Abstract

Background: Unlike iron ruthenium is not an essential element for life. However, the behavior of ruthenium compounds in biological systems and, in particular their use as antitumour agents has attracted much attention recently. This study aims to determine antitumour properties against human tumour cell line HeLa of pentamethylcarboxylate ruthenocene. Methods: This is an in vitro study by applying an experimental within post only control group design. Cisplatin, a clinical used medicine was applied for control. The human tumour cell line Hela was used for the test. The cells were cultured at 370C in 5% CO2/air in Roswell Park Memorial Institute Medium 1640 and seeded overnight in 96 well microtitre plates. The penthamethylcarboxylate ruthenocene was dissolved in 1,2 dimethoxy ethane and diluted with culture media. The plates were assays by measuring optical density in the range of 490-655 nm. The D37 values were then calculated. Results: The pentamethylcarboxylate ruthenocene compound tested was found to be more cytotoxic than cisplatin (with D37 = 422 nM compare to D37 = 705 nM for cisplatin). Conclusions: The compound tested, pentamethylcarboxylate ruthenocene was found more potent as an antitumor compare to cisplatin a clinical used antitumor for curing testicular carcinoma.

Abstract

Background: This research aims to find a cure for anti-inflammation, based on the utilization of red piper crocatum. The research was started with descriptive study to explore active components of red piper crocatum leaf and followed by experimental study to investigate red piper crocatum activity of the leaf extract in anti-inflammation induced Wistar rat. In this research observed three dominant components: caryophyllene bicyclo [5.2.0] none, 2 methylene-4,8,8-trimethyl-4-vinyl; phytol; 5,9-propano-5H- benzocycloheptene, 6,7,8,9-tetrahydro-7,11-bis(methylene); 4,4-ethynedioxy-2-hexadecen-15-15 olide 1,4,9-trioxaspiro [4,15] eic os-6-en-8-one, 10 methyl; 1H-1,2,4-triazole-5(H)-thione,4-allyl-3-(3-furyl); Benzofuran,2,3-dihydro-2-methyl-7-phenyl which are possibly active to inhibit anti-inflammation to atherosclerosis. Bad eating habits also can cause various health problems, such as obesity, dyslipidemia, inflammation to atherosclerosis. This study was conducted to investigate of red piper crocatum leaves extract as an anti-inflammation through decrease of biochemistry markers TNF-? and IL-6 levels. Method: This is a true experimental with randomized pre-test and post-test control group design, using 50 Wistar rats that are divided into 5 groups: control group using 0 mg/kg B red piper crocatum leaves extract, treatment group 1 using 50 mg/kg BW red piper crocatum leaves extract, treatment group 2 using 100 mg/kg BW red piper crocatum leaves extract, treatment group 3 using 150 mg/kg BW red piper crocatum leaves extract, and treatment group 4 200mg/kg BW red piper crocatum leaves extract. Results: It was observed that intake of 150 mg/B red piper crocatum leaves extract results in the highest significance decrease of 45.63% of TNF-? levels from (28.62 ± 1.25 to 15.56 ± 7.20 ?g/mL) and a significance decrease of 15.42% of IL-6 level from (134.64 ± 1.98 to 113.87 ± 4.30 ?g/mL). Conclusion: It can be concluded that intake of red piper crocatum leaves extract acts as antiinflammation for Wistar rats with atherosclerosis through decrease of TNF-? and IL-6 levels. Further research is required to determine whether the application of red piper crocatum leaves extract on human will result in similar effects of anti-inflammation.

Abstract

High cholesterol diet leads to increase of plasma cholesterol and subsequently will end up with hypercholesterolemia. Ignorance of healthy food and lacks of activity are the trend of modern life style. These conditions exacerbated the incidence of hypercholesterolemia in either developed or developing countries. This research aims to determine potential of anredera cordifolia leafs extracts as an anti-hypercholesterolemia by lowering of malondialdehyde (MDA) and 8-hydroxy-diguanosine (8-OHdG) blood levels in hypercholesterolemia Wistar Rat. This is an experimental study applied post only control group design. A number of 15 male wistar rats were used in this study. Rats were randomly divided into 3 groups, i.e. negative control group (C1), positive control group (C2) and anredera cordifolia leaf extract at dose of 100 mg/kg BW group (T). MDA was determined based on malondialdehyde assay (NWLSSTM).  8-OHdG was determined based on oxidative DNA Damage (Cell Biolabs, Inc). The mean difference between control and treatment was assessed by applying ANOVA and consider significant at p < 0.05. In this study, we observed that there were a significant of MDA and 8-OHdG levels between group C1 and T; C2 and T within p < 0.05. This result indicates anredera cordifolia leafs extract has an ability to inhibit hypercholesterolemia in wistar rat fed with high cholesterol diet.

