Abstract

Isolation and identification of active toxic compounds from nyamplung skin bean (Calophyllum inophyllumL). has been carried out. As much as 12,43 g of concentrated methanol extract was resulted from 1.0 kg dry powdernyamplung skin bean that was macerated using methanol. This extract was dissolved into methanol-water (7:3) andthen partitioned with n-hexane and chloroform respectively. The result of toxicity test to both extract showed thatchloroform extract was the most active toxic with LC50 = 239,88 ppmSeparation of chloroform extract using preparative thin layer chromatography (TLC) resulted in F5 fractionsthat was white yellowish as much as 52 mg and LC50 = 151,36 ppm. Infra red and UV-vis spectroscopy wereemployed in order to identify the F5 fraction. The result of identification showed that isolate was aldehidetriterpenoide compounds which had functional group like –OH (3429 cm-1), -CH aldehyde (2922,9 and 2852,9 cm-1),-CºC- (2360 cm-1), -C=O (1742,7 cm-1), -C=C- alkene(1637,0 cm-1), -CH3, -CH2 (1542,9, 1480,7, 1401,9 cm-1), -CO-(1090 cm-1), It showed absorbed at broad peaks at l(nm) (CHCl3): 254, 282, and 342,7.

Abstract

Gayam (Inocarpus fagiferus Fosb) is a tropical plant whose stem bark has been used as traditional medicine. This research aims to identify the flavonoides isolate from stem bark of gayam and identify the class of flavonoids compound as antioxidant. Extraction of 1000 g stem bark powder with 8500 mL of 96 % ethanol to yield 30 g concentrated ethanol extract. Partition of 15 g extract ethanol-water (7:3) with chloroform and n-buthanol to yield 1,5 g concentrated chloroform extract, 5,87 g concentrated n-buthanol extract and 5,63 g concentrated water extract. The flavonoids test using Wilstatter and Bate-Smith Matcalf reagent showed that n-buthanol and water extract contain flavonoids compounds. Antioxidant activity of flavonoids tested with DPPH method and lipid peroxide ammonium thiocyanate method showed that n-buthanol extract had antioxidant activity with IC50 of 24 ppm and n-buthanol extract has ability to inhibit the formation of lipid peroxide with inhibition value of 63,04 %. Separation of n-buthanol extract using thin layer chromatography preparative with chloroform as mobile phase and silica gel GF254 as stationary phase obtained four fractions. Flavonoids test showed that fraction F3 contained flavonoids compounds. FTIR analysis indicated that isolate had functional groups such as  -OH , C=O, C-O alcohol, C=C aromatic, C-H aromatic and C-H aliphatic. UV-vis spectra showed 2 peaks at ? 310 nm  and  261 nm which indicated flavonoids group of  isolfavone with hydroxyl groups at C-5 and  C-7. Antioxidant activity test with DPPH method showed that the isolate had antioxidant activity with IC50 of 1 ppm.

Abstract

Oxidative stress is an imbalance condition between free radicals in the body with the anti-oxidant. Superoxide dismutase (SOD) is one of the endogenous anti-oxidant that can neutralize the excess of Reactive Oxygen Species (ROS). The excess of ROS that caused by stres oksidative can also be neutralized with the help of exogenous anti-oxidants. A soursop plant (Annona muricata L.) has anti-oxidant activity, especially in the leaves. This study aims to determine the activity of the leaves of the soursop (Annona muricata L.) in inhibiting the oxidative stress that conducted on the object hypercholesterolemic rats wistar through increasing SOD activity using variations of the extract dose of 50 mg/kg BW; 100 mg/kg BW and 150 mg/kg BW. In this study the dose gives the best result in inhibiting the oxidative stress through an increase in SOD activity was dose of 150 mg/kg BW with percent inhibition of SOD at (81,42% ± 3.91) where the percent inhibition of SOD is directly proportional to the activity of SOD

