Abstract

The sponge is one of the sea organisms that has a prospect as a source of natural compounds including peptides, steroids, asetogenin, terpenoids, alkaloids, cyclic halide and nitrogen. This research was directed to obtain several species of sponges from the waters of Tasik Ria as well as testing the antibacterial activity of extracts from some of the sponge against the bacteria Escherichia coli and Staphylococcus aureus. From the identification, seven species of sponges were found, which consists of: Amphimedon sp., Axinosa sp., Aaptos sp., Theonella sp., Cribochalina sp., Hyrtios sp., and Lendenfeldia sp. The tests of antibacterial activity of the extracts from these sponges against test bacteria E. coli and S. aureus showed some positive results. Extract from Axinosa sp. sponge(16 mm) showed the strongest antibacterial activity on Escherichia coli bacteria. Followed by Hyrtios sp. extract (13.5 mm), Aaptos sp. extract (13 mm), Lendenfeldia sp. extract (13 mm) and Cribochalinai sp. extract(10.5 mm). While the the tests on Staphylococcus aureus bacteria showed that the strongest antibacterial activity was found from Axinosa sp. sponge extract (16.5 mm), followed by the extract from Aaptos sp. (15 mm), Lendenfeldia sp. extract (14.5 mm), Hyrtios sp. extract(13.5 mm) and Cribochalina sp. extract (11 mm).Keywords: Sponge, antibacterial, Escherichia coli, Staphylococcus aureus ABSTRAK Spons merupakan salah satu biota laut yang sangat prospektif sebagai sumber senyawa bahan-bahan alami antara lain peptide, terpenoid, steroid, asetogenin, alkaloid, halide siklik dan senyawa nitrogen. Penelitian ini diarahkan untuk mendapatkan beberapa spesies spons dari perairan Tasik Ria serta menguji aktivitas antibakteri dari beberapa ekstrak spons terhadap bakteri Escherichia coli dan bakteri Staphylococcus aureus. Hasil identifikasi spons ditemukan sebanyak tujuh spesies yang terdiri dari: Amphimedon sp., Axinosa sp., Aaptos sp., Theonella sp., Hyrtios sp., Cribochalina sp. dan Lendenfeldia sp.. Aktivitas antibakteri dari beberapa ekstrak spons terhadap bakteri uji E. coli dan S. aureus terdapat diameter zona hambat bervariasi yaitu bakteri Escherichia coli menunjukkan aktivitas antibakteri ekstrak spons terkuat pada spons Axinosa sp (16 mm), disusul ekstrak spons Hyrtios sp. (13,5 mm), ekstrak spons Aaptos sp. (13 mm), ekstrak spons Lendenfeldia sp. (13 mm) dan ekstrak spons Cribochalinai sp. (10,5 mm).  Sedangkan pada bakteri Staphylococcus aureus menunjukkan aktivitas antibakteri ekstrak spons terkuat yaitu:  ekstrak spons Axinosa sp. (16,5 mm), disusul ekstrak spons Aaptos sp. (15 mm), ekstrak spons Lendenfeldia sp. (14,5 mm), ekstrak spons Hyrtios sp. Abstract

This study aims to obtain isolates and amplify the associative bacterial 16SrRNA gene in K. alvarezii algae. The K. alvarezii algae was collected from seaweed cultivation area in Belang, Southeast Minahasa, North Sulawesi. Associative bacteria were sampled from K. alvarezii algae, grown in Nutrient Agar and separated based on their morphological characteristics.  Each isolates were extracted their DNA genome and.the16S rRNA gene of each isolate was amplified using PCR.  Eight associative bacterial from K. alvarezii algae were successfully isolated based on morphological characteristics which were dominated by round shape, smooth edges, convex elevation, and white color of the isolates. The results of genomic DNA extraction from each of these isolates were successfully used as templates to amplify the 16s rRNA gene.Key word: Bacteria, Kappaphycus alvarezii, Taxsonomy, 16S rRNAABSTRAKPenelitian ini bertujuan untuk mendapatkan isolat dan mengamplifikasi gen 16SrRNA bakteri asosiatif pada alga K. alvarezii. Metode penelitian diawali dengan pengambilan sampel alga K. alvarezii di lokasi pembudidayaan rumput laut di desa Belang, Minahasa Tenggara, Sulawesi Utara. Selanjutnya, bakteri asosiatif pada alga K. alvarezii tersebut ditumbuhkan dalam media Nutrient Agar, isolat yang tumbuh kemudian dipisahkan berdasarkan karakteristik morfologinya. dan gen 16S rRNA masing-masing isolat diamplifikasi menggunakn PCR.  Delapan isolat bakteri asosiatif pada alga K. alvarezii berhasil diisolasi dengan karakteristik morfologi berbeda yang didominasi dengan bentuk round, tepian smooth, elevasi convex, dan warna putih. Hasil ekstraksi DNA genom dari masing-masing isolat tersebut berhasil digunakan sebagai template untuk mengamplifikasi gen 16s rRNA.Kata Kunci: Bakteri, Kappaphycus alvarezii, Taksonomi, 16S rRNA

