Abstract

A computer application program for processing plasma/serum drug concentration data for bioequivalence test according to the Bioequivalence Study Guideline of The National Agency of Drug and Food Control of The Republic of Indonesia 2004, had been developed using Microsoft® Visual Basic 6.0. The program developed was used to process bioequivalent parameters using eight sets of plasma drug concentration data, and the results were compared to that of manual calculation using Microsoft® Excel 2003 program. Result showed that bioequivalence parameters calculated by this program were similar to those calculated by Microsoft®  Excel 2003.

Abstract

Ekstrak kunyit dan bawang putih telah diketahui mempunyai efek antidiabetik, tetapi mekanisme kerjanya belum diketahui. Penelitian ini mengamati efek tiga ekstrak rimpang kunyit (Curcuma longa L.) dan bawang putih (Allium sativum L.), yaitu ekstrak heksan, etil asetat, dan etanol terhadap kadar glukosa darah dengan tes toleransi glukosa. Selanjutnya, ekstrak yang paling efektif dan komponen aktifnya (kurkuminoid dan S-metil sistein) diuji dengan tes toleransi insulin. Empat puluh ekor tikus galur Wistar dibagi menjadi 8 kelompok, yaitu kelompok normal, kelompok yang hanya diberi emulsi tinggi lemak (kontrol), dan sisanya selain diberi emulsi tinggi lemak juga masing-masing diberi ekstrak kunyit dengan dosis 50 mg/kgBB, ekstrak bawang putih dengan dosis 50 mg/kgBB, kurkuminoid dengan dosis 25 mg/kgBB, S-metil sistein dengan dosis 25 mg/kgBB, kombinasi ekstrak kunyitbawang putih dengan dosis masing-masing 25 mg/kgBB, dan kombinasi kurkuminoid-S-metil sistein dengan dosis masing-masing 12,5 mg/kgBB selama 10 hari. Resistensi insulin dievaluasi dengan tes toleransi insulin.Penelitian dilakukan pada bulan Agustus–Oktober 2010 di Sekolah Ilmu dan Teknologi Hayati (SITH) InstitutTeknologi Bandung (ITB). Hasil penelitian ini menunjukkan bahwa KTTI (konstanta tes toleransi insulin) hewankelompok ekstrak bawang putih (7,2±0,84), kurkuminoid (7,14±0,74), dan kombinasi kurkuminoid-S-metil sistein(7,46±0,64) secara bermakna (p<0,05) lebih besar dibandingkan dengan kelompok kontrol positif (3,2±1,92).Simpulan, ekstrak bawang putih, kurkuminoid, dan kombinasi kurkuminoid-S-metil sistein meningkatkan sensitivitas insulin. However their mechanism of action remain unknown. In this study, we investigated the effect of three turmeric (Curcuma longa L.) and garlic extracts (Allium sativum L.), that are, hexane, ethyl acetate and ethanol extract on blood glucose levels with glucose tolerance test. Furthermore the most effective extracts and its active compound (curcuminoid and S-methyl cysteine) tested with insulin tolerance test. Forty Wistar rats were divided into 8 groups that was normal group, group that treated with a high fat emulsion (control group) and remaining groups were treated with a high fat emulsion and turmeric extract 50 mg/kgBW, garlic extract 50 mg/kgBW, curcuminoid 25 mg/kgBW, S-methyl cysteine 25 mg/kgBW, turmeric-garlic extract combination each 25 mg/kgBW and curcuminoid-S-methyl cysteine combination each 12,5 mg/kgBW for 10 days. Insulin resistance was evaluated by insulin tolerance test. This study conducted from August–October 2010 at Sekolah Ilmu dan Teknologi Hayati (SITH) Institut Teknologi Bandung (ITB). Results of this study showed that insulin tolerance test constanta (KITT) were bigger in animals that treated with garlic extract (7.2±0.84), curcuminoid (7.14±0.74) and combination of curcuminoid-S-methyl cysteine (7.46±0.64) compared with positive control group (3.2±1.92). In conclusions garlic extract, curcuminoid and combination of curcuminoid and S-methyl cysteine improve insulin sensitivity. Abstract

