Abstract

Pakan yang mengandung kadar tinggi lemak dapat menyebabkan hiperlipidemia yang merupakan salah satu faktor pemicu terjadinya perubahan profil lipid. Biji mahoni mengandung flavonoid yang mempunyai kemampuan sebagai agen hipoglikemik. Penelitian ini bertujuan untuk menganalisis pengaruh ekstrak etanol biji mahoni sebagai herba antihiperlipidemia pada tikus putih yang diinduksi ransum tinggi lemak. Tikus putih yang digunakan adalah tikus jantan galur Wistar dengan umur 2 bulan dan berat badan rata-rata kurang lebih 200g. Hewan uji dibagi menjadi 6 kelompok perlakuan dengan 5 ulangan yaitu: Kontrol negatif (P0) diberi ransum komersial; kontrol positif (P1: diberi ransum tinggi lemak); pengobatan P2 (P1+ simvastatin 8mg/g BB); dan P1+ dosis ekstrak etanol biji mahoni sebesar 70;140;280 mg/kg BB(P3;P4 dan P5). Variabel yang diamati adalah kadar kolesterol total, HDL, LDL dan TG. Analisis data menggunakan ANOVA dilanjutkan dengan uji Beda Nyata Terkecil (BNT) dengan tingkat kepercayaan 95% menggunakan software SPSS 10.0. Hasil penelitian menunjukkan bahwa pemberian ekstrak etanol biji mahoni (S. mahagoni) dapat meningkatkan kadar HDL, menurunkan kolesterol LDL dan TG serum darah tikus putih (Rattus norvegicus L.). Foods containing high levels of fat can cause hyperlipidemia which is one of the triggering factors for changes in lipid profiles. Mahogany seeds contain flavonoids which have the ability as hypoglycemic agents. This study aimed to analyze the effect of mahogany seed ethanol extract as an antihyperlipidemic herb in white rats induced by high-fat rations. The white rats used were male Wistar strain rats with the age of 2 months and the average BW of approximately 200g. The test animals were divided into 6 treatment groups with 5 replications, namely: Negative control (P0) was given a commercial ration; positive control (P1: given high-fat ration); P2 treatment (P1+ simvastatin 8mg/g BW); and P1+ the dose of ethanol extract of mahogany seeds of 70;140;280 mg/kg BW(P3;P4 and P5). The variables observed were total cholesterol, HDL, LDL and TG levels. Data analysis using ANOVA was followed by the Least Significant Difference (BNT) test with a 95% confidence level using SPSS 10.0 software. The results showed that the administration of ethanol extract of mahogany seeds (S. mahagoni) could increase HDL levels, reduce LDL cholesterol and TG blood serum white rats (Rattus norvegicus L.).

Abstract

Daun mimba (Azadirachta indica) memiliki banyak manfaat dalam bidang kesehatan. Hasil pengujian yang sudah dilakukan menunjukkan bahwa pemberian ekstrak daun mimba dapat mempengaruhi sistem reproduksi pada hewan jantan, namun efek yang diberikan tidak signifikan terhadap organ testis. Ukuran senyawa yang terlalu besar diduga tidak dapat menembus blood-tissue barrier sehingga perlu dikemas dalam formula dengan ukuran yang lebih kecil, berupa nanopartikel. Tujuan penelitian ini adalah mengetahui pengaruh sediaan nanokitosan ekstrak etanol daun mimba (SNEEDM) terhadap histologis testis tikus putih. Penelitian ini menggunakan 32 ekor tikus umur 2 bulan dibagi menjadi 4 perlakuan yaitu K- (aquades 2ml), K+ (larutan sediaan nanokitosan 2ml). P1 (diberi sediaan nanokitosan dan ekstrak dengan perbandingan 1:0.5), P2 (diberi sediaan nanokitosan dan ekstrak etnaol daun mimba dengan perbandingan 1:1). Hasil penelitian menunjukkan pemberian ekstrak etanol mimba dalam bentuk nanopartikel kitosan berpengaruh tidak nyata terhadap bobot testis, diameter tubulus seminiferus dan skor kerusakan tubulus seminiferus namun berpengaruh nyata terhadap ketebalan epitel tubulus seminiferus (P<0.05), sehingga dapat disimpulkan bahwa pemberian SNEEDM berpengaruh terhadap penurunan ketebalan lapisan epitel tubulus seminiferus. Azadirachta indica have many health benefits. The results of tests that have been carried out show that the administration of neem leaf extract can affect the reproductive system in male animals, but the effect given is not significant to the testicular organs. The size of the compound that is too large is thought to be unable to penetrate the blood-tissue barrier, so it could be packed in a small size particles. The purpose of this study was to determine the effect of neem leaf ethanol extract nanocytosan preparation (SNEEDM) on the histology of white rat testes. This study used 32 rats aged 2 months divided into 4 treatments namely K- (2ml distilled water), K+ (treated by 2ml preparation of nanochitosan solution), P1 (treated by preparation of nanochitosan and neem leaf ethanol extract 1:0.5 in proportion), P2 (treated by preparation of nanochitosan and neem leaf ethanol extract with 1:1 in proportion). The results are the administration of neem ethanol extract in the form of chitosan nanoparticles had no significant effect on testicular weight, seminiferous tubule diameter and seminiferous tubule damage score but had a significant effect on seminiferous tubule epithelial thickness (P <0.05), so it can be concluded that the administration of SNEEDM has an effect on reducing the thickness of the seminiferous tubule epithelial layer.