Abstract

The objective of this research was to obtain probiotic bacteria isolated from the digestive tract of catfish Clarias batracus, to evaluate its effect on growth performances and survival rate of carp. After purified, the bacteria was identified through biochemical tests and selection was carried out through proteolytic, amylolytic, cellulolytic, tests as well as pathogenicity test. Probiotic candidate was administered orally to fish at different concentrations. The fish were fed for 30 days respectively as much as 3% of body weight per day with feeding frequency of two times a day. The results showed that addition of probiotics into feed significantly increased growth performance but not survival rate.Keywords: Probiotic, Clarias batracus,  growth performance, aquaculture. ABSTRAKTujuan dari penelitian ini adalah untuk mendapatkan bakteri probiotik yang diisolasi dari saluran pencernaan ikan lele Clarias batracus, dan mengevaluasi pengaruhnya terhadap kinerja pertumbuhan dan kelangsungan hidup benih ikan mas. Bakteri probiotik diidentifikasi melalui uji biokimia dan seleksi dilakukan melalui proteolitik, amilolitik, tes selulolitik dan patogenisitas. Kandidat probiotik diberikan pada ikan secara oral dengan konsentrasi berbeda Pemberian pakan berprobiotik berlangsung selama 30 hari sebanyak  3% dari berat badan per hari dengan frekuensi pemberian dua kali sehari. Hasil penelitian menunjukkan bahwa penambahan probiotik ke dalam pakan secara signifikan meningkatkan kinerja pertumbuhan namun tidak memengaruhi kelangsungan hidup benih ikan mas.Kata kunci : Probiotik, Clarias batracus, kinerja pertumbuhan, budidaya

Abstract

This study aims to examine the effect of sponge crude extract addition to the fish feed on growth and non-specific immune response of nile tilapia (O. niloticus) and determine the most effective dose of the extract in raising the growth and the non-specific immune response of nile tilapia (O. niloticus). The sponge sample was macerated and then evaporated using a rotary vacuum evaporator until the sample became a paste and mixed with feed. The fish were fed for 14 days respectively as much as 5% of body weight per day with feeding frequency of two times a day. The results showed that addition of sponge crude extract to the fish feed gave good effect on the growth and the non-specific immune response of nile tilapia (O. niloticus) with best dose of  40 g / kg feed.Keywords: Sponge extract, fish immune response, fish growth, immune response, immunostimulant. ABSTRAKPenelitian ini bertujuan untuk menguji pengaruh penambahan ekstrak kasar spons pada pakan ikan terhadap pertumbuhan dan respon imun  non spesifik ikan nila (Oreochromis niloticus) serta menentukan dosis ekstrak kasar spons yang paling efektif dalam meningkatkan pertumbuhan dan respon imun non spesifik ikan nila (Oreochromis niloticus). Sampel spons dimaserasi kemudian dievaporasi menggunakan rotary vacuum evaporator sehingga diperoleh ekstrak kasar berbentuk pasta yang kemudian dicampurkan pada pakan. Ikan uji diberi pakan perlakuan 5% dari bobot biomassa selama 14 hari dengan frekuensi pemberian pakan dua kali sehari. Hasil penelitian menunjukan bahwa penambahan ekstrak kasar spons pada pakan ikan memberikan pengaruh yang baik terhadap pertumbuhan dan respon imun non spesifik ikan nila (Oreochromis niloticus), yang mana dosis yang terbaik adalah 40 gr / Kg pakan.Kata Kunci : Ekstrak spons, respon imun ikan, pertumbuhan ikan, respon imun, imunostimulan.

