Abstract

Abstrak: Uji Daya Hambat Ekstrak Metanol Daging Buah Kawista (Limoniaacidissima L.) dari Daerah Kabupaten Dompu Terhadap BakteriPseudomonas aeruginosa. Tanaman kawista (Limonia acidissima L.) merupakansalah satu jenis tumbuhan obat tradisional. Masyarakat Bima Dompu cenderungmengkonsumsi buah kawista secara langsung sebagai bahan tambahan rujak dansering digunakan sebagai pengobatan diare tradisional. Daging buah Kawista(Limonia acidissima L.) diketahui mengandung flavonoid, alkaloid, dan saponin yangdapat menghambat pertumbuhan bakteri Pseudomonas aeruginosa. Tujuanpenelitian ini adalah untuk mengetahui daya hambat ekstrak metanol daging buahkawista (Limonia acidissima L.) dari daerah kabupaten Dompu terhadappertumbuhan bakteri Pseudomonas aeruginosa. Penelitian ini dilakukan dilaboratorium Riset Fakultas Kedokteran Universitas Islam Al-Azhar menggunakanmetode ekstraksi maserasi dengan pelarut metanol. Rancangan yang digunakandalam penelitian ini adalah Rancangan Acak Lengkap (RAL) dengan 4 kelompokperlakukan dan 2 kontrol (kontrol positif kloramfenikol dan kontrol negatifaqudest). Data yang didapatkan dianalisis menggunakan SPSS versi 23. Hasil daripenelitian ini didapatkan diameter zona hambat pada 4 kelompok perlakuan. Dayahambat terbesar adalah 41,50 mm pada konsentrasi 100%, dan daya hambat  terendah adalah 18,5 mm pada konsentrasi 25%. Berdasarkan hasil analisis yang dilakukan, didapatkan perbedaan yang bermakna antar kelompok perlakuan dengan nilai p<0.05. Ekstrak metanol daging buah kawista (Limonia acidissima L.) dari daerah kabupaten Dompu memiliki daya hambat terhadap pertumbuhan bakteri pseudomonas aeruginosa.

Abstract

Salmonella typhi is a gram-negative bacterium that is an intracellular facultative pathogen, which enters the human body and causes an acute systemic infectious disease called typhoid fever. Among the laboratory tests used to detect the presence of anti-Salmonella typhi IgM in serum are SD Bioline and Tubex TF. This study aims to determinecomparison of the results of the anti-Salmonella typhi IgM examination with the SD Bioline and Tubex TF methods. Analytical observation with cross-sectional design. This research was conducted in RSUD DR. Soedjono Selong. The time of the study was January-April 2022. The sample of this study was patients who underwent typhoid fever examination who were hospitalized at RSUD DR. Soedjono Selong as many as 60 people. This research uses the methodconsecutive sampling,Anti-Salmonella typhi IgM was examined with the SD Bioline kit using the immunochromatography method and the TUBEX TF kit using the Inhibition Magnetic Binding Immunoassay (IMBI) method. Data analysis was performed using SPSS, namely univariate test, and bivariate test with Chi-square. There were 26 (43.3%) male and 34 (56.7%) female patients. The youngest is 2 years old and the oldest is 64 years old with an average age of 25 years ± 19 years. Of the total 60 patients examined with SD Bioline, 5 (8.3%) were positive and 55 (91.7%) were negative. Meanwhile, examination with TUBEX TF found 3 (5%) positive people and 57 (95%) negative people. And there was a significant comparison between the results of the SD Bioline and Tubex TF examination with a value (p = 0.000) and a low level of conformity was obtained with a Kappa value of 0.000. A comparison of the results of the IgM anti-Salmonella typhi examination was obtained using the SD Bioline and Tubex TF . methodswith a very low degree of conformity.

Abstract

ABSTRACT The development of natural materials, in this case phytochemical compounds from herbal plants, is utilized as antibiotic agents in dealing with several cases of antibiotic resistance. Arum manis mango plant (Mangifera indica L. var arum manis) is one of the species of the mango family that is widely distributed in Indonesia and has many benefits ranging from fruit, seeds, fruit skin, leaves and stems. Arum manis mango leaves contain flavonoids, alkaloids, steroids, polyphenols, tannins, and saponins which have the ability to inhibit bacterial growth. To determine the antibacterial activity of ethanol extract of arum manis mango leaves on the growth of Staphylococcus epidermidis bacteria. True Experiment with Posttest Only Control Group Design using the well method. The research subjects amounted to 24 samples divided into 4 treatment groups, namely ethanol extract of arum manis mango leaves with concentrations of 25%, 50%, 75%, 100% and 2 controls (positive control ciprofloxacin and negative control aquadest). The data obtained was analyzed using SPSS version 27. The results of the bivariate test showed a significant difference (p-value 0.05) in the inhibition zone of the treatment group with positive and negative controls (Staphylococcus epidermidis, p-value 0.001). The average inhibition zone formed in the ethanol extract group of arum manis mango leaves on Staphylococcus epidermidis is concentrations of 25% (15.87 mm); 50% (18.25 mm); 75% (19.62 mm) and 100% (20.37 mm). There is antibacterial activity of the ethanol extract of arum manis mango leaves (Mangifera indica L. var arum manis) against the growth of Staphylococcus epidermidis bacteria. Keywords: Antibacterial, Arum Manis Mango Leaves, Staphylococcus Epidermidis, Wells  ABSTRAK Pengembangan bahan alam dalam hal ini senyawa fitokimia dari tanaman herbal dimanfaatkan sebagai agen antibiotik dalam menangani beberapa kasus resistensi antibiotik. Tanaman mangga arum manis (Mangifera indica L. var arum manis) merupakan salah satu spesies dari famili buah mangga yang banyak tersebar di wilayah Indonesia dan memiliki banyak manfaat mulai dari buah, biji, kulit buah, daun dan batangnya. Daun mangga arum manis mengandung senyawa flavonoid, alkaloid, steroid, polifenol, tannin, dan saponin yang memiliki kemampuan menghambat pertumbuhan bakteri. Mengetahui aktivitas antibakteri ekstrak etanol daun mangga arum manis terhadap pertumbuhan bakteri Staphylococcus epidermidis. True Experiment dengan Posttest Only Control Group Designmenggunakan metode sumuran. Subjek penelitian berjumlah 24 sampel dibagi ke 4 kelompok perlakuan, yaitu ekstrak etanol daun mangga arum manis konsentrasi 25%, 50%, 75%,100% dan 2 kontrol (kontrol positif ciprofloxacin dan kontrol negatif aquadest). Data yang didapatkan dianalisis menggunakan SPSS versi 27. Hasil dari uji analisis data menunjukan terdapatnya perbedaan bermakna (p-value 0,05) zona hambat kelompok perlakuan dengan kontrol positif dan negatif (Staphylococcus epidermidis, p-value 0,001). Rata-rata zona hambat yang terbentuk pada kelompok ekstrak etanol daun mangga arum manis pada Staphylococcus epidermidis konsentrasi 25% (15,87 mm); 50% (18,25 mm); 75% (19,62 mm) dan 100% (20,37 mm).