Abstract

Isolation and identification one of cytotoxic fraction have been conducted from the leaves of Andong(Cordyline terminalis Kunth). The fraction having LC50 values of 41,64 ppm contains saponin. Major isolate (7.5 mgwhite amorphous solid) was obtained after a series of chromatographic separations (gravity column chromatography,and preparative high performance liquid chromatography). Identification of the isolate using mass spectrometry withpositive electrospray showed MW of 866 as calculated from the ion peaks m/z 889[M + Na]+, and 867[M + H]+.The ion peaks at m/z 721[(M + H) - 146]+, 703[(M + H) - 164]+, 575[(M + H) - 292]+, 557[(M + H) - 310]+ , 429[(M+ H) - 438]+, and 411 [(M + H) - 456]+ of its fragments indicate the presence of three sugars (two terminal sugarsand one central sugar) from methylpentose moiety with MW 164 eash linked to an agyicone of MW of 428. Protonmagnetic resonance spectrum of the isolate in pyridine-d5 showed characteristic proton signals for three steroidmethyls (two angular methyls and one secondary methyl) at d 1.37 (s), 0.84 (s) and 1.02 (d, J = 6.6 Hz); anexomethylene group at d 4.79 ppm and 4.71 (each br s); an ethylene group at d 5.52 ppm (br d, J = 5.4 Hz); signalsof the protons linked to C26 at d 4.01 and 4.44 ppm (each d, J = 12.0 Hz), and three anomeric protons at d 6.43 ppm(br s); 5.56 ppm (br s) and 4.57 ppm (d, J = 7.0 Hz). From the above data it can be indicated that the isolate resultedis spirostan steroidal saponin.

Abstract

These researchs aim to know of contain total flavonoid, total phenol and antioxidant activity in vitro of gayam stem bark of each extract (Inocarpus fagiferus Fosb). The result of extraction 1 kg stem bark of gayam with 8500 mL ethanol (5 x 1700 mL) to yield 30 g of concentrated extract its brown red. The partition of 15 g concentrated ethanol extract with chloroform, n-buthanol, and water to yields of 1.5g yellowish concentrated chloroform extract,  5.87 g brown red concentrated n-buthanol extract and 5.63 g concentrated water extract respectively. The result of phytochemical test of fourth extract shown that the ethanol, n-buthanol, and water extracts qualitatively contain of flavonoid and phenol compounds, while the chloroform extract only to contain phenol compound with much total flavonoid in ethanol extract 0.14 % and less at n-buthanol extract is 0.09 %. The result of antioxidant activity with DPPH method shown that water and  n-buthanol extract of gayam stem bark have value IC50 was 20 ppm smaller than  IC50 of vitamin E was 25 ppm its mean the water and  n-buthanol extract of gayam stem bark have the highest potent scavenger of free radical. The result of antioxidant activity with lipid peroxide-ammonium thiocyanate method shown the extract n-buthanol extract have the highest potent  inhibition of formed lipid peroxide (63.04%), it is higher than antioxidant activity of vitamin E (50.39%).

Abstract

Isolation and identification of active antifeedant compounds from sirsak seed (Anonna muricata Linn) has been carried out. As much as 13 g of concentrated methanol extract was resulted from 1.9 kg dried sirsak seed that was macerated using methanol. Further, this extract was partitioned with n-hexane to obtained 2 concentrated extracts in n-hexane and methanol. The result of antifeedant test of both extracts showed that n-hexane extract was more active than methanol extract.Separations of n-hexane extract using column chromatography (stationary phase: silica gel 60 and mobile phase: ethylacetate-chloroform 5:2) resulted in three fractions with FA fraction as the most active antifeedant (84,6% at 1600 ppm concentration).The identification using phytochemical test and spectrophotometer analysis showed that the isolate (FA. fraction) was triterpenoid aldehyde compound which absorbed at 227.6 nm, 275.1 nm and it had functional groups such as C-H stretching vinyl (3008.7 cm-1), two bands C-H streching aliphatic (2923.9 and 2854.5 cm-1) showing aldehyde group, and carbonyl group C=O (1743.5-1712,7 cm-1).

Abstract

Isolation and larvacide activity of essential oil from Piper betle Linn were studied. Approximately 13,5 mL of essential oil was obtained from 10,0 kg fresh leaves of Piper betle Linn. The oil is pale yellow with density of 0,7148 g/mL. Phytochemical test indicates that the content of the oil are terpenoid and phenol compounds with toxicity Aedes aegypty larvae 0f LC50= 309,03 ppm. The essential oil was toxic towards Aedes aegypty larvae because it contains phenol compounds, i.e. Eugenol, cavichol.GC-MS analysis reveals that there are 9 compound, i.e. 4-methyl(1-methylnethyl)-3-cyclohexen-1-ol, 1-methoxy-4(1-propenyl) benzene, 4-(2-propenyl)phenol/cavicol, 4-alylphenylacetate, Eugenol, Karyofilen, 3-alyl-6-methoxyphenylacetate, 4-alyl-1,2-diacetoxybenzene and decahydro-4a-methyl-1-methylene-7(1-methylethenyl) naphthalene.Based on the peaks intensity the oil was dominated by 4 compounds i.e. 4-allyl phenyl acetate, 2 methoxy-4-(2 prophenyl) phenol/eugenol, 3-allyl-6-methoxy phenyl acetate, 4-(2-prophenyl)-phenol / cavicol. These four compounds probably act as the active compounds to kill Aedes aegypty larvae, as they are acetate and phenol compounds.