KELARUTAN DAN STABILITAS KIMIA KOMPLEKS DIDANOSIN DENGAN NIKOTINAMID ATAU L-ARGININ. Didanosin (DDI) adalah obat anti HIV yang bekerja dengan menghambat enzim reverse transkriptase yang bertanggung jawab dalam replikasi virus HIV. Ketersediaan hayati DDI pada pemberian secara oral sangat buruk akibat terhidrolisis dalam suasana asam. Penelitian ini bertujuan untuk mengetahui pengaruh pembentukan kompleks antara DDI dengan NKT atau ARG terhadap kelarutan dan stabilitas kimia DDI. Kompleks DDI-NKT dibuat dengan metode slurry menggunakan pelarut campuran etanol:air (7:3), sedangkan kompleks DDI-ARG dibuat dengan metode penguapan pelarut menggunakan pelarut yang sama. Karakterisasi kompleks DDI-NKT dan DDI-ARG meliputi, analisis dengan mikroskop polarisasi, difraksi sinar-X serbuk, uji kelarutan, dan stabilitas kimia pada larutan dapar pH 1,2; 4,5 dan 6,8 di suhu 37 °C. Karakterisasi dengan mikroskop polarisasi dan difraksi sinar-X serbuk menunjukkan terbentuknya kompleks DDI-NKT dan DDI-ARG. Kelarutan kompleks DDI-ARG dan DDI-NKT dalam air berturut-turut 1,86 dan 4,54 kali lipat dari kelarutan DDI murni. Hasil uji stabilitas kimia menunjukkan larutan kompleks DDI-NKT dan DDI-ARG mengalami degradasi lebih lambat daripada DDI murni pada pH 1,2; 4,5; dan 6,8 di suhu 37°C. Hubungan linier antara pH terhadap logaritma tetapan laju degradasi (log kobs) menunjukkan, bahwa degradasi kompleks DDI-NKT, DDI-ARG, dan DDI pada pH 1,2 hingga 6,8 dikatalisis oleh asam spesifik. Hasil penelitian dapat disimpulkan, bahwa pembentukan kompleks DDI-NKT dan DDI-ARG dapat meningkatkan kelarutan dan stabilitas kimia DDI.

Abstract

PENGARUH PENGGILINGAN TERHADAP PADATAN NIMODIPIN. Energi mekanik melalui penggilingan dapat menyebabkan transformasi polimorfik dan kerusakan kristal pada suatu padatan kristalin sehingga akan mempengaruhi sifat fisikokimianya. Tujuan penelitian ini adalah untuk mempelajari pengaruh penggilingan terhadap padatan nimodipin. Penggilingan dilakukan dengan alat Retsch RM100 mortar grinder selama 15 menit, 30 menit, 60 menit dan 120 menit. Karakterisasi padatan hasil penggilingan dilakukan menggunakan difraksi sinar-X serbuk, Differential Scanning Calorimeter (DSC), Scanning ElectronMicroscope (SEM) dan spektrometer Fourier Transform-Infrared (FT-IR). Difraktogramsinar-X serbuk padatan nimodipin hasil penggilingan menunjukkan puncak-puncak yang identik dengan nimodipin sebelum penggilingan.Termogram DSC nimodipin menunjukkan puncak endotermik pada suhu 124,2 oC. Titik lebur dan entalpi nimodipin akibat penggilinganmengalami sedikit pergeseran ke arah yang lebih rendah denganmeningkatnya waktu penggilingan. Fotomikrograf SEMmenunjukkan terjadinya amorfisasi dan agregasi partikel. Spektrum FT-IR tidak menunjukkan adanya pergeseran bilangan gelombang. Data-data tersebut menegaskan bahwa energi mekanik akibat proses penggilingan sampai 120 menit tidak menyebabkan transformasi polimorfik pada padatan nimodipin.

Diabetes mellitus is a metabolic disorder present in the top ten diseases that cause death. Diabetes treatment usually uses conventional drugs, such as gliclazide, which is the first choice in patients with type 2 diabetes mellitus who are intolerant to metformin. In addition to conventional medicine, herbal medicines are in great demand and have become a focus of research to obtain an alternative treatment for type 2 diabetes mellitus. One herb that has been widely used and studied is the bitter melon. This study aimed to determine the effects of bitter melon extract (BME) on the pharmacokinetics and pharmacodynamics of gliclazide when used simultaneously. The study was conducted on three groups of rats with alloxan-induced diabetes. Group 1 received a single dose of gliclazide (33 mg/kg BW, n = 3), group 2 received aqueous extracts of bitter melon fruit (341 mg/kg BW, n = 4), and group 3 received a combination of gliclazide and an aqueous extract of bitter melon (n = 4). The pharmacokinetic profile of gliclazide is affected by BME, where interactions occur during the absorption phase. The blood glucose levels were measured using a glucometer. Decreased blood glucose levels following administration of gliclazide, extract of bitter melon, and a combination of gliclazide and extract of bitter melon 6 hours after dosing were 74.73%, 82.977%, and 86.457%, respectively. This study demonstrated the interactions between gliclazide and BME in the absorption phase of gliclazide and its effect on blood glucose levels.
