Abstract

God mushroom is a functional food. It has pharmacological activity. This content has activity as an anticancer. The purpose of this study was to conduct a preliminary test to obtain an active ingredient capable of having anticancer activity on MCF-7 cells. The stages of the research are stratified extraction with several solvents. Extraction using maceration. The resulting extract was identified and tested for the anticancer activity of MCF-7 cells. The test uses the TLC method. The stationary phase with silica GF254, the mobile phase is n-hexane: ethyl acetate. Anticancer activity test using the MTT method. The results of the identification of each extract contained terpenoid secondary metabolites. Anticancer activity of MCF-7 cells with IC50 extract of n-hexane 15.0923 µg/ml, dichloromethane extract 17.4213 µg/ml, ethyl acetate extract 10.9132µg/ml with strong cytotoxicity criteria, while for ethanol extract IC50 675, 1236 µg/ml.

Abstract

ABM (Agaricus blazei Murill) is a basidiomycetes fungus. ABM is used by people for the treatment of diabetes, antihypertention, anticholesterol, anticancer, and immunostimulant. ABM contains terpene, steroids, agaritine, vitamin C, vitamin E, and betaglucane. In this research, ABM extract was tested as an anti-breast cancer in vitro using MCF-7 breast cancer cells. The extract was obtained from the multistage extraction process of several solvents in turn, the solvent used, among others, n-hexane, dichloromethane (DCM), chloroform, ethyl acetate, butanol, and water. The results of the research were the obtained IC50 value from n-hexane extract 247,17 μg /ml; extract DCM 227μg/ml ; chloroform extract 215,64 μg /ml ; extract of ethyl acetate 234,9 μg/ml ; butanol extract 500,78 μg/ml; while the water extract was inactive. Based on these results can be considered for further research to fractionate in order to know which class compounds have the potency as anticancer within the extracts.Key words : Agaricus blazei, multistage extraction, MCF-7 cells.

Abstract

Dynamics of compound 1,3-β-glucan in Agaricus Blazei Murill (ABM) powder and the effect of renal and hepatic histopathology performed on 24 male rats aged 2 months, were randomly conducted in three treatments there are control, ABM powder, and the pure compound 1.3-β-glucan for 28 days. Provision of treatment given in the morning and then 1, 3, 8, 12, and 24 hours after administration of treatment blood was collected, while for urine and feces were collected every 8 hours. Giving treatment was continued until day 28. The results showed that the levels of 1,3-β-glucan in pure compounds is higher than the Agaricus Blazei Murill (ABM) powder. The effects on the kidneys and liver histopathology showed that cells necrosis more on pure compounds from the mushroom powder gods, both the kidneys and liver. This study shows that the use of pure compounds to be more careful than Agaricus Blazei Murill (ABM) powder in determining the doses used.

Abstract

Dewa mushroom is a saprophytic mushroom that can function as medicine, namely antihypertensive, antibacterial, anticancer, immunostimulant, anticholesterol, antiviral, and antioxidant. The content of secondary metabolites of terpenoids, alkaloids, phenols have antibacterial activity. The extract was obtained by stepwise extraction with hexane, dichloromethane, and ethylacetate as solvents. Testing of antibacterial activity by diffusion method. The results obtained that hexane extract, dichloromethane extract, and ethylacetate extract had inhibitory power against Escherrichia coli and Staphylococcus aureus. There were significant differences in the three extracts against Escherrichia coli. Whereas in Staphylococcus aureus there was a significant difference between dichloromethane extract and ethylacetate extract, and for hexane extract to dichloromethane extract and ethyl acetate extract there was no significant difference (having the same inhibitory power)

Abstract

Latar Belakang: Kayu Bidara Laut (Strychnos ligustrina Blume) memiliki banyak efek farmakologis sebagai obat tradisional. Standarisasi simplisia kayu bidara laut diperlukan untuk penyediaan bahan baku sediaan obat tradisional.Tujuan untuk mengetahui simplisia kayu bidara laut memenuhi standar mutu fisik dan skrining sesuai Farmakope Herbal Indonesia edisi II. Penelitian ini termasuk jenis penelitian deskriptif. Metode: Metode penelitian secara deskriptih. Langkah-langkah yang dilakukan adalah: (1) determinasi tanaman, (2) Pembuatan simplisia kayu bidara laut, (3) pengujian makroskopis simplisia, (4) pengujian mikroskopis dan mikroskopis, (5) krining fitokimia, dan (6) pengujian mutu fisik. Hasil: Determinasi menunjukkan familia loganiaceae, marga Strychnos. Hasil makroskopis dan mikroskopis sesuai dengan FIH II. Kandungan metabolit skunder golongan senyawa alkaloid, flavonoid, saponin, terpenid, dan tannin. Mutu fisiknya berupa kadar air dan susut penegeringan 7,56%±0,02; kadr abu total 3,47±0,002; kadar abu tidak larut asam 0,05%±0,0001.