Abstract

Lifestyle changes to consumption of variegated instant food may be associated several heath hazards, such as obesity, dyslipidemia, and atheroschlerosis. This study was conducted to investigate the effects of orally administered Sardinella longiceps oil as an anti inflammatory agent on the serum levels of TNF-? and IL-6 considered as biomarkers for atherosclerosis.The study design is an true experimental with randomized pretest and posttest control group design, using 50 Wistar rat equaly divided into 5 groups, i.e. placebo control group 0% and 4 treatment groups each treated daily with 10%, 15 %, 20 % and 25 % fish oil respectively for 6 weeks. Before the treatment was started, all rats were orally fed daily with a high cholesterol diet for 13 weeks to induce atherosclerosis. Our study showed that the intake of 20% fish oil had resulted in the significantly greatest decrease of 45,63 % in the TNF-? serum levels,from 28.62 ± 1.25 to 15.56 ± 7.20 ?g/mL and similar significant decrease 15,42% in of IL-6 serum levels from 134.64 ± 1.98 to 113.87 ± 4.30 ?g/mL. The overall results of our study seemed to imply than in the Wistar rats, oral intake of Sardfinella longiceps oils signifacantly decreased serum levels of TNF-? and IL-6 probably through their anti-inflamatory effects. Futher research to determin the magnitude of effects sardinella longiceps oils on the serum levels TNF-? and IL-6 human.

Abstract

Research on IR64 rice shell ash activation with KOH was carried out. Then, this activated ash known asadsorbent was applied to absorb H2O2 solution. This adsorbent was then applied to absorb peroxide number oftraditional coconut oil.The research includes activation of rice ash with 5%, 10%, and 15% KOH, followed by determination ofrate and isotherm adsorption towards H2O2. Finally, the ash was applied to absorb traditional coconut oil, in order todecrease peroxide number of the oil.Research results indicated that, adsorption rate of the ash with KOH activation is higher than without KOHactivation. Adsorption isotherm patern of the ash with and without activation is following Langmuir adsorption. Riceshell ash activated with 15% KOH can increased adsorption capacity of H2O2 about 297.16 mg/g, whereas withoutactivation the adsortion capacity is about 271.66 mg/g. Activated ash with 15% KOH was the applicated to decreaseperoxide number of traditional coconut oil. In this study, we obtain that the ash can decreased peroxide number until84.4%.

Abstract

Excessive fat consumption can increase blood cholesterol level. Phytosterol composition in the plant can decrease blood cholesterol level. One of the plants that contain phytosterol is beans. This study was to prove the effect of antihypercholesterol the ethanol extract of the beans (Phaseolus vulgaris L.) with variety of doses of 50 mg/Kg BW; 100 mg/Kg BW and 150 mg/Kg BW in male Wistar rats with high fat level condition. Subjects of this study were 24 Wistar rats divided into 6 groups randomly with posttest control group study design. First group as negative control, second group as positive control, and the third (high cholesterol diet and ethanol extract in dose of 50 mg/kg,bw), fourth ( high cholesterol diet and ethanol extractin dose of 100 mg/kg,bw), fifth ( high cholesterol diet and ethanol extract in dose of 150 mg/kg,bw),  sixth (high cholesterol diet and simvastatin drug). The data was analyzed with ANOVA Tukey test. The result of this study shows total cholesterol levels decrease 23,88%; 30,14%; 35,82%. Cholesterol LDL levels decrease 38,09%; 52,38%; 61,35%. Cholesterol HDL levels increase 21,86%; 56,56%; 63,84%. Dose variant on this study giving the best result to decrease total cholesterol levels and LDL was 150 mg/Kg BW. Furthermore, 100 mg/Kg BW dose gave the best result to increase HDL cholesterol level. Based on the results it can be suggested that beans ethanol extract is able to decrease total cholesterol level, LDL and increase HDL cholesterol levels.

