Abstract

The recent research concerned in method development for determination of nitric oxide metabolites in serum using spectrophotometry visible based on Griess reaction. The reduction of nitrate to nitrite was performed using copper coated zinc. The method was optimized and validated. The determination of total nitrite in samples was started with deproteinization using ZnSO4 and NaOH. The supernatants were reducted using copper coated zinc, reacted with diazotization and coupling reagents, then incubated. The absorbance was measured using spectrophotometer visible. The optimum reductant mass of copper coated zinc is 1.5 g with reduction time 10 minutes, coefficient variation ? 5%. Linierity of analytical method gives a coefficient correlation, r > 0.995. Relative procedural standard deviation (Vx0) was ? 5%, precision ? 15%, and recovery 85-115%.

Abstract

Cases of psychotropic abuse have occurred in Indonesia. Drug abuse or detrimental use is the use of psychotropic without medical supervision. The case of drug abuse is a case that requires forensic toxicology examination. Forensic toxicology examination consists of the stages of screening tests, confirmation tests, and interpretation of results. This study aims to determine the psychotropic compounds contained in urine samples. The screening test method used is the immunoassay technique using a strip test, the results obtained that urine samples show positive results on the barbiturate group test strips. Confirmation test method is done by TLC-Spectrophotodensitometry method, the results obtained that the barbiturate class compounds contained in the sample are phenobarbital compounds. Furthermore, a determination test was performed and the results were obtained that the levels of phenobarbital in urine in a 4 µl drop volume with replication 3 times were 22.97 ng / µl, 34.66 ng / µl, and 22.42 ng / µl.

Abstract

Dichlorvos and phenthoat are organophosphate group pesticides which have toxic effects on humans and can cause fatal deaths. Suicide cases using organophosphate pesticides (dichlorvos and phenthoat) are one of the postmortem toxicology analysis studies. As a forensic toxicologist, competency is needed to be able to handle suicide deaths that are thought to be caused by dichlorvos and phenthoat. Handling suicides due to dichlorvos and phenthoate is done from sample preparation, screening tests, assurance tests, determination, and data interpretation. Forensic toxicology analysis in the case showed that high concentrations of dichlorvos and phenthoate were found to have toxic effects on the victim's body. Based on the toxicological analysis, it can be concluded that it is true that the victim died from organophosphate poisoning (dichlorvos and phenthoat).

Abstract

ABSTRAK Zat pewarna sintetis digunakan karena komposisinya lebih stabil dan relatif lebih murah. Contoh pewarna sintetis adalah sunset yellow FCF yang memberi warna oranye, karmoisin yang memberikan warna merah, serta Tartrazine untuk warna kuning. Zat warna yang dilarang digunakan dalam pangan tercantum dalam Peraturan Menteri Kesehatan RI Nomor 239/MenKes/Per/V/85 mengenai Zat Warna Tertentu yang Dinyatakan sebagai Bahan Berbahaya. Salah satu zat warna yang dinyatakan bahan berbahaya adalah rodamin B. Rodamin B masih banyak ditemukan pada produk kosmetik dan juga berbagai komoditas pangan. Paparan jangka panjang dengan rodamin B dapat menyebabkan kanker dan resiko cacat kelahiran. Oleh karena itu akan dikembangkan metode analisis kromatografi lapis tipis (KLT) untuk identifikasi dan penetapan kadar rodamin B di produk jajanan tradisional. Dikembangkan enam fase gerak untuk memisahkan senyawa rodamin B dari pewarna lain. Dari hasil yang diperoleh,fase gerak 6 (Etanol:air:asam asetat:dietilamin (95:5:5:1) adalah fase gerak yang paling mampu memisahkan beberapa pewarna sekaligus. Uji kualitatif dengan KLT-spektrofotodensitometri didapatkan bahwa sampel yang positif mengandung rodamin B adalah sampel KM3 dan KM 5. Hal ini disimpulkan drai Rf dan spectrum yang sama dengan rodamin, Yaitu Rf 0,72. Persamaan garis regresi yang diperoleh adalah y = 564,534 + 1,799x dan r = 0,9946 dengan rentang linieritas 200-1500 ppm. Pada penetapan kadar sampel yang mengandung rodamin B, yaitu pada sampel KM3 dan KM B, secara berturut-turut kadar yang diperoleh adalah 0,676 mg/1 gram sampel dan 2,751 mg/1 gram sampel

Abstract

Opiates are a group of narcotics that are often abused. Forensic experts play crucial roles in handling this. The screening test results that the sample containing opiate compounds. Confirmation testing is carrie out by using TE and TAE mobile phase systems. The error window values ??obtained in the TE and TAE systems are 10 and 8. Data collection of compounds in the range of hRfc 16.09 to 36.09 in the TE system obtained two compounds, Morphine and Codeine). And in the range of hRfc 25.1-9.1 also found Morphine and Codein compounds. The results of the correlation spectrum of the sample and spectrum in the library showed a correlation value of 0.99079 (TE system) and 0.99477 (TAE system) to the codeine compound. Determination testing is carried out using the TE mobile phase. This test results obtained that the concentration of codeine compounds is 20.88 ng / µL. These results indicate that the sample contains codeine in the lethal dose range.

