Abstract

The developed Leydig cells-conditioned medium (LCM) contains bioactive materials secreted by Leydig cells in vitro.  LCM was used to evaluate the ability of bone marrow mesenchymal stem cells differentiation. Bone marrow mesenchymal stem cells (1x 106 cell/ml) were cultured in : 1) DMEM supplemented with 10% NBCS as a control (M), 2) M supplemented with 10 ng/ml testosterone; 3) M supplemented with 50%  LCM ; 4) M supplemented with 50% LCM and 2.5 IU/ml hCG. Bone marrow mesenchymal stem cells that were cultured with  LCM has a positive reaction (57.4%) to histochemistry staining 3β-HSD and produced 1.87 ng/ml testosterone. Supplementation of hCG to LCM  increased the positive number of Leydig cells and testosterone production by 74.6% and 12.33 ng/ml (P<0.05). It can be concluded that Leydig cells-conditioned medium can support differentiation of bone marrow mesenchymal stem cells into Leydig cells.

Abstract

This research aims to evaluate the protective potency of freeze-dried egg yolk powder on Garut ram sperm during the liquid semen preservation process. Semen with good sperm quality was divided into three groups and diluted using the following Tris diluents: fresh egg yolk-Tris (FEY-Tris), commercial egg yolk powder-Tris (CEY-Tris), and freeze-dried egg yolk powder-Tris (DEY-Tris). Semen that had been diluted was observed every 12 hours until sperm progressive motility was 50%. Sperm quality was tested for progressive motility percentages, viability, intact plasma membrane (IPM), and intact acrosome (IA). Fresh egg yolk had a particle size of 14460.00330.76 nm, with polydispersity index value (PI) and zeta potential (ZP) being 1.000.11 and -9.220.30 mV, respectively. The particle size of commercial egg yolk powder was 877.90168.86 nm with PI value 0.340.04 and ZP-28.71.24 mV. The particle size of freeze-dried egg yolk powder was 1296.0086.73 nm, with PI value 0.700.04 and ZP-34.50.64 mV. Progressive motility percentage showed that DEY-Tris diluent managed to survive to 51.116.06% for 168 hours, compared to FEY-Tris (51.256.74%) which survived for 156 hours, although no longer than CEY-Tris diluent (53.476.33%) which survive for 180 hours (P0.05). Higher sperm motility was also supported by viability percentage, IPM, and IA, which were all higher (P0.05). In conclusion, freeze dried egg yolk powder mixed with Tris buffer was able to maintain sperm quality during the liquid semen preservation process.

Abstract

ABSTRACTThe objective of this research was to evaluate the quality of chilled canine semen in Tris-egg yolk (TEY) extenders containing different concentrations of sericin. Semen were collected from four dogs by massage method. Canine semen was collected using sterile urine pots and evaluated. Sperm-rich fractions were pooled and divided into four equal aliquots, which were then diluted with TEY extenders supplemented with different concentrations of sericin (0%, 0.1%, 0.25%, 0.5%). The diluted semen aliquots were preserved at 4 in sterile centrifuge tubes and were then evaluated for spermatozoa motility, viability, plasma membrane integrity and acrosome integrity every 12 hours up to 72 h. The TEY extenders supplemented with 0.25% and 0.5% sericin resulted in higher spermatozoa motility and viability at 72 h compared to other TEY extenders (P0.05). The integrity of plasma membrane and acrosome of spermatozoa showed no significant differences among the groups extenders at 72 h. In conclusion, sericin in concentration of 0.25% and 0.5% were able to prevent the motility and viability of canine spermatozoa after storage for 72 h.

