Abstract

The Growth Response of Orchid Plantlets of Dendrobium sp. by Aplicación of Bean Sprouts Extract and Coconut Water in Organic Medium The aim of this researh is to obtain the effect of coconut water and bean sprouts extract and the best combination added to organic medium on growth of orchid planlets in vitro. The experiment was conducted at Tissue Culture Laboratory of Agriculture Faculty, Udayana University, Pegok Denpasar from Mei until Oktober 2012. The research design was Completely Randomized Design (RAL) with ten treatments and six replications. The results of this ezperiment showed that addition of coconut water at concentration of 150 ml/l and bean sprouts extract of 50 ml/l resulted in the best growth of orchid planlet which includes the height of plantlets (4,10 cm), the number of leaf (3,33 leaves), length of root (1,07 cm). The total fresh weight of plantlet was 0,20 g and total oven dry-weight of plantlet was 0,021 g.

Abstract

Lygodium circinnatum (Burn.f.) Swartz is a wild Pteridophyta which is used as a material for handicraft. Recently, Lygodium has been rare in nature due to overgathering. Domestication of this wild plant may required to overcome this problem. A research concerning domestication of Lygodium was conducted at Faculty of Agriculture of Udayana University during March to October 2009. Dose of urea fertilizer in varied levels was applied to 8 month-old sphorophyte. The level of urea doses were 0, 100, 200, 300 and 400 mg urea per plant. The results showed that urea fertilizer significantly stimulated the number of “sulur” (“sulur” is an organ of Lygodium that used as material for handicraft) , but did not affect the number of leaves and plant height. Dose of 300mg urea per plant resulted in the highest number of the average of “sulur” number per plant.

Abstract

Micropropagation of Phalaenopsis Orchid Using Explants of Flower Stalk. This studyaimed to obtain optimum media formulations for bud multiplication of micropropagation ofPhalaenopsis orchid using flower stalk. This study employed Completely Randomized Designwith 5 different treatments, i.e. M0 = MS + 150 ml/l coconut water + 2 g activated charcoal,M1 = MS + 1 ppm IBA+ 1 ppm BAP + 150 ml/l coconut water + 2 g/l activated charcoal M2= MS + 1ppm IBA+ 2 ppm BAP+ 150 ml/l coconut water + 2 g/l activated charcoal, M3 =MS + 1 ppm IBA+ 3 ppm BAP + 150 ml/l coconut water + 2 g/l activated charcoal, M4 = MS+ 1 ppm IBA+ 4 ppm BAP + 15 0ml/l coconut water + 2 g/l activated charcoal. Eachtreatment consists of 5 replicates, so there were 25 units experiment, with 3 explants each.Results of this study revealed that M2 media exhibited the best growth, as shown by swellingbuds (23.77 Days After Planting/DAP), earliest buds formation (49.33 DAP) and height ofbuds of 2,12cm.

Abstract

Callus Induction on Phalaenopsis Pollen using 2,4-Dichlorophenoxyasetic acid. Tissue culture is one of the modern methods of plant propagation. In tissue culture hormone has an important role. The goals of this research was to determine the effect of 2,4-D hormone on callus growth of Phalaenopsis pollen explants on NP media and to know the most optimal concentration for the formation and growth of explants. This research used the complete randomized design with five treatment of 2,4-D dosage such as 0 ppm, 0.5 ppm, 1 ppm, 1.5 ppm, and 2 ppm. The variables observed in this research was time of explant swollen, percentage of explant swollen, time of explants detached, percentage of explants detached, time of callus growing, percentage of explants grow callus, callus colour, and callus texture. The result of analysis was 1,5 ppm of 2,4-D treatment gave the best effect to explants, with the fastest swollen explants time of 3 days after culture, the highest swollen explants (86.6%), the fastest detached explants (7.4 days after culture), and the highest detached percentage (66.6%), time of explant growth callus (28.4 days after culture) and percentage of explant growth callus (53.3%).

Abstract

The Effect of Combination of BAP-NAA and Explant Types on Micropropagation of Orchidof Vanda tricolor Lindl. var. suavisThe objectives of the research was to investigate the effect ofcombination of NAA–BAP, type of explant and their interactions on shoot initiation in micropropagationof Vanda tricolor L. var. suavis. The factorial experiment consisted of two factors that was arrangedby completely randomized design (CRD) with three replications. The first factor was combination ofNAA-BAP i.e. M0= MS medium without any plant growth regulator; M1= MS + 0,1 mgL-1 NAA +0 mgL-1 BAP; M2= MS + 0,1 mgL-1 NAA + 0,1 mgL-1 BAP; M3= MS + 0,1 mgL-1 NAA + 1 mgL-1 BAP ; and M4= MS + 0,1 mgL-1 NAA + 10 mgL-1 BAP. The second factor was explant type i.e.shoot tip (B1) and shoot base (B2). The results showed that there was no interaction between factorsfor all variable observed. Medium M0 (MS medium without any plat growth regulator) and explant ofB1 (shoot tip) gave the highest number of the percentage of explants producing shoots and the highestnumber of shoots per explant.

