Abstract

Abstrak Pengembangan minuman fungsional dari ekstrak sirih merah yang ditambahkan dengan beberapa ekstrak rempah telah dilakukan sebelumnya. Akan tetapi, hasil uji sensori masih tergolong rendah karena masih terdapat rasa pahit pada minuman fungsional tersebut. Penelitian ini bertujuan untuk mengembangkan minuman fungsional berbasis ekstrak daun sirih merah dengan teknologi nanoenkapsulasi dan mengevaluasi total fenolik, aktivitas antioksidan in vitro serta mutu sensori. Pembuatan minuman fungsional nanoenkapsulasi menggunakan carrier agent berupa gum arab dan maltodekstrin (1:3; 1:1; 3:1), sedangkan active agent berupa campuran ekstrak daun sirih merah dan rempah-rempah (kayu manis, jahe merah, jeruk nipis). Aktivitas antioksidan menggunakan metode CUPRAC. Hasil penelitian menunjukkan bahwa minuman ready to drink memiliki nilai total fenol (786,23±1,52 mg GAE/g) dan aktivitas antioksidan (1583,97±85,52 µM troloks/g ekstrak) lebih baik dibandingkan dengan minuman fungsional nanoenkapsulasi. Akan tetapi, minuman fungsional nanoenkapsulasi memberikan mutu sensori yang lebih disukai dibandingkan dengan minuman ready to drink. Nanoenkapsulasi dapat memperbaiki mutu sensori minuman fungsional sirih merah, tetapi menurunkan nilai total fenol dan aktivitas antioksidan in vitro. Kata kunci: gum arab, maltodekstrin, minuman fungsional AbstractThe development of functional drink from red betel extract which is added with several spice extracts has been previously conducted. However, the result of the sensory test was still not good because there was still a bitter taste in the functional drink. This study aimed to develop functional drink based on red betel extract with nanoencapsulation technology and evaluate total phenolic, in vitro antioxidant activity, as well as sensory value. The nanoencapsulated functional drink was made from carrier agent, which were arabic gum and maltodextrin (1:1; 1:3; 3:1), while the active agent were a mixture of red betel extract and spices (cinnamon, red ginger, lime juice). Antioxidant activity used the CUPRAC method. The result showed that ready to drink (RTD) drink which had a total phenolic value (786.23±1.52 mg GAE/g) and antioxidant  activity (1583.97±85.52 µM trolox/g extract), was better than the nanoencapsulated functional drink. However, nanoencapsulated functional drink provide sensory quality which is more preferable than the RTD drink. The nanoencapsulated technology could improved sensory value of red betel functional drink, but it decreased total phenolic value and antioxidant activity in vitro. Abstract

Since 500 years ago sea cucumber has been used as wound healing medicine. Sea cucumbers are thought to function as a cell growth factor and have the potential as an antibacterial agent. This study aims to determine whether the methanol extract of sea cucumber has antibacterial activity against Staphylococcus aureus and Pseudomonas aeruginosa so that it can be used to help diabetic wound healing of people infected with the same bacteria. The extraction method used was maceration with methanol to dried frozen simplicia. The simplicia was obtained by freeze drying to liquid preparation of heated fresh sea cucumber. The antibacterial activity test was performed using well diffusion method. There were no any antibacterial activity found in the two methanol extracts of sea cucumber. Abstract

This study is aimed for finding alternative ways to treat diabetes. Many studies have beendone before, but focused on identification of α-glucosidase inhibition mechanism. This study wasconducted by extracting Caesalpinia bonduc seed using two methods maceration by ethanol 96%and the others boil by water. The ethanol extract was then fractioned into three fractions, n-hexane,ethyl acetate, and ethanol. To find the bioactive compounds, the ethanol extract was then tested byseveral phytochemistry tests. The phytochemistry test showed positive results for alkaloid, flavonoid,saponin and triterpenoid. The inhibition mechanism was tested using pNPG. The inhibition testshown that the mechanism was a competitive inhibition, by the Dixon plot. The calculated inhibitionconcentrations for 50% substrate (IC50) for ethyl acetate fraction are 1655.8079 μg/ml for 5 mMsubstrate and 803.9521 μg/ml for 10 mM substrate. From the results of this study, we concluded thatthe extract of Caesalpinia bonduc has inhibition activity toward α-glucosidase enzyme. Abstract

