Abstract

Background: Several epidemiological data’s reported significant correlation between fructose consumption and diabetes mellitus type 2 by inducing insulin resistance. Leptin resistance induced by fructose was proposed as one novel mechanism that induce insulin resistance but the exact mechanism remains unclear.  We hypothesize that fructose diminish the type b leptin receptors in hypothalamus and intestine.Aim: This study was aimed to elucidate fructose effect on the expression of leptin receptor type b in hypothalamus and intestine of Rattus novergicus.Method: twenty eight rats were used and divided into 4 groups: Group 1 was control, group 2 was given fructose 10%, group 3 was given fructose 30% and group 4 was given fructose 55% for 2 months. At the end of treatment, the animal were sacrificed and then the hypothalamus and intestine were collected. The expression of type b leptin receptor were measured by immunohistochemistry technique with primary antibody from Bioss antibodies type Leptin receptor polyclonal antibody bs-0109R using Staining kit Skytec Laboratories and DAB chromogen. A positive expression can be seen as a brown colour in cell cytoplasm and counted in 100 cell. The expression then analysed using SPSS 18 with anova one way tes (p0,05) followed by post hoc test after the data showed normality and homogeneity using Saphiro wilk and Levene test (p0,05).Result: There was significant differences in type b leptin receptors found in hypothalamus between each group (p0.05). The significant differences also could be seen in the expression between control and group fructose 30 and 50% in intestinal cell (p0.05).Conclusion: the consumption of Fructose 55% for 2 months attenuates the expression of type b leptin receptors in hypothalamus and intestine of Rat novergicus.

Abstract

Kanker  serviks  merupakan  penyebab  utama  kedua  kematian  kanker  pada wanita  di seluruh  dunia, menyebabkan  240.000 kematian  setiap  tahunnya.  Ekstrak  daun  Annona  muricata  berpotensi  baik  sebagai  obat  anti  kanker .  T ujuan  dari  penelitian ini  adalah  untuk  mengetahui  pengaruh  ekstrak  etanol  daun  sirsak  (Annona  muricata)  dalam  menghambat  dan menginduksi  aktivasi apoptosis yang dimediasi oleh stabilisasi p53 pada pertumbuhan kanker serviks.  Sebuah percobaan in vitro dilakukan dengan menggunakan kultur sel HeLa. Penghambatan proliferasi diukur dengan MTT assay. Apoptosis dideteksi  dengan  menggunakan  flowcytometry  Annexin  V  Biotin  Kit  Apoptosis  Detection.  Konfirmasi  dan  perhitungan ekspresi  p53  ditentukan  oleh  imunocytochemistry.  Ekstrak  etanol  dari  daun  Annona muricata  secara  efektif  menghambat proliferasi sel HeLa dengan dosis tertentu pada paparan 48 jam. Nilai  IC50 ekstrak etanol daun Annona muricata adalah 111,75  ug/ml.  200  mg/ml  ekstrak  menginduksi  apoptosis  sel  HeLa  tergantung  dari  lama  pemaparan.  Ekstrak  ini  juga meningkatkan  ekspresi  p53  sel  HeLa  tergantung  dosis  pemberian  (25,  50,  100,  200  μg/ml)  dan  lama  pemberian  (24  dan  48 jam).  Dapat  disimpulkan  bahwa  ekstrak  etanol  daun  sirsak  menghambat  pertumbuhan  sel  HeLa  dan  menginduksi apoptosis.  Mekanisme  ekstrak  daun  sirsak  menghambat  proliferasi  dan  apoptosis  inducing  berhubungan  dengan stabilisasi  dan  aktivasi  p53.Kata  Kunci:  Daun  Annona  muricata,  apoptosis,  ekstrak  etanol,  proliferasi,  ekspresi  p53

