Abstract

Research on the potential of the Surian tree (Toona sinensis) for medicine has been carried out, especially for the leaves. However, the bark section has not been done much. This study aimed to determine the inhibitory activity of -glucosidase and IC50 extract as well as the fraction of surian bark as antidiabetic in vitro. The reflux method with water at 90oC and maceration using 70% ethanol at room temperature was carried out to obtain aqueous and ethanol extracts. Furthermore, the ethanol extract was fractionated successively using n-hexane, diethyl ether, and ethyl acetate. Phytochemical screening test showed that the aqueous extract contained flavonoids, saponins, and tannins. Meanwhile, the 70% ethanol extract and its fraction showed terpenoid content, in addition to flavonoid compounds, saponins, and tannins. The best -glucosidase inhibition was shown by 70% ethanol extract, which was 70.45 ppm followed by ethyl acetate fraction, n-hexane fraction, diethyl ether fraction, and water extract of surian bark with values of 81.96 g/mL, 98.76 g/mL, respectively. , 140.25 g/mL, and 242.85 g/mL. Kinetics of -glucosidase inhibition showed that 70% ethanol extract of surian bark inhibited through a non-competitive inhibition mechanism, characterized by a decrease in Vmax from 8.29x10-3 mM/min to 1.65x10-3 mM/min and a decrease in Km from 0.17 mM to 0.03 mM.

Abstract

Abstract

Red ginger (Zingiber officinale Roxb. var. Rubrum) contains phenolic compounds that have potential to reduce blood glucose levels (antihyperglycemic) in patients with diabetes mellitus. This study aimed to determine the inhibitory activity toward α-glucosidase, antioxidant activity in reducing DPPH free radicals, and total phenolic content of aqueous extract of red ginger. The red ginger was extracted using water solvent at 100 oC for 15 minutes. Analysis of antioxidant activity, inhibitory activity of α-glucosidase, and total phenolic content using the spectrofotometry. Antioxidant activity showed that the extracts from the first, second and third replicates were not significantly different (p<0.05). α-glucosidase inhibition activity showed that the 1st replicate extract had the highest value of 97.729% and was significantly different (p<0.05) from the 2nd and 3rd replicate extracts and acarbose. The total phenolic content showed that the 2nd replicate extract had the highest value, namely 27,624 mg GAE/g extract and was significantly different (p<0.05) from the 1st and 3rd replicate extracts. The 1st, 2nd, and 3rd replicates of red ginger aqueous extract has the same antioxidant activity and equivalent glucosidase inhibitory activity as acarbose.

Abstract

Red betel (Piper crocatum) has antihyperglycemic activity for diabetic treatment. The purpose of this study was to investigate the activity of DPPH free radical scavenging, the α-glucosidase inhibitory activity, and total phenolics of the aqueous extract of P. crocatum (1st extract, 1st extract residue, and 2nd extract residue). The infundation method was used with spectrophotometric test techniques. The red betel leaves extract in all extracts and extract residues had a small of α-glucosidase inhibition activity ranging from 0.76% - 1.43%. Meanwhile, the antioxidant activity of DPPH and total phenolic levels showed significantly different results (p<0.05) in the 1st extract, 1st extract residue, and 2nd extract residue. The highest antioxidant capacity and total phenolic content were found in the first extract, it were 59.64 ± 0.18 mg AAE/g extract and 55.50 ± 0.14 mg GAE/g, respectively. Gradual extraction reduced DPPH free radical scavenging activity, α-glucosidase inhibition activity and phenolic total content.

Abstract

Tamanu or nyamplung oil, derived from Calophyllum inophyllum L., is commonly used in traditional medicine and occasionally used as a moisturizer in skincare cosmetics. In silico predictions suggested that tamanu oil contained compounds capable of inhibiting tyrosinase activity. This study aimed to measure the surface tension of tamanu oil and evaluate its tyrosinase inhibitory activity in vitro. The surface tension of tamanu oil was measured using the du-Nouy ring method with a tensiometer, while tyrosinase inhibitory activity was assessed by spectrophotometry using a microplate reader. The study revealed that the surface tension of tamanu oil was measured at 41.83±0.76 mN/m, and the tyrosinase inhibitory activity of tamanu oil was determined to be 83.75±0.41%. Notably, the inhibitory activity of tamanu oil was comparable to that of the positive control, kojic acid, which exhibited a tyrosinase inhibition of 84.59±2.04%. We concluded that tamanu oil has potential as a natural surfactant raw material and may serve as an effective tyrosinase inhibitor.   Keywords: Calophyllum inophyllum L., nyamplung, surface tension, tamanu oil, tyrosinase inhibitor

Abstract

Ngadireso Village has significant agricultural potential, particularly in cultivating horticultural commodities such as cabbage, chili, and oranges, which serve as the main sources of livelihood for the local community. However, agricultural practices in Ngadireso Village are still largely dominated by the use of chemical-based fertilizers, pesticides, and herbicides that pose risks to human health and the environment, while also increasing production costs. The community service activity conducted in Ngadireso Village aimed to enhance public understanding of the production of compost fertilizer, natural biopesticides, and bioherbicides that are more environmentally friendly, easy to make, and cost-effective. The methods used included socialization, demonstration of the production process, and hands-on practice with the Sri Rahayu 1 farmer group in Ngadireso Village. The materials used were locally sourced and easily obtainable within the village area. The results of the activity showed that the community was highly enthusiastic in participating in the training and gained a better understanding of the importance of sustainable agriculture. In addition to reducing dependency on synthetic chemicals, this activity also encouraged the utilization of local waste and promoted farmers’ self-reliance. It is expected that through this initiative, the people of Ngadireso Village will continue to develop a healthier, more economical, and environmentally conscious agricultural system.