Abstract

High accumulation of lipids in hepatocytes can trigger an increase in the production of radical compounds, leading to fatty liver disease. This condition induces histological changes in the liver, especially in hepatocytes, and affects the surrounding structures. Mahogany seeds have the potential to act as a hypolipidemic agent in treating fatty liver. Mahogany seeds are known to contain phytochemical compounds which act as antioxidants. This study aimed to determine liver histopathology of rats by inducing high-fat diet after giving mahogany seeds ethanol extract which was observed through the percentage of steatosis, hepatocyte diameter, and hepatosomatic index. This study involved thirty male Sprague Dawley rats, divided into 6 groups: P0 (given commercial feed), P1 (given high-fat diet), P2 (given high-fat diet and simvastatin at a dose of 8 mg/200 g BW), P3, P4, and P5 (given high-fat diet and mahogany seeds ethanol extract at a dose of 14, 28, and 56 mg/200 g BW). The results showed that the hepatosomatic index among all treatment groups did not differ significantly; however, the percentage of steatosis and hepatocyte diameter in the groups given mahogany seeds ethanol extract significantly differed from the group given only high-fat feed. Based on these results, it can be concluded that mahogany seed ethanol extract can maintain the histological structure of the liver in male Sprague Dawley rats with fatty liver. Abstract

Agents of immunomodulation are compounds that have the ability to balance the immune system throughout the body. The neem plant is one of the herbal plants that is often used as traditional medicine. Neem leaves (Azadirachta indica A. Juss) contain antioxidants that have the potential to be antiinflammatory, antiviral, and as an immunomodulatory agent. The constraint in giving medicine orally is the low bioavailability of drugs and the distribution of active compounds of herbal plants. This problem can be solved by packaging herbal plant extracts in nanochitosan. The purpose of this study was to analyze the effect of induced of a nanochitosan ethanol extract of neem leaves on the histological structure of the spleen tissue of female Wistar white rats. This study used a randomized design (CRD) which was divided into 3 treatment groups with 4 repetitions. Treatments include control (P0), an ethanol extract of neem leaves 2 ml/head/day (P1), and nanochitosan preparation of neem leaves 2 ml/head/day (P2). The parameters measured were the diameter of white pulp, the diameter of germinal centre, the area of white pulp, and the area of germinal centre in the spleen. Data on the diameter and the area of the white pulp and germinal centre were analyzed statistically parametrically with ANOVA with a 95% level of confidence, if the data were not normally distributed, the Kruskall Wallis nonparametric test was carried out. Data that were significantly different were carried out by Duncan’s further test. The results showed that the treatment of nanochitosan preparations of neem leaf ethanol extract had a significant difference in the diameter of the white pulp and germinal centre as well as the area of the white pulp and germinal centre (P<0,05). The conclusion of this study was that the nanochitosan preparation ethanol extract of neem leaves at a dose of 14 mg/kg BB can increase the diameter and the area of the white pulp and germinal centre.

