Abstract

The process of freeze drying caused detrimental effect on plasma membrane and acrosome of the spermatozoa, even it potentially could alter the chromatin and DNA integrities. On the other hand, DNA integrity is essential for spermatozoa to participate in pronucleus formation during fertilization event. Therefore the evaluation of DNA integrity should be carried out to study the effect of freeze drying process. EDTA, EGTA, and PBS were used as dilution media of spermatozoa prior to freeze drying process to protect the DNA. Toluidine blue staining and comet assay methods were used to evaluate the alteration on chromatin and DNA integrities of spermatozoa, respectively. The results revealed that the highest compacted chromatin after 6 months storage of freeze-dried spermatozoa were observed from EGTA-3 (98%) and EGTA-1 (97%) treatments that had significant differences compared to all PBS treatments (90-92%), but not for fresh spermatozoa (100%). Whereas, the highest compacted DNA integrity of freeze-dried spermatozoa were observed from EGTA-2 (92%) and EGTA-3 (92%) but had no significant differences compared to other treatments including fresh spermatozoa (97%). These results demonstrate that EDTA and EGTA tend to be able to protect chromatin and DNA integrities of ram spermatozoa during freeze-drying and storage compared to PBS. Abstract

The developed Leydig cells-conditioned medium (LCM) contains bioactive materials secreted by Leydig cells in vitro.  LCM was used to evaluate the ability of bone marrow mesenchymal stem cells differentiation. Bone marrow mesenchymal stem cells (1x 106 cell/ml) were cultured in : 1) DMEM supplemented with 10% NBCS as a control (M), 2) M supplemented with 10 ng/ml testosterone; 3) M supplemented with 50%  LCM ; 4) M supplemented with 50% LCM and 2.5 IU/ml hCG. Bone marrow mesenchymal stem cells that were cultured with  LCM has a positive reaction (57.4%) to histochemistry staining 3β-HSD and produced 1.87 ng/ml testosterone. Supplementation of hCG to LCM  increased the positive number of Leydig cells and testosterone production by 74.6% and 12.33 ng/ml (P<0.05). It can be concluded that Leydig cells-conditioned medium can support differentiation of bone marrow mesenchymal stem cells into Leydig cells.

Abstract

Since the discovery of cryopreservation method for bull semen, cryopreservation become an alternative method for maintaining gamet resources of certain animal which is threatened or near extinction. This technology was then applied to the preservation of embryo, oocyte, ovary and testis. The application of intracytoplasmic sperm injection (ICSI) for which sperm motility is unnecessary had supported the effort to create simplified method such as freeze-drying for sperm preservation. Due to the benefit of ICSI over the conventional in vitro fertilization (IVF) the spermatozoon could be mechanically driven to pass through the zona pellucida and entering the cytoplasm of oocytes prior to fertilization. The freeze-drying method is an alternative method in sperm preservation which ignored the motility of sperm. The sperm resulted from this technique is in drying state, therefore, it might be stored in room temperature or in refrigerator. Many reports have claimed that freeze-dried sperm which is not motile but has an intact DNA was able to fertilize oocytes, even produced offspring in mouse.

Abstract

The aim of the present study was to determine the number of follicles, oocyte quality and maturation rate of oocytes from pairs of ovary with different reproductive status in two maturation medium, TCM-199 as control and CR1aa as treatment. The pairs of ovary were classified into four groups: (i) ovaries with corpus luteum (CL) and dominant follicle (DF), (ii) ovaries with CL, without DF, (iii) ovaries with DF, without CL, (iv) ovaries without both CL and DF. Results of the experiment revealed that the greatest number of follicles was observed from ovary with CL without DF (15.88 + 10.68), although not significantly different (P > 0.05) with other status of ovaries. The lowest number (P < 0.05) of A grade oocytes was found from ovary with DF without CL (1.20 + 1.10). The percentage of Metaphase II was highest in TCM-199 (75.51%) with oocytes from ovaries with CL and DF, and the lowest with oocytes from ovaries with DF without CL in TCM-199 and CR1aa (42.86 and 30.95%). The study suggested that the number of oocytes with A grade were influenced by the reproductive status of ovaries. The maturation rate of A grade oocytes was influenced by the quality of oocytes and the composition of maturation medium. Abstract