Abstract

In Planta Transformation of SoSPS1 Gene Through Agrobacterium tumefaciens in Balinese Grapevine (Vitis Vinifera L. var. Alphonso Lavalle). Vitis vinifera L. var. Alphonso Lavalle is one of the cultivar of grapevine cultivated in Bali. The sour taste of fruit is one of the problem of these cultivar that make it is less prefer by consumer. Research regarding improvement of fruit quality through genetic engineering has been investigated. The aims of the research were to find out the most proper methods of in planta transformation mediated with Agrobacterium tumefaciens applied on V. vinifera. The T-DNA harbored the SoSPS1 gene under the control of promoter of the CaMV from Cauliflower Mosaic Virus and contained the NPTII gene, a kanamycin resistant gene as a selectable marker for transformant selection. V. vinifera seedlings from stem cutting aged 40-50 days were ripped and then dipped with the bacterial suspension for 1 hour, 2 hours, 1 day, and 2 days. Visualization of V. vinifera samples after PCR showed that ripping and dipping for 2 hours method was positively integrated with NPTII gene. The dipping of plants wound for 2 hours treatment was effective for inserting gene of interest in ripping and dipping methods.

Abstract

Asymbiotic Germination of Dendrobium bicaudatum Orchid Seeds on Media with In Vitro Addition of Tomato Extract. Dendrobium bicaudatum is a natural orchid that needs to be preserved. Orchid seeds propagation naturally (symbiotic) has a low success rate. Orchid seeds do not have an endosperm. Asymbiotic germination with in vitro culture techniques can be an alternative solution for the propagation of orchid seeds. This study aimed to determine the effect of giving tomato extract on modified media on the asymbiotic germination of Dendrobium bicaudatum orchid seeds. This research used the Completely Randomized Design with five treatments of tomato extract concentration, namely 0%, 5%, 10%, 15%, 20%, with 4 replications. The variables observed in this study were the time and percentage of phase 1; time and percentage of phase 2; time and percentage of phase 3; time and percentage of phase 4; time and percentage of phase 5; and time and percentage of phase 6. The results showed that the tomato extract treatment had a significant or very significant effect on the germination of Dendrobium bicaudatum orchid seeds and the best treatment for the fastest and the highest percentage of germination from protocorm to First Leaf Primodium was the addition of tomato extract with a concentration of 15% or 20%, and a concentration of 0% tomato extract for the early stages of swelling of the embryo.

Abstract

The effect of inoculation of a combination of the arbuscular mycorrhizal fungus of Glomus and Acaulospora on micropropagated banana plantlets cultivar Raja from Bali during the acclimatization phase was investigated. The experiment was conducted at the Experimental Station of the Faculty of Agriculture, Udayana University, Denpasar, Bali, Indonesia, over the period of March to June 2022. The objective of this research was to evaluate the impact of various doses of mycorrhizae on the growth of tissue culture banana plantlets that had undergone acclimatization for a period of six weeks. A total of five different doses of mycorrhiza were applied to the acclimatized plantlets, namely M0=without mycorrhiza and M5=5g mycorrhiza.kg-1, M10=10 g mycorrhiza.kg-1, M15=15 g mycorrhiza.kg-1, M20=20 g mycorrhiza.kg-1. A significant effect was discovered at M15; however, compared to M20, M20 gave better results for root length and fresh weight of the above-ground part, indicating the best dose was 20 g mycorrhiza kg-1. This study concluded that mycorrhiza has a very significant role in supporting the growth of micro propagated banana plantlets at post-acclimatization. Variables observed, such as the average number of leaves, stem diameter, above-ground plant fresh and dry weight, and root fresh and dry weight, increased with the application of AMF (Glomus and Acaulospora).

Abstract

Determination of D. crumenatum mutant obtained from in vitro mutation breeding needs a long time due to its long-life cycle. SSR molecular markers can be used for early mutant detection. Specific SSR markers developed in D. crumenatum are not yet available. Alternative published SSR markers were developed from D. catenatum. The aims of this study are to screen the most informative SSR markers generated from D. catenatum tested in irradiated D. crumenatum population and to determine the gamma irradiation dose resulting the most mutants. Ten SSR markers were randomly selected and tested in 25 individuals of D. crumenatum plantlets irradiated with several doses (0, 5, 10, 15, and 20 Gy; 5 plantlets each dose). The result showed 7 of 10 primers were polymorphic and the other three were  monomorphic. All seven polymorphic primers can be used to identified intraspecific variation in the D. crumenatum mutant population. Markers dnsr28 and dnsr98 were the most informative, with the highest polymorphic information content (PIC) value of 0.5. Irradiation D. crumenatum protocorms using 10-15 gray doses were detected as the highest mutant percentage obtained up to 100% in the sample tested. This resulting marker information can be used to screen wider mutant population to decrease the non-mutant individuals in the population for maintenance and cost efficiency. The 10-15 Gy can be used as reference doses for gamma irradiation in 3 months old D. crumenatum protocorm materials.

Abstract

The large number of in vitro media circulating in the market is a challenge to find the most effective media for sachinoka strawberry meristem culture, because the need for in vitro media depends on genetic intelligence. The purpose of this study was to find the best media from the various tested media to reproduce the Sachinoka variety strawberry seedlings in a relatively short time using the meristem culture method. This study used a completely randomized design (CRD) with 6 media treatments and repeated 7 times, namely: M1 (WPM without BA), M2 (WPM + 4 ppm BA), M3 (NP without BA), M3 (NP + 4 ppm BA), M5 (BMM without BA), M6 (BMM + 4 ppm BA). The variables observed were the time of emergence of shoots, the number of shoots and the number of leaves. The results of this study indicate that there is no significant difference for the emergence of shoots variable. Treatment M1 produced the highest number of shoots (12.29) but not significantly different from M5 (10.14), both were significantly different from both treatments. The highest number of leaves is in M1 (27.14). It can be concluded that the best medium for the growth of sachinoka strawberry meristem culture is WPM media with the addition of 4 ppm BA, while the second-best medium is BMM media with the addition of 4 ppm BA.