Abstract

The marine sponge collected from tropical coral reefs in Manado North Sulawesi Indonesia was screened for antimicrobial activities. In the screening program to search for antituberculotic inhibitors, the result found that the ethanol extract ODS fractions 4 – 6 with concentration 5 mg/mL of an Indonesian marine sponge Auletta sp. was exhibited inhibitory activity against nonpathogenic Mycobacterium smegmatis at concentration 10, 20, 30µg/disc each fraction respectively. Fractions 4 - 5 with concentration 10µg/disc were most active, the inhibition zone 11, 12 mm, respectively.Keywords:  marine sponge, antimicrobial, antituberculosis, Mycobacterium smegmatis

Abstract

Soft corals ar known to have a variety of bioactivity capabilities, one of them is antibacterial. Antibacterial is a compound that can interfere with the growth of bacteria or even kill the bacteria. The soft coral samples, Lobophytum sp., obtained from  Pangalisang, Bunaken Island, wer extracted by evaporative process, then fractionated into three fractions by aqueous partitioning technique with aqueous solvents, ethyl acetate, methanol and n-hexane. Antibacterial testing, with bacterial samples of Pseudomonas aeruginaso and Staphylococcus aureus were analyzed by agar diffusion / Kirby Bauer method. The results of the experiment showed that every fraction had antibacterial activity, where the largest activity was produce at concentration of 50 disc paper sampes with  7 mm (water fraction), 8 mm ( metanol fraction), 7,2 mm (n-hexane fraction) in Pseudomonas aeruginosa and 7 mm (water fraction), 8,7 mm ( metanol fraction) in Staphylcoccus aureus. It can be concluded that the  inhibition zone from soft corals, Lobophytum sp., is classified as a weak activity, so it does not potential to be developed into an antibacterial drug.Keywords: Antibacterial, Lobophytum sp., Pseudomonas aeruginosa, Staphylcoccus aureus.ABSTRAKKarang lunak diketahui mempunyai berbagai bioaktivitas salah satunya antibakteri. Antibakteri merupakan senyawa yang dapat mengganggu pertumbuhan atau bahkan dapat mematikan bakteri. Sampel karang lunak Lobophytum sp. yang diperoleh dari perairan Pangalisang Pulau Bunaken diambil ekstrak kentalnya melalui proses evaporasi, selanjutnya difraksinasi menjadi tiga fraksi dengan teknik partisi cair dengan pelarut aquades, etil asetat, metanol dan n-heksan. Pengujian antibakteri terhadap bakteri Pseudomonas aeruginosa  dan Staphylococcus aureus dianalisis dengan metode difusi agar (disc diffusion Kirby and Bauer). Hasil penelitian menunjukan ketiga fraksi memiliki aktivitas antibakteri, dimana aktivitas terbesar dihasilkan pada konsentrasi sampel dalam kertas cakram 50 μl dengan zona hambat 7mm (fraksi air), 8 mm (fraksi metanol), 7,2 mm (fraksi n-heksan) pada Pseudomonas aeruginosa dan 7 mm (fraksi air), 8,7 mm (fraksi metanol) pada Staphylococcuss aureus. Dapat disimpulkan bahwa zona hambat yang dihasilkan karang lunak Lobophytum sp. tergolong lemah sehingga tidak berpotensi untuk dikembangkan sebagai obat antibakteri.Kata kunci: Antibakteri, Lobophytum sp., Pseudomonas aeruginosa, Staphylcoccus aureus.

Abstract

Sponge samples collected around Manado waters were obtained 30 species and their crude extracted have been tested in vitro for their activity in inhibiting bacterial growth. Based on the results of antibacterial screening in 30 sponge extracts, there were 23 sponge extracts which had bioactivity in inhibiting the growth of S. aureus, E. coli, S. saprophyticus and P. aeruginosa, Sponge extract No. 43 (of 30 sponge extracts tested) was the most active in inhibiting bacterial growth and had the widest inhibition zone diameter.Keywords: screening, sponge, crude extract, antibacterial, ManadoABSTRAKSampel spons dikoleksi di sekitar Perairan Manado sebanyak 30 jenis/spesies, dimana ekstrak kasarnya telah diuji secara in vitro aktivitasnya dalam menghambat pertumbuhan bakteri. Abstract