Abstract

Tanaman mimba merupakan tanaman herbal yang memiliki kandungan antioksidan tinggi dan berpotensi sebagai obat. Kendala dalam pemberian secara oral adalah rendahnya bioavaibilitas obat dan distribusi senyawa aktif tanaman herbal. Salah satu yang bisa dilakukan untuk mengatasi masalah tersebut adalah dengan mengemas ekstak tanaman herbal ke dalam bentuk partikel lebih kecil berukuran nano. Tujuan penelitian ini adalah untuk melihat respon struktur dan fungsi hepar tikus putih betina dengan pemberian sediaan nanokitosan ekstrak etanol daun mimba. Penelitian ini menggunakan 12 ekor tikus umur 2 bulan yang di bagi menjadi 3 perlakuan dengan 4 ulangan yaitu P0 (kelompok tikus normal yang di induksi dengan aquades 2ml), P1(Tikus normal yang di induksi dengan ekstrak etanol daun mimba 2ml). P2 (tikus normal yang di induksi dengan nanokitosan : ekstrak etanol daun mimba 2:1). Desain penelitian menggunakan RAL (Rancangan Acak Lengkap). Analisis data menggunakan software SPSS versi 24 dengan uji ANOVA taraf signifikansi 5% menunjukkan bahwa pemberian sediaan nanokitosan ekstrak etanol daun mimba 1:2 berpengaruh tidak nyata terhadap Hepatosomatic index, diameter hepatosit, namun hasil uji Kruskal Wallis yang dilanjutkan dengan uji Mann Whitney berpengaruh nyata terhadap skor kerusakan hepatosit (P<0,05), sehingga dapat disimpulkan bahwa pemberian sediaan nanokitosan ekstrak etanol daun mimba dapat mengantarkan senyawa bioaktif daun mimba ke organ target yaitu hepar serta dapat mengurangi kerusakan histologis hepar akibat senyawa toksik mimba. Neem are herbal plants that have high antioxidant content and have potential as medicine. One that can be done to overcome this problem is by packaging herbal plant extracts into the form of smaller nano-sized particles. The purpose of this study was to see the response of the structure and function of female white rats by giving nanochitosan preparations of neem leaf ethanol extract. This study used 12 mice aged 2 months which were divided into 3 treatments, each treatment had 4 repeats, namely P0 (group of normal rats induced with 2 ml aquades), P1 (normal mice induced with 2 ml neem leaf ethanol extract). P2 (normal mice induced with 2 ml nanochitosan: neem leaf ethanol extract 2:1). The study design used RAL (Complete Randomized Design). Data analysis using SPSS software version 24 with ANOVA test significance level of 5% showed that administration of neem leaf ethanol extract nanochitosan preparation 1:2 had no real effect on hepatosomatic index, hepatocyte diameter, but the results of the Kruskal Wallis test followed by the Mann Whitney test had a real effect on hepatocyte damage score (P<0.05), so it can be concluded that giving neem leaf ethanol extract nanochitosan preparations can delivering neem leaf bioactive compounds to the target organ, namely the liver and can minimize histological damage and liver function due to neem toxic compounds.

Abstract

Doksisiklin merupakan antibiotik yang digunakan untuk mengatasi infeksi. Doksisiklin dengan dosis berlebih dapat menimbulkan radikal bebas yang mampu menyebabkan toksik bagi tubuh. Bunga telang (Clitoria ternatea L.) merupakan tanaman yang mengandung senyawa antioksidan, seperti flavonoid, antosianin, asam fenolik, prosianidin, dan flavonol glikosida. Antioksidan yang terkandung di dalam bunga telang berfungsi dalam menetralisir radikal bebas dalam tubuh. Tujuan dari penelitian ini, yaitu menganalisis histomorfometri ginjal tikus putih (Rattus norvegicus L.) strain Sprague Dawley setelah pemberian doksisiklin dan ekstrak etanol bunga telang (Clitoria ternatea L.). Tikus diaklimatisasi selama 1 minggu dan diberi perlakuan selama 2 minggu. Kelompok perlakuan yang diujikan pada penelitian ini, yaitu P0 (1 mL aquades), P1 (1 mL larutan doksisiklin), P2 (1 mL larutan ekstrak etanol bunga telang), P3 (1 mL larutan doksisiklin dan 1 mL larutan ekstrak etanol bunga telang). Hasil uji ANOVA satu arah menunjukkan bahwa hasil berbeda tidak nyata (P>0,05) terhadap diameter glomerulus, jarak ruang kapsula Bowman, tebal epitel tubulus kontortus distal, tebal epitel tubulus kontortus proksimal, bobot ginjal, dan konsumsi air minum pada tikus setelah perlakuan. Kesimpulan dari penelitian ini, yaitu pemberian doksisiklin dan ekstrak etanol bunga telang tidak memberikan dampak negatif pada struktur histologi ginjal tikus putih.