Abstract

Protium is the main genusl in the family of Burseraceae which has been used traditionally as medicine, cosmetic as well as insecticide. This research aimed to isolate and identify antifeedant active compounds from leaf of Protium javanicum Burm, F which is locally known as tenggulun. Epilachna sparsa (E.sparsa) larvae were used as bioindicator for antifeedant activity assay. The assay was performed both on crude extract, fractions, and isolate. Dried leaf powder of tenggulun (1000 g) was extracted with methanol to yield 20.89 g dark green extract. This extract showed 71.61% antifeedant activity at 0.1% (b/v). This active extract was then fractionated into n-hexane, chloroform and water fractions. The chloroform fraction was the most active which give 89.62% activity at 0.1% (b/v) compared to n-hexane (36.83%), and water fractions (1.96 %). Purification of chloroform fraction was done using silica gel column chromatography with n-hexane : chloroform (3:4) as eluent and three groups of fractions (isolates) were obtained. The most active isolate which showed 70.53% antifeedant activity at 10 ppm was found relatively pure, therefore it was then identified.  Based on phytochemical reaction, analysis of infrared spectra and ultraviolet-visible spectra, the antifeedant active isolate was identified as triterpenoid class of compounds which have functional group such as OH, CH3, CH2, C=O, C=C and ?max at 245 and  416  nm.

Abstract

Sembukan adalah tumbuhan liar yang batang dan daunnya telah dimanfaatkan sebagai obat tradisional karena terdapat metabolit sekunder seperti flavonoid, alkaloid, minyak atsiri, dan steroid. Penelitian ini bertujuan untuk mengidentifikasi golongan senyawa flavonoid serta menguji aktivitas antioksidannya. Hasil maserasi 1,2 kg daun sembukan kering menghasilkan ekstrak kental etil asetat 0,34 g, n-butanol 3,12 g, dan air 26,85 g yang positif flavonoid. Pemisahan 3,12 g ekstrak n-butanol dengan kromatografi lapis tipis (KLT) preparatif dengan fase gerak n-butanol : asam asetat glasial : akuades (BAA) menghasilkan 5 fraksi dan 2 fraksi diantaranya yaitu fraksi F2 dan F4 positif flavonoid. Hasil analisis Fraksi F2 dengan FTIR menunjukkan bahwa isolat memiliki gugus fungsi C-H aromatik dan C=C aromatik, OH, C=O, CH alifatik, C-O alkohol, dan C-O eter, sedangkan analisis dengan spektrofotometer UV-Vis menunjukkan isolat menyerap pada panjang gelombang 315,60 nm (bahu) dan 283,80 nm sehingga diduga isolat adalah senyawa flavonoid golongan flavanon dan mempunyai gugus hidroksi pada atom C-3, C-3’, dan C4’. Uji aktivitas antioksidan menunjukkan bahwa isolat dapat menangkap radikal bebas 1,1’-diphenyl-1-picrylhydrazyl (DPPH) sebesar 21,59 % pada konsentrasi 50 ppm.

Abstract

Identification  of active toxic compounds from chloroform extract Nyamplung skin bean (Calophyllum inophyllum L) have been conducted. As much as 2,85 g n-hexane extract, 7,42 chloroform extract, and 4,71 water extract were resulted from 1 kg Nyamplung skin bean. Separation of  2 g  chloroform extract  using column chromatography (stationary phase: silica gel 60 and mobile phase chloroform-methanol 9,5:0,5) resulted  in four fraction with yellowish fraction (0,473 g) as the most toxic fraction (LC50 8,241 ppm). Identification using GC-MS showed that the isolate was hexadecanoic acid (21,02%), 8-octadecenoic acid (59,55%), and oktadecanoic acid(19,43%).