Abstract

Mephedrone or 4-Methylmethcathinone is one type of NPS (New Psychoactive Substance) which is included in the katinon derivative. Mephedrone has been banned in Indonesia and in other countries such as the Netherlands. Mephedrone is used as a recrerational drug that can increase enthusiasm for its users. Misuse of mephedrone can cause toxicity to death for its users. In reviewing this article we will summarize the cases of mephedrone abuse in Indonesia, the pathway for mephedrone synthesis, the pharmacological, pharmacokinetic and pharmacodynamic effects of mephedrone and forensic toxicology analysis on cases of misuse of mephedrone compounds.

Abstract

A current research of screening and determination of codeine using Al-TLC Spectrophotodensitometry has been carried out. The separation was performed using TAEA mobile phase. Screening and confirmation were investigated by comparing hRfc of the analyte and hRfc those on data library. Combination hRfc vs correlation also been used as confirmation method, which can be enhanced by spectrum shift reaction using HCl 10% and KOH 0,1 M. The validation parameters were precission, linierity, limit of detection and quantitation (LOD & LOQ) and recovery.   TAEA mobile phase performed a good separation among morphine, codeine, caffeine, papaverine and bromhexine standards, which gave Rs value > 1. Test of linierity using regression gave correlation value > 0,99 . LOD and LOQ of Al-TLC were 137.85 ng and 459.63 ng. The recovery of codeine in human plasma using isopropanol as the precipitating agent was 77.1% ± 4.9 (6.5%).

Abstract

Virgin coconut oil (VCO) dan madu telah banyak digunakan sebagai bahan baku sediaan topikal untuk perawatan kulit. Salah satu khasiat yang telah terbukti adalah aktivitas antioksidannya. Namun efek VCO dan madu sebagai antioksidan umumnya masih dibuktikan dalam bentuk bahan tunggal tidak dalam bentuk kombinasi keduanya. Pengujian aktivitas antioksidan dengan metode DPPH memerlukan sampel uji yang bersifat polar. Fraksi metanol dari VCO dan madu telah diketahui bersifat polar. Tujuan penelitian ini adalah menentukan kombinasi fraksi metanol VCO dan madu kele Bali yang memberikan aktivitas antioksidan paling kuat. VCO dan madu kele Bali diperoleh dari Desa Aan, Bali. Skrining fitokimia fraksi metanol VCO dan madu kele Bali ditentukan ditentukan dengan metode yang sesuai. Aktivitas antioksidan (IC50) fraksi metanol VCO, madu kele Bali, dan kombinasi keduanya ditentukan dengan metode DPPH. Terdapat 5 kombinasi kedua bahan tersebut yang diuji. Data dianalisis menggunakan ANOVA oneway-LSD dengan taraf kepercayaan 95%. Fraksi metanol VCO mengandung flavonoid dan tanin, sedangkan madu kele Bali mengandung saponin, fenol, flavonoid dan steroid/triterpenoid. Fraksi metanol VCO dan madu kele Bali memiliki aktivitas antioksidan dengan nilai IC50 berturut-turut sebesar 7,20±0,23 mg/mL dan 14,56±0,25 mg/mL. Perbandingan jumlah kombinasi fraksi metanol VCO dan madu kele Bali berpengaruh secara bermakna terhadap aktivitas antioksidannya (p<0,05). Kombinasi 0,0063 g fraksi metanol VCO dan 1 g madu kele Bali menunjukkan aktivitas antioksidan paling kuat (IC50 8,50±0,17 mg/mL). Fraksi metanol VCO dan madu kele Bali serta kombinasinya tergolong antioksidan lemah, namun terjadi peningkatan aktivitas antioksidan yang bermakna pada kombinasi ini dibandingkan madu kele Bali tunggal (p<0,05).

Abstract

Abstrak Reaksi oksidasi yang berlebihan pada tubuh dapat menyebabkan terbentuknya radikal bebas yang sangat aktif, sehingga merusak struktur dan fungsi sel tubuh. Antioksidan dapat menstabilkan radikal bebas dengan melengkapi kekurangan elektron yang dimiliki radikal bebas serta dapat menghambat terjadinya reaksi berantai dari pembentukan radikal bebas. Herba suruhan (Peperomia pellucida (L.) Kunth) mengandung senyawa antioksidan seperti flavonoid, fenol, serta tanin yang berpotensi sebagai antioksidan. Review artikel ini bertujuan untuk mengetahui nilai total fenol dan flavonoid ekstrak herba suruhan serta mengetahui nilai aktivitas antioksidan ekstrak herba suruhan (Peperomia pellucida (L.) Kunth) berdasarkan pelarut, metode ekstraksi, serta metode pengujian aktivitas antioksidan. Review artikel ini dibuat dengan metode studi literatur dengan menggunakan artikel penelitian terdahulu yang diperoleh melalui database online yang memenuhi kriteria inklusi. Hasil menunjukkan bahwa aktivitas antioksidan ekstrak herba suruhan memiliki perbedaan hasil berdasarkan perbedaan jenis pelarut, metode ekstraksi, serta metode pengujian aktivitas antioksidan yang digunakan. Penelitian lebih lanjut dengan variasi parameter pengujian serta faktor-faktor yang berpengaruh terhadap hasil disarankan dilakukan untuk mendapatkan hasil yang lebih spesifik dan jelas.