Current researches for appropriate hypoglycemic agents focused on plants for traditional medicine. Traditionally in diabetic treatment, people used decoctions of Piper crocatum (Piperaceae). However, there is no phytochemical data of decoctions extract of P. crocatum. Hence, the aims of this study were to explore the phytochemical of P. crocatum decoctions and its antihyperglicemic activity. Fresh leaves of P. crocatum were boiled in water to obtain decoction and were examined phytochemical compounds by using Harbone assay. Antihyperglicemic activity of P. crocatum decoction extract was orally fed to alloxan induced diabetic rats. Results showed that P. crocatum decoction extract contained flavonoids, tanins, and alkaloids. Ten days of daily treatment of various doses decoction extract of P. crocatum led to reduce blood sugar level by 10-38% and prevent fall in body weight level by 5-52%. This result showed the same activity as Daonil treatment, which was the diabetic drug. Hence, this extract showed antihyperglicemic activity in alloxan-induced diabetic rats and increasing of their body weight. Key words: Piper crocatum, antihyperglicemic, phytochemical

Abstract

The excessive number of free radicals in human body could increase lipid peroxides and causes a variety of degenerative diseases. Piper crocatum is a plant containing natural chemicals that may have an antioxidant ability to inhibit fatty acid oxidation and reduce free radicals, however, there is no published scientific reports up to now. Therefore, the objective of this research is to study the ability of ethanol extract of P. crocatum as an antioxidant. The leaves of P. crocatum (25 g) were extracted with 100 ml of 70% ethanol. Antioxidant activity was measured by thiobarbituric acid method and compared with 2,2-diphenil-1-picryl hydrazyl method with α-tocopherol as the standard antioxidant. The results showed that the extract of 200 ppm inhibited fatty acids oxidation by 80.40% and IC50 for free radical scavenging was 85.82 ppm. There was no significant different (α = 0.05) inhibition of unsaturated fatty acids oxidation between the sample and α-tocopherol at 200 ppm. This study suggested that the leaves extract of P. crocatum has a potential natural antioxidant.

Abstract

Piper crocatum and Cinnamomum burmannii formulations is known to be a new diabetes functional drink. Thus, its toxicological profile needs to be studied. At present, the formulation was evaluated for the repeated dose toxicity study. The Sprague dawley albino rats were treated with P. crocatum and C. burmannii formulations (0, 630, 1260, and 1890 mg/kg) and administered orally for a period of 28 days in albino rats. The effects on body weight, food and water consumption, organ weight, hematology, clinical biochemistry as well as histology were studied. There were no significant differences in the body weight, organ weights and feeding habits between control and treated animals. Hematological analysis showed no marked differences in any of the parameters examined in either the control or treated groups. There were no significant changes that occurred in the blood chemistry analysis including glucose, cholesterol, triglycerides, creatinine, SGPT, and SGOT in experimental animals. Pathologically, neither gross abnormalities nor histopathological changes were observed. The formulation of P. crocatum and C. burmannii was found safe in repeated dose toxicity studies.