Abstract

Identification of antifeedant active compounds from Soursop seeds (Annona muricata Linn) as natural pesticides against Plutella xylostella has been carried out. 1800 g Soursop seed powder  maceration was applied using methanol and n-hexane resulted in 46.42 g extract of methanol and 94.19 g extract of n-hexane. Viscous extract of methanol was diluted with the mixture of methanol:water (7:3), then evaporated until the entire methanol was removed, and after that it was partitioned with chloroform. The antifeedant activity of viscous extract of n-hexane, chloroform and water was tested which showed that the extract of n-hexane had highest activity compared to the others. Separation of 2 g viscous n-hexane extract with Coloumn Chromatography using Gel Silica 60 and n-hexane:aceton (4:1) as the mobile phase resulted in 5 fractions. The fractions were tested against Plutella xyllostella. The results showed that fraction C had the highest antifeedant activity. The isolate of fraction C was then identified using Gas Chromatography-Mass Spectrometry (GC-MS) based on the data base (Database\NISTO2.L and Database\wiley7n.1) suggested the active compounds were ethyl hexadecanoate ester and 11-octadecanoic acid.

Abstract

Changes in consumption behaviour to instant food can cause various health problems, such as obesity, dislipidemia, and atheroschlerosis. A study was conducted to investigate sardinella longiceps oil as anti dislipidemia through increasing HDL-C levels.This is a true experimental with randomized pre and posttest control group designs, emplyoying 50 Wistar rats grouped into five groups, i.e. control group (0% fish oil), treatment 1 group (10% fish oil), treatment 2 group (15% fish oil), treatment 3 group (20% fish oil), and treatment 4 group (25% sardinella longiceps oil). This research observed that intake of 20% sardinella longiceps oil resulted in the higest significant increase of HDL-C of 22.78 % (from 55.97 ±1.25 to 68.72 ± 6.25 mg/dL) for dislipidemia study. Therefore, it can be concluded that intake of sardinella longiceps oil in Wistar rat acts as anti dislipidemia by increasing the HDL-C levels. Further research is required to determine whether the application of sardinella longiceps on human will result in a simillar way, as anti inflammation and anti dislipidemia.

Abstract

Isolation and identification of anti-oxidant  active isolate from soursop leave (Annona muricata L.) was carried out in this study. Methanol was used to macerate an 158 gram sample. This crude extract showed 77,22% of inhibition ability to reduce oxidation of DPPH (difenil-picrylhydrazine) at the fifth minute and 85,42% at the sixtieth minute inhibition time. Petroleum benzene, chloroform, n-buthanol, and water were applied to separate it. N-buthanol extract has the highest anti-oxidant reduction percentage which was 91,10% of inhibition ability to reduce oxidation at the fifth minute and 97,90% at the sixtieth minute inhibition time. From this research, it was obtained that the best dose of n-buthanol fraction applied to decrease uric acid was 200 mg/Kg BW with a percentage decrease of 86,29% (p=0,001).n-buthanol  fraction was identified by gas chromatography-mass spectrometer. The active compounds observed were: benzofuran,2,3-dihidro; tetradecana;  3-ethoxy-1,4,4a,5,6,7,8,8a-octahydroisoquinoline;  2-cyclohexen-1-one, 4-hydroxy-3,5,6-trimethyl-4-(3-oxo-1-butenyl). In conclusion, soursop leave (Annona muricata L.) extract  was active for decreasing uric acid of rat tested. For future research, this method could be applied to human in decreasing uric acid concentration in blood.

Abstract

Oxidative stress is an imbalance condition between free radicals in the body with the anti-oxidant. Superoxide dismutase (SOD) is one of the endogenous anti-oxidant that can neutralize the excess of Reactive Oxygen Species (ROS). The excess of ROS that caused by stres oksidative can also be neutralized with the help of exogenous anti-oxidants. A soursop plant (Annona muricata L.) has anti-oxidant activity, especially in the leaves. This study aims to determine the activity of the leaves of the soursop (Annona muricata L.) in inhibiting the oxidative stress that conducted on the object hypercholesterolemic rats wistar through increasing SOD activity using variations of the extract dose of 50 mg/kg BW; 100 mg/kg BW and 150 mg/kg BW. In this study the dose gives the best result in inhibiting the oxidative stress through an increase in SOD activity was dose of 150 mg/kg BW with percent inhibition of SOD at (81,42% ± 3.91) where the percent inhibition of SOD is directly proportional to the activity of SOD