Abstract

Data Globocan tahun 2020, jumlah kasus baru kanker payudara mencapai 68.858 kasus (16,6%) dari total 396.914 kasus baru kanker di Indonesia. Sementara itu, untuk jumlah kematiannya mencapai lebih dari 22 ribu jiwa kasus. Usaha pencarian obat antikanker dari bahan alam yang mempunyai target molekuler yang spesifik, selektifitas yang tinggi, dan efek samping minimum, amat diperlukan dalam pengobatan penyakit kanker. Salah satu tanaman tingkat rendah yang dapat menghambat pertumbuhan kanker adalah jamur dewa (Agaricus blazei Murill/ABM). Tujuan penelitian ini adalah mengidentifikasi profil metabolit sekunder ABM yang diekstrak menggunakan pelarut etanol 96% dan uji aktivitas antikanker payudara pada sel MCF-7. Metode ekstraksi yang digunakan adalah maserasi, filtrat dikeringkan dengan freezdry. Penentuan profil metabolit sekunder menggunakan metode reaksi warna dan KLT. Uji aktivitas antikanker menggunakan metode uji MTT untuk menentukan IC50 . Hasil uji profile metabolit sekunder secara reaksi warna dan KLT bahwa ekstrak etanol 96% ABM mengandung terpenoid dan alkaloid. Pengujian aktivitas antikanker diakukan secara in-vitro menggunakan sel MCF-7 bahwa ekstrak etanol 96% ABM memiliki aktivias antikanker dengan IC50 89,8419µg/ml. Nilai IC50 tersebut termasuk strong citotoxicity

Abstract

The use of natural ingredients for cancer treatment is something that needs to be considered to obtain treatment according to the right molecular target cells with high selectivity with minimal side effects. Temu giring (Curcuma heyneana) has the highest antioxidant activity among the rhizome groups; the content of secondary metabolites is polyphenols and flavonoids. There was a correlation between antioxidant activity and cancer activity. The aim of this study was to examine the profile and anticancer activity of MCF-7 cells treated with a 96% ethanol extract of temu giring. Extraction using the maceration method. The profile of temu giring extract uses chemical reactions and thin layer chromatography. Anticancer testing using the Microculture Tetrazolium Technique assay method. Testing using MCF-7 cells with extract concentrations of 400µL, 200µL, 100µL, 50µL, and 25µL. Extract yield: 5.17%. Secondary metabolites in turmeric extract are flavonoids, terpenoids, polyphenols, and anthraquinones. The metabolites have the ability to inhibit MCF-7 cells. The anticancer potential of MCF-7 cells with IC50 60.38±9.03µL (strong cytotoxicity).

Abstract

Soap is a cleaning product produced from the reaction between oil or fat with potassium hydroxide or sodium hydroxide base. Based on its consistency, soap is divided into two types, namely liquid soap and solid soap. Solid soap is more often used by the public. Utilization of natural ingredients as an innovation for soap is still rarely done. The content of coffee is able to prevent premature aging of the skin, function to smooth the skin, and neutralize irritated skin. This study aims to determine the physical quality and pH of solid soap mixed with coffee extract. Production of coffee extract using the maceration method with 70% ethanol solvent. This type of research is descriptive, with the aim of knowing the physical quality and pH of solid body soap by adding coffee extract. Based on the results of the research conducted, it can be concluded that the results of the evaluation of the physical quality of the coffee extract solid soap preparations fulfilled the physical quality test requirements, which included organoleptic tests, water content tests, foam stability tests, and pH.

Abstract

Tyrosinase is a critical enzyme in melanin synthesis, where its overactivity can result in hyperpigmentation disorders. Despite the availability of synthetic tyrosinase inhibitors, concerns about safety have prompted the search for natural alternatives. The objective of this study is to evaluate the potential of Kepok banana (Musa acuminata x balbisiana) corm extract as a natural tyrosinase inhibitor, using in vitro and in silico approaches. The extract was prepared by maceration with etanol 96%, followed by sonication and lyophilization. LC-MS analysis was performed on a Shimadzu LCMS 8040 using a Shim Pack FC ODS column with 90% methanol as the mobile phase under isocratic conditions. Samples were ionized via ESI in positive mode and scanned over m/z 10–1000. Peaks were identified based on their m/z values and MS/MS fragmentation patterns. In vitro and in silico analyses were applied, respectively, to study the biological process of kepok banana corm extract as a natural tyrosinase inhibitor. The result of the kepok banana corm extraction process was 12.5% dry powder with bioactive components identified via LC-MS, including flavonoids, phenolics, and terpenoids. The kepok banana corm ethanol extract exhibited strong tyrosinase inhibition with an IC₅₀ of 91.074 µg/mL, falling below the 100 µg/mL threshold for strong inhibitors and showing a similar inhibition pattern to the standard kojic acid (IC₅₀ = 10.87 µg/mL). Molecular docking revealed that major compounds, particularly flavonoids, exhibited high binding affinity with the tyrosinase enzyme through hydrogen bonding and hydrophobic interactions, similar to kojic acid, a known inhibitor. These findings suggest that kepok banana corm extract may serve as a promising natural tyrosinase inhibitor, with potential applications in hyperpigmentation treatment and cosmetic formulations.