Low nuclear maturation and fertilization rate is one obstacle in the in vitro embryo production which decrease embryo yield. This problem is presumable related with high production of reactive oxygen species (ROS) during maturation process. Glutathione (GSH) as an antioxidant is well known to overcome effect of ROS production. GSH synthesis in the cytosol part of the oocyte cytoplasm is influenced by cysteine availability. It is therefore, this research was conducted to evaluate the ability of cysteamine to provide cysteine availability as GSH precursor on the nuclear maturation and fertilization rate of sheep oocytes. Results of this experiment revealed additional cysteamine at 150 µm and 200 µm could significantly improve nuclear maturation rate. On the other side, although additional of cysteamine at 50 µm could not improve nuclear maturation rate, however 50 µm cysteamine in the maturation medium could significantly improve the fertilization rate. Based on those experiment results, it seems that the additional cysteamine might be improve not only GSH availability but also the oocyte quality which characterized the ability of pronuclear formation. This finding strongly suggested that additional cysteamine in the maturation medium could improve nuclear maturation and fertilization rate of sheep oocytes.

Abstract

PO cattle have weaknesses to show clear estrus signs which cause difficulty in artificial insemination implementation. The present study was designed to obtain ovarian dynamic as effect of GnRH injection in ovsynch protocol based on progesterone intravaginal device. Heifers (18) and cows (n= 12) were allocated to one of three groups. Cuemate-PGF2α (CP) group inserted with a Cuemate on day 0-7 and injected with prostaglandin on day 7. Cuemate-PGF2α-GnRH (CPG) group was treated as CP group with the addition of GnRH injection on day 9. GnRH-Cuemate-PGF2α-GnRH (GCPG) group was treated as CPG group with addition of GnRH injection on day 0. Ultrasonography was performed on days 0-3, day 7 until ovulation and 7 days after ovulation. Percentage of ovulation synchronization increased significantly (P<0.01) between CP, CPG, and GCPG, respectively, both in heifers (16%, 50%, and 85%, respectively) and cows (0%, 60%, and 100%, respectively), on day 11. Preovulatory follicle diameters between CP, CPG, and GCPG treatments were not different significantly both in heifers (11.9±0.5, 11.9±0.5, and 12.1±0.6 mm, respectively) and cows (11.7±0.4, 11.8±0.7, and 11.1±0.6 mm, respectively). This study concluded that GCPG protocol increased the synchrony of ovulation rate both in cows and heifers, without affecting the follicle preovulatory and CL diameters.Key words: ovarian dynamic, GnRH, ovsych, PO Cattle

Abstract

An experiment was carried out to analyze the application of laparoscopic technique for oocyte collection, in vitro embryo production and embryo transfer in sheep. The first experiment was conducted to observe effect of gonadotropin stimulation on follicle development and laparoscopic technique for oocytes aspiration. In the second experiment, effect of culture system on the embryo development in vitro was assessed and in the third experiment, the application of laparoscopic for embryo transfer has been conducted. The result showed that single dose of gonadotrophin was sufficient to support follicle development significantly and it could help follicle visualization. It also showed that laparoscopic ovum-pick up could be conducted weekly without any restriction The second series experiment showed CR1aa culture system was better than TCM 199 (29.90°/o vs 8.00%) and the changing of media was required to ensure better metabolism process for embryos. The third experiment revealed that embryo transfer could be conducted with an aid from laparoscope. In conclusion, single dose PMSG stimulation is sufficient to support follicle development for /aparoscopic ovum-pick up, the culture media changing affects the successful rate of in vitro embryo production (8% vs 25.66%) and the laparoscopy technique can be used safely for embryo transfer on sheep.Keyword: laparoscopic, oocyte, embryo transfer, sheep

