Abstract

Kecapi (Sandoricum koetjape (Burm.f.) Merr) merupakan salah satu tanaman obat tradisional dari famili Meliaceae, mengandung senyawa flavonoid, saponin dan polifenol yang dapat digunakan sebagai antifungi. Penelitian ini bertujuan untuk mengetahui senyawa dalam kulit batang S. koetjape yang bersifat antifungi terhadap C. albicans. Penelitian dilakukan dengan dua tahap yaitu ekstraksi dengan metode maserasi menggunakan pelarut metanol dan fraksinasi dengan metode kromatografi kolom menggunakan pelarut n-heksan,kloroform, etil asetat dan metanol. Selanjutnya dilakukan uji aktivitas terhadap C. albicans dan identifikasi fraksi aktif dengan Gas Chromatograpy-Mass Spectrometry (GC-MS). Hasil penelitian menunjukkan bahwa ekstrak metanol kulit batang S. koetjape memiliki aktivitas antifungi terhadap C. albicans sebesar 39,65% b/v. Ekstrak metanol di fraksinasi berdasarkan gradient polaritasmenghasilkan6 fraksi dengan aktivitas antifungi terbesar yaitu fraksi III (43,02%b/v), diikuti dengan fraksi V (39,66%b/v), fraksi IV (39,02%b/v), fraksi II (37,91%b/v), fraksi VI (35,49%b/v) dan fraksi I (33,72%b/v). Identifikasi senyawa dengan GC-MS dilakukan pada fraksi III, dan diperoleh senyawa yang berpotensi sebagai antifungi adalah α-gurjunene, trans-caryophyllene, aromadendrene, α-humulene, β-caryophyllene,δ-cadinene, alloaromadendren, octadecanoic acid (as. stearat), hexadecanoic acid metil ester (metil palmitat), hexadecanoic acid (as. palmitate), 9-octadecenoic acid metil ester (metil elaidate), 9-octadecenoic acid (as. oleat)

Abstract

Penelitian ini bertujuan untuk melakukan penapisan dengan cara ekstraksi dan dilanjutkan dengan kromatografi lapis tipis untuk melihat profil senyawa dan melakukan uji aktivitas sitotoksik dan induksi apotosis dari ekstrak tersebut. Ekstraksi batang brotowali dilakukan dengan maserasi menggunakan etanol 96% kemudian di fraksinasi dengan n-heksan, kloroform, etilasetat berdasarkan tingkat polaritas dan dilanjutkan kromatografi lapis tipis menggunakan fase diam silika gel GF 254 dan fase gerak campuran, spot yang ada dilihat dibawah UV 254 nm dan 366 nm serta disemprot dengan reagen penampak noda. Ekstrak dan fraksi kemudian diuji aktivitas sitotosiknya dengan metode MTT dan induksi apoptosis dengan metode double staining dan flowcytometri. Hasil penelitian menunjukkan bahwa ekstrak etanolik mengandung senyawa flavanoid, fenolik dan terpenoid. Ekstrak mempunyai efek sitotoksik IC50sebesar 314 µg/ml dan fraksi F3 sebagai fraksi aktif mempunyai efek sitotoksik dengan IC50 sebesar 146 µg/ml. Pada konsentrasi 25 µg/ml fraksi mampu menginduksi apoptosis sebesar 28,9 %, kontrol sel 17,90% dan doksorubisin 63,12%. Pemacuan apoptosis ditunjukkan dengan adanya warna orange dan terjadinya banyak badan apoptosis pada sel Widr dengan perlakuan ekstrak yang lebih banyak dibandingkan dengan kontrol sel.

