Abstract

TThe prevalence of dyslipidemia in Indonesia continues to increase. Anredera cordifolia and Curcuma xanthorrhiza are empirically used in the community to overcome dyslipidemia disorders. Flavonoids as the main compounds in A. cordifolia and curcumin in C. xanthorrhiza have potential as antioxidant activity. This study aims to determine the in vitro mechanism of A. cordifolia and C. xanthorrhiza to reduce cholesterol associated with antioxidant activity. The testing was carried out on a combination of ethanol extracts of A. cordifolia leaves with C. xanthorrhiza rhizomes (1:1) and their single extract. The antioxidant activity test was conducted using DPPH method by making serial dilution of samples and ascorbic acid with adding DPPH. The inhibitory activity of HMG CoA and lipase enzymes was carried out enzymatically using ELISA as well as simvastatin as the comparison. The results showed that the highest antioxidant activity is in C. xanthorrhiza extract samples IC50 16.21 ± 1.74 µg/mL, but was less potential compared to ascorbic acid IC50 7,63±0,85 µg/mL. The most potential inhibition of HMG CoA and lipase is in C. xanthorrhiza samples IC50 8.35 ± 0.02 and 22.35 ± 1.26 µg/mL. The combination of A. cordifolia and C. xanthorrhiza (1: 1) is better than A. cordifolia extract but lower than C. xanthorrhiza extract in antioxidant activity as well as the inhibition of HMG CoA reductase and lipase. The result showed a correlation that the higher antioxidant activity, the higher inhibitory of HMG CoA reductase and lipase respectively.

Abstract

The incidence of malaria in Indonesia is about two million cases annually. Dihydroartemisinin-piperaquine (DHP) is the first line therapy recommended for uncomplicated malaria treatment, whereas DHP is still fully imported. The generic DHP tablet formulation has the potential to become the first of DHP drug which is locally produced. This study is aimed to formulate generic DHP film coated tablets for antimalaria drug. Tablets were compressed with the combination of wet granulation for piperaquine phosphate (PQP) and direct compression method for DHA and coated with a moisture barier coating material. The parameters to evaluate the quality of DHP tablets are physical properties, assay, and dissolution test. DHA and PQP assay were performed by HPLC method. The dissolution testing was conducted by in house method using HCl 0.1 N medium. The result shows physical properties of filmcoated tablets meet the requirement, i.e. uniform weight, 7.0-8.5 kp hardness, 0.02% friability and 3 minute 22 seconds disintegration. The assay to determine DHA in tablet was 95.17% and PQP was 97.05%. The result of dissolution testing shows the content of DHA and PQP in the tablet were 113.51% and 96.55%, respesctively. The formulation which is developed meets the general requirement of API in tablet 90–110% and dissolution requirement >75%.

Abstract

Gambir extract contains a high content of catechin. Catechin has so many benefits, such as antioxidant, antimicrobial, and anti-atherosclerosis.Gambir are widely available in Indonesia, especially in West Sumatra, might be a sustainable source of catechin. Catechin is easily oxidized and hygroscopic. Film coating is used to protect catechin contained in a tablet from moisture and light. Study of film-coated tablet of catechin has not been conducted before. This study aims to formulate film-coated tablet of catechin from gambir and evaluate its physical properties. Catechin was percolated from gambir extract using ethyl astetat. The ethyl acetate fraction of gambir extract contains 92,48±0,28% catechin. Tablets were compressed with direct compression method and coated with a polymer coating material. The physical properties of core tablet and film-coated tablet were evaluated toward weight uniformity, hardness, friability, and disintegration time. Friability testing was conducted only on core tablet. The formulation produced tablets with uniform weight, 16,6 ±1,97 Kp hardness, and 4 minutes 15 seconds disintegration time. Weight uniformity and disintegration time of the tablet meet the requirements.

Abstract

Cinnamomum culilaban is one of the native Indonesian plants that has been used as medicinal plant. The local community on Seram Island uses the stem bark to treat toothaches. Eugenol, a chemical compound found in Cinnamomum culilaban, is used in dental practice and has antibacterial activity. Research on Cinnamomum culilaban is very limited, its activity against bacteria that cause oral infections has not been reported. This study aims to investigate the antibacterial activity of Cinnamomum culilaban bark extract against oral pathogens Porphyromonas gingivalis, Streptpcoccus mutans, and Enterococcus faecalis. The extraction was carried out using 96% ethanol with ultrasound-assisted extraction method. Disc diffusion assay was conducted to investigate the antibacterial activity. The concentration of extract used were 50%, 25%, and 12.5% (b/v) in DMSO. Eugenol (50% v/v in DMSO) was used as positive control. The phytochemicals screening was carried out to investigate the chemical compounds contained in Cinnamomum culilaban bark extract. Extraction of Cinnamomum culilaban bark using 96% ethanol with the ultrasound-assisted extraction method obtained a yield of 23.36±0.49%. The extract contains alkaloids, flavonoids, tannins, saponins, phenolics, and steroids/terpenoids. The ethanolic extract of Cinnamomum culilaban bark has inhibitory activity against Streptpcoccus mutans and Enterococcus faecalis. Cinnamomum culilaban bark has the potential as an antibacterial agent.