Abstract

The aims of this study were to investigate the freezing capability of Pasundan bull spermatozoa in Tris-egg yolk (TEY), Tris-soy (TS), and AndroMed as diluents. Semen were collected twice a week from four Pasundan bulls aged 3-5 years old using an artificial vagina and evaluated macro- and microscopically. Semen had 70% sperm motility, 800x106/mL sperm concentration, and less than 20% sperm abnormalities were divided into three parts and each of them diluted with TEY, TS, or AndroMed. After an equilibration step at 5C for four hours, diluted semen were packaged in 0.25 mL straw, frozen in liquid nitrogen for ten minutes and kept in liquid nitrogen container until examination. Motility test on fresh, diluted, equilibrated, and after-thawed semen was done using Androvision. The results showed that after thawing motility of sperm diluted in AndroMed (58.640.72%) was higher than in TEY (49.451.22%) and TS (39.346.33%). Sperm motility of Pasundan bulls diluted in these three diluents reduced around 33.272.45% during freezing process.

Abstract

This research was aimed to study the effect of administration of glycerol and dimetilacetamida (DMA) in BTS and MIIIextender supplemented with sodium dodecyl sulphate (SDS) on boar frozen semen. A number of four boars were used in this study for semen collection (n=20). The collected semen was evaluated both macroscopically and microscopically. In this study, only the semen that demonstrated70% sperm motility, 200.106/mL sperm concentration, and20% sperm abnormalities were used and divided into eight tubes. A number of 4 tubes were diluted with 5 mL of BTS, and the rest with 5 mL MIII. The sampel was stored at 20-22 C for 2 hours, followed by centrifugation for 15 minutes (at 2000 rpm), and taken of pellet with 1 ml supernatant. The pellet that was resulted from centrifugation using BTS, then re-diluted with BTS-glycerol 5% (BTSG), BTS DMA 5% (BTSD), BTS-glycerol 5% and SDS (BTSG-S), BTS-DMA 5% and SDS (BTSD-S). Four other pellets that were centrifuged with MIII also re-diluted with MIII-glycerol 5% (MIIIG), MIII-DMA 5% (MIIID), MIII-glycerol 5% and SDS (MIIIG-S), MIII-DMA 5% and SDS (MIIID-S). Next, all of diluted semen were inserted into 0.5 mL straw and equilibrated for 2 hours (4 C), then frozen and stored in liquid nitrogen. The evaluation of frozen semen quality was conducted at 24 hours after frozen. The result of this study showed that post-thawing motility of spermatozoa in BTSD-S (40.170.2%) was found higher (P0.05) compared to seven other dilution processes. Therefore, it is concluded that the concentration of 5% DMA that supplemented with SDS in BTS dilution much better for maintaining boars frozen semen quality.

Abstract

Penelitian ini bertujuan menggambarkan morfologi pertumbuhan ranggah velvet (RV) dua ekor muncak jantan (M1 dan M2) yang dipelihara di kandang penelitian secara terpisah. Tanggal lepas ranggah dan hari pertama percabangan RV serta awal pengelupasan kulit velvet diamati dan dicatat. Pengukuran meliputi panjang dan diameter ranggah velvet utama (RVU) dan ranggah velvet cabang (RVC) dexter et sinister, serta durasi pertumbuhan RV. Rataan SD panjang dan diameter maksimum RVU dan RVC muncak M1 adalah 148,251,12 mm; 46,260,68 mm (panjang), 19,212,35 mm; 9,120,21 mm (diameter) dengan durasi pertumbuhan 104 hari; sedangkan muncak M2 berturut-turut adalah: 146,441,58 mm; 46,941,97 mm (panjang); 13,651,27 mm; 8,490,29 mm (diameter) dengan durasi pertumbuhan 98 hari. Tidak terdapat perbedaan tahapan pertumbuhan RVU dan RVC antara muncak M2 dan M1, namun berbeda pada ukuran RV dan durasi pertumbuhannya. Muncak M2 memasuki ranggah keras setelah pengelupasan kulit velvet lebih cepat dibandingkan M1. Dapat disimpulkan bahwa ukuran pertumbuhan RV berkorelasi erat dengan faktor umur, berat badan, dan postur tubuh muncak jantan.