Abstract

Free radical in the body could cause degenerative diseases such as diabetes mellitus and hyperlipidemia, which could be prevented by the supplementation of antioxidant whether it is synthetic or natural. Flavanoids is a phenolic compound, was extracted from natural sources is known for its antioxidative potential. This research aimed to investigate the active compounds in brotowali (Tinospora crispa) stem to find new antioxidative potential using its ability to bind 2, 2’-diphenyl-1-picrylhydrazyl (DPPH). Brotowali stem was extracted using ethanol and fractioned using n-hexane, ethyl acetate, and water. Water fraction was hydrolyzed by chloric acid. The antioxidant activity of ethanol extract, water fraction, and hydrolized water fraction (subfraction I and subfraction II) were determined using spectrophotometry (DPPH methods). The compound of flavonoid was identified using KLT with AlCl3 reagent spray. The results showed that ethanol extract and water fraction of brotowali stem contained flavonoid glycoside of flavonol group. Antioxidant activity, quantified using IC50, of ethanol extract, water fraction, subfraction I and subfractiont II were 49.92μg/mL, 38.25 μg/mL, 36.12 μg/mL, and 16.18 μg/mL, respectively. In addition, this research was found that hydrolysis of water fraction using chloric acid like in subfraction I and subfraction II was able to improve its antioxidant activity.

Abstract

Background: Recently, with the high prevalence of diarrhoea caused by bacterial infection, the usage of antibiotics has increased. Antibiotic overuse might lead to several side effects and resistance, suggesting the development of an alternate antibacterial agent. A mangrove plant, Acanthus Illinois, contained triterpenoid, which has antibacterial properties. Aim: This study aimed to evaluate the antibacterial effect of the Acanthus Illinois n-hexane fraction against Escherichia coli and Shigella dysenteriae. Method: The Acanthus Illinois was fraction using n-Hexane, identification of secondary metabolite compound using GC-MS, and evaluation of antibacterial activity against Escherichia coli and Shigella dysenteriae under paper disc methods. This study was designed using a fully randomized design (CRD) with concentrations of fractions 1%, 2%, and 4%. The GC-MS results were compared to the WILEY 9 library and analysed. A prism graph was used to measure the inhibition zone of antibacterial activity. Result: The n-hexane fraction yield is 3.3% and contains sesquiterpene compounds (trans (beta.)-caryophyllene, alpha humulene, naphthalene decahedron-4A-methyl), terpenoid alcohol (3,7,11,15-tetramethyl -2-hexadecane-1-ol), and fatty acids (hexadecanoic acid methyl ester. Acanthus Illinois n-hexane fraction has antibacterial activity against Escherichia coli and Shigella dysenteriae in a dose-dependent manner. Conclusion: The n-hexane fraction of leaves Acanthus ilicifolius contains sesquiterpene, alcohol terpenoids, and fatty, and has antibacterial activity against Escherichia coli and Shigella dysenteriae

Abstract

Background: Butterfly pea (Clitoria ternatea) contains secondary metabolites, including flavonoids, saponins, terpenoids, tannins, and anthocyanins which have antioxidant activity. Objective: This research aims to produce the effervescent granule preparations of the butterfly pea flower ethanol extract with the optimal concentrations of citric acid and tartaric acid. Methods: Butterfly pea flower was extracted using 70% ethanol. Effervescent granules were made using the wet granulation method in eight formulas containing citric acid and tartaric acid. The physical properties of granules were evaluated, including extract quality, flow rate, dissolution time, and pH. Results: The concentration of the mixed components of citric acid 48.65 mg and tartaric acid 576.30 mg was the most optimal combination of acid sources for effervescent granules of butterfly pea flower extract with a desirability value of 1,000. Conclusion: The variation in the concentration of citric acid and tartaric acid affected flow rate, dissolution time, and pH of the effervescent granule preparation.