Abstract

Background: The Faculty of Medicine and Health Sciences of the State Islamic University (FKIK UIN) Malang needs to build recognition in realizing its superior curriculum, namely hajj health through hajj pilgrimage interprofessional education (HIPE) course. On the other hand, there is an opportunity from the Batu City Government and Ministry of Religion to collaborate in fostering the waitingperiod of Prospective Hajj Pilgrims (CJH) to achieve istitha'ah.Objective: This study aimed to compile and evaluate the learning instructional design of the Hajj Pilgrimage Interprofessional Education (HIPE) program as a manifestation of the superior curriculum of FKIK UIN Malang which is part of the prospective hajj pilgrimage waiting period program of the Batu city government.Research Method: This research was conducted from June to December 2023. The participants of the study included the executive board of faculty incluring dean and vice dean, head of medical bachelor and bachelor of pharmacy educational program, head of the ministry of religion, the hajj and umrah organization division, head program director of the health service district and from 5 primary health center, along with hajj pilgrimage candidate of Batu city in 2023. The data used was primary data from the program satisfaction survey questionnaire and secondary data in the form of formal documents.Results: The learning instructional design has been approved by the faculty leader, head of medical and pharmacy bachelor program, head of ministry of religion, and head of Batu city health office in Focus Group Discussion meeting with the topic of Hajj Health Superior Curriculum marked by the issuance of the MoU, the organizational structure of the faculty unit called Unit of Medical and Health Profession Education (UMHPE), and the Dean's Decree for the unit in charge. The organizational structure involved all parties including the ministry of religion of Batu city, the health officer and hajj program director in 5 primary health centers in the Batu city, and the Faculty with its study program. Longitudinal courses in 2 bachelor program was arranged with a spiral curriculum entitled HIPE 1, 2, 3. HIPE 3 as the final course applied the Project Based Learning (PJBL) approach providing intervention program to achieve istitha'ah which is implemented through hajj pilgrimage candidate serial home visits. The quality cycle has been applied in the entire series of learning programs. The results of the satisfaction survey from hajj pilgrimage candidate and external stakeholders showed a positive response to the HIPE program and the objectives of HIPE itself.Conclusion: The HIPE learning prototype as a superior curriculum of FKIK UIN Malang to foster hajj pilgrimage candidate during the waiting period in Batu City already facilitated a mutual collaboration between FKIK UIN Malang, the Health Office and the Ministry of Religion of Batu City.

Abstract

Background: Poor blood pressure control of hypertensive patients is caused partially by bad lifestyle management, which increases the risk of a cardiovascular event by stimulating the defective endothelial function. Objective: This study aimed to investigate the effect of night sleep duration on systolic blood pressure and Nitric Oxide concentration in hypertensive patients. Methods: 60 hypertensive patients age 40 years based on the medical record participated in this study. The sleep duration and sleep quality were performed using the PSQI questionnaire. The 40 participants thus selected randomly for the NO concentration assay using NO Salivary Elisa Kit ab65238. Results: 53 participants experienced night sleep duration ≤ 6 hours, and 44 (73.7%) participants had poor sleep quality. Night sleep duration was associated significantly with hypertensive patients' systolic blood pressure (p = 0,000 0.05; Odds ratio=0.008). Conclusion: The longer duration of sleep at night among hypertension patients, the lower systolic pressure were found. There was no correlation between night sleep duration on NO concentration (p 0.05). These findings provide evidence that night sleep duration is associated strongly with the increased systolic blood pressure but not directly caused by the alteration of endothelial function.Keywords: sleep duration. systolic blood pressure, hypertension, endothelial function, Nitric Oxide

Abstract

Introduction Modern life style and high burden of academic tasks contribute to the upsurge of stress, poor sleep quality, lack of sufficient physical activity in medical student society nowadays. The unhealthy life style act as one cardiovascular risk factor which can be predicted using Cardiorespiratory Fitness (CRF) examination. Objectives to identify the association between stress, sleep quality, physical activity with CRF level among medical students. Design The cross-sectional data was collected between September until November 2021. A total of 38 male and 73 female students of Faculty of Medicine and Health Sciences UIN Maulana Malik Ibrahim Malang were participated in this study. Stress level was assessed with DASS-21, sleep quality with PSQI, physical activity with GPAQ questionnaire.  The level of CRF was measured using the Harvard Step Test. Statistical analysis for bivariate correlation ordinal scale was tested using Spearman Rank. Results 83 participants (74,8%) had low CRF. Meanwhile, 76 participants (68,5%) experienced no stress. In addition, 75 participants (67,6%) had poor sleep quality. A total number of 79 participants (71%) had low physical activity. There was no significant correlation between stress level, sleep quality, physical activity with CRF ( p=0,962; 0,772; 0,114, respectively). The participants who had low physical activity and low CRF was found in 56 participants (50,45%). Conclusions There was no association between stress level, sleep quality, physical activity with CRF among medical students. Keywords: Cardiorespiratory fitness; Stress; Sleep quality; Physical activity.