Abstract

Mimba merupakan tanaman obat yang umumnya dikonsumsi secara oral, konsumsi obat secara oral memiliki tingkat bioavaibilitas yang rendah dalam tubuh. Nanokitosan mampu mengenkapsulasi senyawa aktif dan mendistribusikannya ke sirkulasi sistemik serta meminimalisir efek toksik. Tujuan dari penelitian ini adalah menganalisis pengaruh paparan sediaan nanokitosan ekstrak etanol daun mimba (Azadirachta indica A. juss) terhadap struktur histologi duodenum tikus putih (Rattus norvegicus) betina. Metode penelitian menggunakan desain Rancangan Acak Lengkap (RAL) terdiri atas 3 perlakuan dan 4 pengulangan. Perlakuan terdiri dari P0 (kontrol), P1 (ekstrak etanol daun mimba), dan P2 (nanokitosan ekstrak etanol daun mimba) masing-masing perlakuan diberikan dalam 2 ml/ekor/hari selama 21 hari. Variabel yang diukur berupa diameter duodenum, tebal lapisan muskularis, panjang vili, dan skor kerusakan deskuamasi vili melalui skoring dengan skor 0-3 Data dianalisis secara statistik parametrik dengan ANOVA pada taraf kepercayaan 95. Hasil penelitian menunjukkan bahwa diameter duodenum, tebal lapisan muskularis, dan panjang vili tidak berbeda nyata (P>0,05). Skor kerusakan deskuamasi vili menunjukkan P0 menunjukkan vili normal (skor 0), P1 terdapat kerusakan sedang (skor 2), dan P2 terdapat kerusakan ringan (skor 1). Kesimpulan menunjukkan bahwa sediaan ekstrak etanol daun mimba dalam bentuk nanopartikel mampu memberikan efek proteksi terhadap kerusakan struktur duodenum tikus.  Neem is a medicinal plant that is generally consumed orally, oral conpsumption of drugs has a low level of bioavailability in the body. Nanochitosan able to encapsulate active compounds and distributing them throughout the systemic circulation and minimize toxic effects.The purpose of this study was to analyze the effect of treatment of nanochitosan ethanol extract of neem leaves (Azadirachta indica A. juss) on the histological structure of the duodenum of female white rats. This study used a Completely Randomized Design (CRD) consisted of 3 treatments and 4 replication. The treatments was devided into P0 (control), P1 (ethanol extract of neem leaves), and P2 (nanochitosan ethanol extract of neem leaves), each treatment was given 2 ml/head/day for 21 days. The variables measured were duodenal diameter, muscularis layer thickness, villi length, and villi desquamation damage score by scoring with scores 0-3. Data were analyzed with ANOVA at a confidence level of 95%. The results showed that the duodenum diameter, muscularis layer thickness and villi length had no significant difference (p>0,05). The villi desquamation damage score P0 showed normal villi (score 0), P1 had moderate damage (score 2), and P2 had slight damage (score 1). The conclusion shows that treatment of neem leaf ethanol extract in the form of nanoparticles was able to protect the structure of the rat duodenum from damage.

Abstract

Tujuan penelitian ini adalah untuk mengetahui mikroanatomi ginjal tikus putih (Rattus norvegicus L.) galur Sprague-Dawley hiperlipidemia setelah pemberian ekstrak etanol biji mahoni (Swietenia mahagoni L. Jacq) terhadap. Model hewan hiperlipidemia dikondisikan dengan diberi perlakuan pakan tinggi lemak. Metode yang digunakan adalah dengan memberi ekstrak etanol biji mahoni secara oral selama 4 minggu. Parameter pengamatan yaitu bobot ginjal, diameter glomerulus, tebal ruang kapsula bowman, diameter tubulus kontortus proksimal, tebal sel epitel tubulus kontortus proksimal, diameter tubulus kontortus distal dan tebal sel epitel tubulus kontortus distal. Data dianalisa menggunakan ANOVA dan uji Kruskal-Wallis. Hasil penelitian menunjukan bahwa pemberian ekstrak etanol biji mahoni dengan konsentrasi 14mg/200gBB efektif dalam menurunkan diameter glomerulus, tebal ruang kapsula bowman dan diameter tubulus kontortus distal, konsentrasi 28mg/200gBB efektif menurunkan tebal sel epitel tubulus kontortus proksimal dan tebal sel epitel tubulus kontortus distal, konsentrasi 56mg/200gBB efektif dalam menurunkan bobot ginjal dan diameter tubulus kontortus proksimal. Kesimpulan dari penelitian ini, yaitu pemberian ekstrak etanol biji mahoni dosis 14 mg dan 28 mg dapat dikatakan memiliki potensi yang sama dengan obat simvastatin dengan dosis 8 mg dalam menekan efek hiperlipidemia dan mempertahankan mikroanatomi ginjal tikus putih hiperlipidemia.  The purpose of this study was to determine the microanatomy of the kidneys of white rats (Rattus norvegicus L.) of the Sprague-Dawley strain of hyperlipidemic after administration of ethanol extract of mahogany seeds (Swietenia mahagoni L. Jacq) against. The animal model of hyperlipidemia was conditioned by being given high-fat feed treatment. The method used was to give ethanol extract of mahogany seeds orally for 4 weeks. Observation parameters were kidney weight, glomerular diameter, thickness of Bowman's capsule space, diameter of proximal convoluted tubule, thickness of proximal convoluted tubule epithelial cells, diameter of distal convoluted tubule and thickness of distal convoluted tubule epithelial cells. Data were analyzed using ANOVA and the Kruskal-Wallis test. The results showed that the administration of mahogany seed ethanol extract with a concentration of 14 mg/200 gBW was effective in reducing glomerular diameter, Bowman's capsule space thickness and distal convoluted tubule diameter, a concentration of 28 mg/200 gBW was effective in reducing the thickness of proximal convoluted tubule epithelial cells and the thickness of distal convoluted tubule epithelial cells, a concentration of 56 mg/200 gBW was effective in reducing kidney weight and proximal convoluted tubule diameter. The conclusion of this study, namely the administration of mahogany seed ethanol extract at doses of 14 mg and 28 mg can be said to have the same potential as simvastatin with a dose of 8 mg in suppressing the effects of hyperlipidemia and maintaining the kidney microanatomy of hyperlipidemic white rats.