Abstract

The purpose of the analysis of the feasibility of the contents of the mathematics learning textbooks for fourth grade SD/MI students in the Mediatama issue of Angle Measurement material is to determine the accuracy and suitability in the preparation of student textbooks based on KI and KD. The method in this writing uses a qualitative approach. The data collection technique used in this research is a documentary study with a content analysis approach. This data collection was carried out to determine the condition of students' textbooks in learning mathematics for grade IV SD/MI and their suitability with K-13. This is done as an effort to improve the quality of student textbooks in grade IV SD/MI mathematics learning used by educators and fourth grade students of SD/MI. In this paper, the documentation guidelines used are based on the knowledge and skills dimensions set by BSNP. Furthermore, data collection uses a scoring technique. This technique is done by putting a checklist on the scoring sheet according to the assessment criteria. The next data analysis technique is to calculate the percentage of each aspect. The results of the analysis on mathematics learning textbooks for grade IV SD/MI on Angle Measurement material published by Mediatama are as follows: (1) The feasibility of the knowledge dimension is 96% or in the very feasible category. (2) Eligibility on the skill dimension is 91% or in the very feasible category.

Abstract

This research is motivated by the books being recalled and disturbing the world of education, not just general textbooks. However, there are also religious books that were withdrawn from circulation because they contained deviant teachings. The aim is to describe the character values in the 2013 curriculum theme 5 My Experience and the benefits of this research can be used as a reference for improving character through an integration model in elementary school subject matter and also as a reference in the improvement of subsequent books and used as a guide for book users. 2013 curriculum text. This research is a Content Analysis research with the sub-theme subject in the 2013 Curriculum Textbook, My Experience theme. As the object of research is the character values contained in the textbook. This research technique was carried out by analyzing and recording carefully the 2013 Curriculum Theme 5 My Experience textbook in the first semester of the second semester of Elementary School. The content analysis technique used in this study uses a content analysis technique consisting of six steps. The six steps are, "Unitizing, sampling, recording, reducing, inferring, narrating".

Abstract

Background: The imbalance of free radicals can be mitigated by antioxidants. Natural sources, such as the blue pea flower (Clitoria ternatea L.), have gained attention as potential antioxidant alternatives. Objective: This study aimed to evaluate the antioxidant potential of the blue pea flower by extracting its fractions using n-hexane and ethyl acetate. Methods: The study employed an experimental design involving maceration for extraction, followed by fractionation using n-hexane and ethyl acetate solvents. Antioxidant activity was assessed using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Data from both qualitative identification and antioxidant tests were analyzed descriptively and quantitatively to determine the IC50 values. Results: The analysis revealed that both the extracts and fractions of the blue pea flower contain bioactive compounds with antioxidant potential, including flavonoids, tannins, steroids, and terpenoids. The IC50 values for antioxidant activity were as follows: n-hexane fraction, 85.28 µg/mL; ethyl acetate fraction, 82.94 µg/mL; and ethanol fraction, 78.78 µg/mL. Conclusion: Flavonoids, steroids, and terpenoids were identified in both the blue pea flower extract and ethyl acetate fraction, while tannins were present only in the ethanol extract. Strong antioxidant activity was observed in all fractions, including the ethanol extract, n-hexane, and ethyl acetate fractions.

Abstract

Zingiber ottensii (bangle hantu) is a member of the Zingiberaceae family, known for its pharmacological effects. Several studies have demonstrated immunomodulatory potential of Zingiberaceae rhizomes, including Zingiber officinale, Zingiber zerumbet, and Zingiber cassumunar. This study aimed to evaluate the immunomodulatory effects of Z. ottensii extract on mouse peritoneal macrophages and lymphocytes. The extract was prepared using a maceration method with 96% ethanol (1:6) and the extract compound was observed by phytochemical screening with thin layer chromatography (TLC). Phagocytotic macrophage activity was quantified using a phagocytotic assay, which use mouse peritoneal macrophages. Lymphocyte proliferation was assessed with the MTT assay and absorbance measured at 595 nm. TLC results, showed that Z. ottensii extract tested positive for flavonoids and terpenoids. The Z. ottensii extract stimulated macrophage phagocytosis activity, significantly increasing the phagocytotic index at 25 and 50 µg/mL concentrations compared to the control. Additionally, significant lymphocyte proliferation was observed with the treatment of the extract. The Z. ottensii extract may be developed for adjuvant therapy to enhance the immune responses, offering a promising natural approach to immunomodulation.