Abstract

Penelitian ini bertujuan untuk mengamati dan mengkaji hubungan suhu, kelembaban dan manajemen ternak terhadap efisiensi reproduksi sapi perah di Kabupaten Bogor. Suhu dan kelembaban udara diamati pagi, siang dan sore hari. Data suhu dan kelembaban udara dikonversi ke nilai indeks suhu kelembaban udara (THI). Pengamatan parameter efisiensi reproduksi dilakukan dengan menghitung nilai Days Open (DO), Conception Rate (CR),  Service per Conception (S/C) dan First Service. Hasil penelitian menunjukkan bahwa nilai THI Acep Farm Kunak (77,21±0,71) lebih tinggi (P<0,05) dari Cifa Farm (71,27±0,50) dan Erif Farm (71,22±0,41). Days Open Acep Farm Kunak (110,76±37,34 hari) lebih panjang (P<0,05) dibandingkan dengan Cifa Farm (88.39±23,80 hari) dan Erif Farm (88,20±22,39 hari). Nilai CR Acep Farm (75,76±0,11%) dan Cifa Farm (66,13±0,10%) tidak berbeda (p>0,05) dibandingkan dengan Erif Farm (42,00±0,08%). Nilai S/C di tiga peternakan tidak menunjukkan perbedaan (p>0,05) masing-masing nilai S/C Cifa Farm (1,42±0,64), Erif Farm (1,52±0,64) dan Acep Farm Kunak (1,64±0,82). First Service Cifa Farm (76,11±13,84) dan Erif Farm (75,17±13,17) berbeda (P<0,05) dengan Acep Farm Kunak (96,42±35,49). Dapat disimpulkan bahwa suhu, kelembaban udara dan manajemen pemeliharaan dapat memengaruhi DO sapi perah di ke tiga peternakan sapi perah di ke tiga lokasi penelitian. Lebih lanjut, diindikasikan THI memengaruhi zona nyaman peternakan sapi perah, namun tidak ditemukan korelasi positif antara nilai THI dengan tingkat efisiensi reproduksi.

Abstract

Penelitian ini dilakukan untuk mengevaluasi penggunaan metode swim up untuk persiapan spermatozoa sebelum fertilisasi terhadap tingkat fertilisasi in vitro spermatozoa kauda epididimis pasca penyimpanan selama 48 jam. Kauda epididimis domba disimpan pada suhu 4 oC selama 0 hari (H-0), 1 hari (H-1) dan 2 hari (H-2), kemudian spermatozoa dikoleksi dan dibekukan. Spermatozoa ejakulat beku digunakan sebagai kontrol. Oosit yang telah matang difertilisasi secara in vitro dengan spermatozoa asal kauda epididimis pasca penyimpanan dan ejakulat menggunakan metode persiapan spermatozoa dengan dan tanpa swim up. Hasil penelitian menunjukkan bahwa spermatozoa asal kauda epididimis yang dikoleksi segera setelah kematian hewan (H-0) memiliki kemampuan yang sama dengan spermatozoa ejakulat (P>0,05). Tingkat fertilisasi spermatozoa kauda epididimis pasca penyimpanan selama 2 hari mengalami penurunan seiring bertambahnya waktu simpan. Penggunaan metode swim up dan tanpa swim up menunjukkan kemampuan fertilisasi yang sama pada spermatozoa ejakulat dan spermatozoa kauda epididimis yang disimpan. Dapat disimpulkan bahwa metode swim up tidak berpengaruh terhadap tingkat fertilisasi in vitro spermatozoa asal kauda epididimis yang disimpan pada suhu 4 °C selama 2 hari. Kemampuan fertilisasi spermatozoa asal kauda epididimis domba yang disimpan pada suhu 4 °C mengalami penurunan sampai hari kedua, namun spermatozoa tersebut masih mampu membuahi oosit secara in vitro.

Abstract

Penelitian tentang sinkronisasi estrus pada sapi perah Friesian Holstein (FH) menggunakan cloprostenol telah dilakukan. Penelitian ini bertujuan untuk mengamati respons dan karakteristik estrus serta pengukuran nilai hambatan arus listrik lendir vagina. Penelitian dilakukan pada 20 ekor sapi FH dengan dua perlakuan injeksi cloprostenol, yaitu injeksi tunggal dengan keberadaan corpus luteum dan injeksi ganda dengan selang waktu injeksi 11 hari. Pengamatan estrus dan pengukuran lendir dilakukan lima hari berturut-turut setelah injeksi cloprostenol yang terakhir. Parameter pengukuran yang diamati meliputi respons, onset, dan durasi estrus, serta nilai hambatan arus listrik lendir vagina. Hasil penelitian menunjukkan bahwa injeksi ganda menghasilkan respons estrus yang lebih baik dibandingkan injeksi tunggal (90% vs 70%), dengan onset estrus lebih pendek (47,55 jam vs 53,28 jam), dan durasi estrus lebih pendek (24,85 jam vs 23,55 jam). Data hambatan arus listrik lendir vagina pada kelompok injeksi ganda lebih rendah dibandingkan dengan injeksi tunggal (187,77 vs 192,14), dengan pola tinggi sebelum estrus, rendah pada saat estrus, dan tinggi kembali setelah estrus, baik pada kelompok injeksi tunggal maupun injeksi ganda. Dapat disimpulkan bahwa respons estrus pada kelompok injeksi ganda lebih baik dibandingkan dengan kelompok injeksi tunggal.