Abstract

Functional drinks containing the leaves of Java tea has been shown to offer many health functionalities, particularly to lower blood glucose levels, due to its bioactive compounds. Nanoencapsulation technology was reported being able to protect these bioactive compounds and also enhance their bioavailability. This study examined the use of nanoencapsulation techniques to improve the bioavailability of Java tea-based functional effervescent drink and enhance its antihyperglycemic activity. Three versions were prepared in this study: ready to drink (RTD) as the control, microencapsulated, and nanoencapsulated. They were all measured for their total phenolic content (TPC) before being tested for their antihyperglycemic activity. The TPC of RTD, microencapsulated, and nanoencapsulated Java tea-based functional drinks (JTFD) were 998,425, 735,433, and 663,517 ppm, respectively. The antihyperglycemic activities were evaluated by feeding each beverage to streptozotocin-induced diabetic Sprague Dawley rats for fourteen days. Nanoencapsulated JTFD was found to be more superior than the other two formulations to decrease the blood glucose level (7.98%), maintain the feed intake, body weight, improve the viability of Langerhans and beta-cells by 49.09%, 32.50%, respectively.

Abstract

The aim of this study was to explore the analysis proximate and toxicity of the decoctions of P. crocatum. Fresh leaves of P. crocatum were boiled in water in order to obtain decoction and were examined for its chemical compounds by using SNI 01-2891-1992 method for proximate analysis. Toxicity of decoction extract of P. crocatum was orally fed to rats (Sprague dawley). The results showed that P. crocatum leaves contains 9.27% water, 14.33% ash, 3.96% fat, 22.63% proteins, and 59.08% carbohydrates. Acute toxicity test showed that all rats were still alive after 7 days treatment with P. crocatum decoction for all dose groups (0, 5, 10, and 20 g/kg BB). LC50 value of P. crocatum decoction was 544.82 ppm, meaning that the decoction was relatively harmless and bioactive.

Abstract

This study aims to determine the stability of total phenolic content, antioxidant activity, α-glucosidase inhibitory activity, physical properties (total dissolved solid, pH, color), and total microorganism of antidiabetic functional beverage at certain storage period. The functional beverage made from red betel leaf, cinnamon, and red ginger by using infuse method. Parameters were analyzed for 28 days on 10°C and 30°C of storage temperature. Antioxidant activity was measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, α-glucosidase inhibitory activity was determined in vitro, and total microorganism was measured by total plate count (TPC) method. The results showed that there were increasement in total dissolved solid of 1.92% (10°C) and 3.85% (30°C), and pH values of 15.30% (10°C) and 13.57% (30°C). In contrary, there were decreasement in total phenolic content of 61.63% (10°C) and 79.73% (30°C), antioxidant activity of 46.80% (10°C) and 53.19% (30°C), and α-glucosidase inhibitory activity of 5.43% (10°C) and 0.39% (30°C). Total microorganism at 10oC was <1.0×100 CFU/mL until day 28, while at 30oC was 2.0×100 CFU/mL on day 28. Based on the results, the functional beverage was good in physical and microbiological quality and still have biological activity as an antidiabetic beverage during storage.

One of the efforts of cancer treatment is the utilization of natural compounds. One of the microorganisms having the potential as anticancer agent is endophytic fungi. Endophytic fungi from the marine habitat can be isolated from sea weeds, sea grasses, sponges, and mangroves. Xylaria psidii KT30, a marine fungus used in this study was isolated from red seaweed Kappaphycus alvarezii. Xylaria psidii KT30 was cultivated in potato dextrose broth medium for nine days at room temperature 27-29°C in shaking condition. This study aimed to obtain protein fractions from X. psidii KT30 and determine their toxicity againt Chang and HeLa cells. The fractionation process was conducted using DEAE Sephadex A-50 column chromatography and the toxicity was determined by Brine Shrimp Lethality Test (BSLT). The metabolites excreted in the culture broth was extracted using 90% of ammonium sulphate. The extract was then tested for their toxicity against HeLa and Chang cells by Microculture Tetrazolium Technique (MTT) assay.The results revealed that LC50 of the protein extract of X. psidii KT30 was 104.95 ppm and IC50 was 69.9 ppm. Based on the National Cancer Institute (NCI), this value showed moderate cytotoxicity against HeLa cells.