Abstract

Nanokitosan adalah jenis polisakarida hasil turunan kitosan yang bersumber dari eksoskeleton arthropoda laut seperti krustasea (udang, lobster, kepiting, teritip, kril) serta kutikula serangga. Nanokitosan telah disetujui sebagai feed aditif oleh Badan Pengawas Makanan dan Obat-obatan Amerika Serikat tetapi masih jarang digunakan oleh para peternak lokal di Indonesia. Berdasarkan potensi tersebut dilakukan penelitian dengan tujuan menganalisis pengaruh kadar pemberian aditif pakan nanokitosan dalam pakan terhadap bobot badan dan somatometri itik pengging yang ditunjukkan dengan panjang badan, lingkar toraks, lingkar abdomen, panjang leher, paruh, sayap, tibia, dan tarsometatarsus. Penelitian ini menggunakan Rancangan Acak Lengkap yang terdiri atas 5 perlakuan meliputi P0 (pakan basal 100%), P0 adalah pakan basal tanpa aditif pakan nanokitosan (100% pakan basal); P1, P2, P3, dan P4 adalah pakan basal yang diberi aditif pakan nanokitosan, masing-masing dengan kadar 2,5; 5; 7,5; dan 10 g/kg pakan, tiap perlakuan diulang sebanyak lima kali. Pakan dan minum diberikan secara ad libitum. Data dianalisis menggunakan uji Kruskal Wallis. Berdasarkan hasil analisis penelitian disimpulkan bahwa nanokitosan sebagai aditif pakan dapat mempertahankan bobot badan dan ukuran somatometri pada itik pengging berdasarkan indikator yang diukur, meliputi panjang badan, lingkar toraks dan abdomen, panjang sayap, leher, paruh, tibia, dan tarsometatarsus. Penggunaan nanokitosan sebagai aditif pakan dapat diberikan pada itik pengging (Anas platyrhynchos) umur 4 minggu (fase starter) agar dapat diperoleh efek yang optimal terhadap proses pencernaan, metabolisme, dan performa produktivitas yang terkait dengan bobot badan dan ukuran somatometri pada itik pengging.

Abstract

Neem leaves (Azadirachta indica A. Juss) contain compounds that have antifertility potential. The constraint in giving medicine orally is the low bioavailability and distribution of active compounds in herbal plants. This problem can be solved by packing herbal plant extracts in nanochitosan. The aim of this study was to analyze the effect of treating with a nanochitosan ethanol extract of neem leaves on the histological structure of the white rat uterine. This study was conducted for 8 months used a randomized design (CRD), which was divided into 3 treatment groups with 4 repetitions. Treatments is given for 21 days, including P0 (an aquades 2 mL/animal/day), P1 (an ethanol extract of neem leaves 2 mL/animal/day, and P2 (a nanochitosan preparation of neem leaves 2 mL/head/day. The variables measured were endometrial thickness, number of uterine glands, uterine diameter, and uterine weight. Numeric datas were analyzed statistically parametrically using the ANOVA test at a confidence level of 95%. The results showed that the endometrial thickness, uterine diameter, number of uterine glands, and uterine weight of P0 were normal, and did not significantly different (P >0.05) from P1 and P2. The conclusion of this study is that the treatment of nanochitosan ethanol extract of neem leaves has the same potential with ethanol extract of neem in affecting the histological structure of white rats uterine.
