Abstract

Enzim kitosanase merupakan enzim yang mengkatalisis reaksi endohidrolisis ikatan (1,4) β- glukosida pada kitosan menjadi serangkaian kitooligosakarida atau oligomer kitosan. Diantarasemua organisme penghasil kitosanase, bakteri mendapat perhatian khusus karena bakteri mampu memproduksi secara cepat kandungan biomassa sehingga senyawa bioaktif bisadiproduksi lebih mudah, cepat dan banyak dalam skala bioteknologi Kitosanase dari mikroba memberikan hasil yang baik dalam memproduksi kitooligosakarida, namun sangat mahal untuk digunakan di industri dalam skala besar. Penelitian ini bertujuan untuk memproduksi dan karakterisasi ekstrak kasar enzim kitosanase dari Klebsiella sp. Enzim kitosanase diproduksi pada medium fermentasi yang mengandung 0,5% koloidal kitosan selama 60 jam dengan aktivitas 0,309 U/mL (5,235 U/mg), bekerja optimum pada suhu 40 ºC dan pH 8, kecepatan maksimal enzim pada konsentrasi substrat (soluble kitosan) 1 %, diaktifkan oleh Co2+; Ca2+ dan Ni2+ serta dihambat oleh Zn2+; Mg2+ dan Cu2+.Kata kunci: Kitosanase, Klebsiella, kitooligosakarida Kitosanase enzyme is an enzyme to catalyze the bonding endohidrolisis (1.4) β- glucoside on chitosan into a series chitosan oligomers or kitooligosakarida. Among all organismschitosanase, bacteria have a particular concern because the bacteria are able to produce quickly the content of the biomass so that the bioactive compounds could be produced more easily ,faster and more in scale biotechnology. Chitosanase of microbes give good results in producing kitooligosakarida, but very expensive for use in large-scale industry. This study aims to manufacture and characterization of enzymes chitosanase crude extract of Klebsiella sp. Kitosanase enzyme produced in the fermentation medium containing 0.5 % colloidal chitosan for60 hours with the activity of 0.309 U / mL ( 5,235 U / mg ) , work optimally at 40 ° C and pH 8, the maximum speed on the enzyme substrate concentration (soluble chitosan ) 1 %, powered by Co2+ ; Ca2+ and Ni2+ and inhibited by Zn2+ ; Mg2+ and Cu2+ .Keyword : Chitosanase, Klebsiella, Chitooligosaccaride

Abstract

Moringa plants are one of the plants that contain lots of nutrients both minerals and vitamins. This study aims to determine the content of Vitamin C extract of Moringa oleifera Lam extract with different heights in Baubau. Moringa leaves were extracted by maceration using water and Sodium Oxalate (Na2C2O4) and vitamin C levels were analyzed using the UV-Vis Spectrophotometer Method. The results showed that the vitamin C content of Moringa leaves for a height of 20 masl (KP 20) in the coastal area was 0.59%, 50 masl (KM 50) 0.53%, while for an altitude of 296 masl (LBW) was 0.83% . From the data obtained shows that the highest levels of vitamin C are Moringa leaves located at a height of 296 mdl (LBW) compared to areas with low altitude in this case Moringa that grows on the coast (KP 20) and medium height (KM 50). From the results of this study it can be concluded that the vitamin C content of Moringa leaf extract which grows in the low area is 296.40 ppm, medium height 267,12 ppm and for the altitude region is 413.71 ppm.

Abstract

Chitosan oligomers or chitooligomer is a mixture of oligomers of D-glucosamine which are formed through a process of depolymerization of chitosan with cuting its β-glycosidic bonding. Chitosan oligomer has received much attention in various fields because of the specific biological activity 10 times greater than chitosan. Chitosan oligomer having biological activity as antibacterial, antiviral, antioxidant, anti-tumor and anti-cancer. Before it is applied to humans, the bioactive compounds to be used as pharmaceutical products must pass a preliminary test using test animals. BSLT method is often used to praskrining against bioactive compounds. This method uses shrimp larvae are widely used in searches that are toxic bioactive compounds from natural materials. This study aims to determine the toxicity of chitosan oligomers enzymatic (crude) as a preliminary test before being used as a pharmaceutical product either as anti-cancer or anti microbial with BSLT method. The results showed chitosan oligomer (crude) hydrolysis of leather tiger shrimp chitosan (DD 60%) with chitosanase enzyme for each time of incubation has strong toxicity properties with a value of 36.90 ppm LC50 (1 hour incubation); 47.43 ppm (incubation 2 hours) and 104.86 ppm (incubation 3 hours).
