtained were analyzed descriptively because the research data did not meet the requirements for statistical tests. In the nursery, shoots of wani var. Ngumpen Bali is affected by the connection time. The success of shoot grafting in wani ngumpen Bali, best during the day, at 13.00-14.00, with percentage of seedlings successfully growing 35,51%, shoot length 21.10 cm and number of leaves 13 strands.

Abstract

Influence of 2,4-D to induce callus from strawberry leaves on alternative media of AB Mix hydroponic nutrientThis research was conducted to find out the most optimum 2,4-D concentration to induce callus from strawberry leaves on alternative media of AB Mix hydroponic nutrient. This research used Completely Randomized Design with 5 levels of treatment concentration of 2.4-D hormon, first: AB Mix hydroponic nutrients (as basic media) without 2,4-D, second: basic media + 0.5 ppm 2.4 -D, third: basic media + 1 ppm 2,4-D, fourth: basic media + 1.5 ppm 2,4-D, fifth: basic media + 2 ppm 2,4-D. The variables observed were the day explant curled up, day explant swelled, percentage of eksplan curled up (%), percentage of explants swelled (%), the day callus appears, and percentage of explant browning (%). The best percentage of the treatment in callus induction of the strawberry leaves is on the P3 treatment with the concentration of 2,4-D as much as 1.5 ppm, with the fastest curled up explants time of 3 days after culture, the highest curled up eksplants (93.3%), the fastest swelled explants (17.0 days after culture), and the highest swelled percentage (40.0%). Callus formed 32 days after swelling. Successful callus formed only in treatment basic media with 1,5 ppm 2,4-D concentration.

Abstract

Growth of Plantlets of Cattleya Orchid on The Fooliar Fertilizer-Based Medium added with Benzyl Amino Purine. The aim of the current research was to investigate the appropriate concentrationof plant growth regulator BAP on the growth of Cattleya plantlet. The experiment was utilized a Randomized Completely Design with five treatments and six replications. The basal medium used was modification of foliar fertilizer of Growmore (trade mark) with addition of fish emulsion, Vitamin B1 and active charcoal. BAP concentration used as treatment were 0 ppm, 0.5 ppm, 1.0 ppm, 1.5 ppm and 2.0 ppm. Variables observed were height, leaf number, root number, root length, fresh weight and dry weight of plantlets. The result showed that concentration of 1 ppm BAP resulted in the highest value for plantlet height (5.67 cm), leaf number (4.67), root length (2.07 cm), fresh weight (0.36 g) and dry weight (0.043 g).

Abstract

Growth of Plantlets of Vanda tricolor Orchid In-Vitro on the Cassava Pulp-Enriched MS Medium.Addition of organic matter into the culture medium is commonly used for increasing the source of sugar, vitamins and amino acids. The aim of this study was to determine the effect of the cassava pulp on the growth of Vanda tricolor plantlet, and to investigate appropriate dose for the optimal growth of plantlets. The experiment utilised a Randomized Completely Design with six treatments i.e. UO (MS medium without any organic matter), U1 (MS medium + 35 g/L cassava pulp), U2 (MS medium + 40 g/L cassava pulp), U3 (MS medium + 45 g/L cassava pulp), U4 (MS medium + 50 g/L cassava pulp), and U5 (MS medium + 45 g/L cassava pulp) with five replicates. The result showed that addition of cassava pulp significantly increased number of leaf, number of root and dry weight of plantlets. The highest number of root (i.e. 5.8) was obtained with addition of 50 g/L cassava pulp, while the highest dry weight of plantlet (i.e. 0.28 g) was on addition of 55 g/L cassava pulp.

Abstract

In Vitro Transformation of Flowering Gene through Agrobacterium tumefaciens on Vandatricolor Orchid. Research concerning of Agrobacterium-mediated transformation of Vanda tricolor orchid had been done during March to August 2014. Objectives of this research wasto investigate the effects of period of inoculation with Agrobacterium suspension and application of acetosyringone (AS) on the percentage of transforman candidates. The T-DNA harbored the PaFT gene under the control of promoter of Ubiquitin and the hpt gene, ahygromycine resistant gene as a selectable marker for transformant selection. This T-DNA was constructed in the PGAS vector and transferred to the plant genome mediated by A. tumefaciens. The period of inoculation with Agrobacterium and application ofAcetosyringone (AS) were trialed in this research. The result showed that a period of one hour inoculation with Agrobacterium suspension added with 25 ppm AS resulted in the highest number of transforman candidates. i.e. 7.04% compared to 4.10% (one hour inoculation without AS), 3.76% (two hours inoculation without AS), and 4.48% (two hours inoculation with 25 ppm AS).

Abstract

Callus Induction with Aplication of 2,4-D in Micropropagation of Strowberry(Fragraria xananassaDuch cv. Rosalinda). Strawberryis grown and well planted in many countries and has ahigh economic value. In Bali,thecentraproduction is in Pancasari Village,Sukasada District,BulelengRegency. The limited number and quality of seedling are the problems currently faced bystrawberry farmers at Pancasari Village. Seedlings produced by conventional propagation are limitedand are not able to meet the needs of seedling available for planting area. This study offers a solution tothe problem by applyingmicropropagationtechnique through callus induction. The research aim is tofind the optimal concentration of 2,4 - D to induce callus of strowberry. Callus that forms will be usedas the material for the next growing into young plants regenerated seedlings. This experiment used acompletely randomized design with 2 factors. The first factor was theconcentrations of 2,4 - D withfive levels of treatments, i.e.0 ; 0.5, 1.0, 1.5, and 2.0 ppm and the second factor was type of explantsused, including tip , middle and base of leaf. Results showed that2,4-D concentration of 1 and 2 ppmapplied on the base of the leaf is able to induce 100 % callus at 14 days after planting. Callus formedwas greenish white. The callus will be regeneratedinto plantlets or young plants.