This study aims to analyze the relation of marine debris to gastropods in the mangrove ecosystem. This research was conducted from October to December 2019 in the Tongkaina mangrove forest. The method used in this research is the line transects method with 2 different stations. Then do the data analysis of waste density, gastropod abundance index, frequency, diversity index, uniformity, and dominance. Then, analyzing a simple linear regression and correlation is performed to see the relationship between marine debris and gastropod abundance in a mangrove ecosystem. The results showed that the highest inorganic marine debris in the Tongkeina mangrove ecosystem is plastic and followed by cloth, wood, metal and the least was glass. The species of Gastropods found in the mangrove ecosystem are Littoraria scabra, Nerita undulata and Terebralia sulcata. The highest abundance of gastropod species was found at station 2 in transects 1 by Littoraria scraba. The results of the linear regression analysis show that there is a relationship between waste and abundance of gastropods. The increasing amount of waste, lower the abundance of gastropods. Where the correlation value is -0.20506.Keywords: Correlation; Marine debris; Gastropods; Mangrove Ecosystem. ABSTRAKPenelitian ini bertujuan untuk menganalisis  hubungan  sampah laut terhadap gastropoda di ekosistem mangrove. Penelitian ini dilakukan pada bulan Oktober sampai Desember 2019 di hutan mangrove Tongkaina.  Metode yang digunakan adalah metode line transek dengan 2 stasiun berbeda. Kemudian dilakukan analisa data kepadatan sampah, indeks kelimpaha gastropoda, frekuensi, indeks keanekaragaman, keseragaman dan dominasi. Selanjutnya dilakukan analisa regresi lineier sederhana serta korelasi untuk melihat hubungan yang terjadi antara sampah dan kelimpahan gastrooda. Hasil penelitian menunjukkan bahwa sampah anorganik jenis plasik yang tertinggi di ekosistem mangrove pantai Tongkaina diikuti sampah jenis Kain, Kayu, Logam dan yang paling sedikit adalah sampah jenis kaca. Jenis gastropoda yang ditemukan di ekosistem mangrove yaitu Littoraria scabra, Nerita undulata dan Terebralia sulcata. Nilai kelimpahan jenis gastropoda tertinggi terdapat di stasiun 2 di transek 1 oleh Littoraria scraba. Hasil analisa regresi linier menunjukan bahawa ada hubungan yang terjadi  antara sampah dan kelimpahan gastropoda yang dilihat dari anlisis regresi linier sederhana. Semakin meningkatnya jumlah sampah maka semakin rendah kelimpahan gastropoda. Dimana nilai korelasinya sebesar -0.20506.Kata Kunci : Korelasi; Sampah Laut; Gastropoda; Ekosistem Mangrove.

Abstract

Ascidians are marine invertebrates that have been classified with the subphylum Urochordata (Tunicata). These organisms are very important because they contribute a lot to the stability of the marine ecosystem. This study aims to determine the species composition, density, ecological index (diversity, uniformity, dominance), distribution patterns, and substrate occupied by Ascidian. Data were collected at Mike’s Point Bunaken waters at two depths 7 m and 14 m. The method used a direct observation along the 50 m and 2 m width belt transect. In this study, the transect was applied parallel to the coastline at each depth, where 3 replications were performed. The results show  26 species consisting of 13 species at a depth of 7 m and 22 species at a depth of 14 m were identified. The average density of each species was  0.05 ind/m2 at both depths. Diversity Index of 1.76 at 7 m depth and 2.24 at 14 m depth was determined. Uniformity Index was 0.68 at 7 m depth and 0.73 at 14 m depth. Dominant Index of 0.67 at 7 m depth and 0.88 at 14 m depth was also determined. The distribution pattern of species was dominated by uniform distribution, and the substrate most commonly occupied by Ascidianns was dead coral overgrown with algae.Keywords: Ascidian, composition and density, ecological index, distribution pattern, substrate  AbstrakAscidian adalah Avertebrata laut yang termasuk dalam subfilum Urochordata (Tunicata). Organisme ini sangat penting karena mereka banyak berkontribusi pada stabilitas ekosistem laut. Penelitian ini bertujuan untuk menentukan komposisi spesies, kepadatan, indeks ekologi (keanekaragaman, keseragaman, dominasi), pola distribusi, dan substrat yang ditempati oleh Ascidian. Data dikumpulkan di Perairan Mike's Point Bunaken pada dua kedalaman 7 m dan 14 m. Metode yang digunakan adalah pengamatan langsung sepanjang transek sabuk 50 m dan lebar 2 m. Transek dipasang sejajar dengan garis pantai dan pada setiap kedalaman.  Pengambilan data  dilakukan 3 ulangan. Hasil dari penelitian ini adalah  ditemukan  26 spesies yang terdiri dari 13 spesies pada kedalaman 7 m dan 22 spesies pada kedalaman 14 m. Kepadatan total Ascidian pada kedalaman 7 m (0,67 ind/m2) dan pada kedalaman 14 m (1.11 ind/m2). Indeks Keanekaragaman adalah 1,76 pada kedalaman 7 m dan 2,24 pada kedalaman 14 m. Indeks Keseragaman adalah 0,68 pada kedalaman 7 m dan 0,73 pada kedalaman 14 m. Indeks Dominansi 0,67 pada kedalaman 7 m dan 0,88 pada kedalaman 14 m. Abstract

DNA isolation and gene amplification of algae are significantly influenced by various factors such as characteristics and components of the algae cell wall. Therefore techniques and methods of DNA isolation in certain algae, sometimes only succeed in one particular species and can not be applied to another algae species. Based on that issue, this study was conducted with the aims to determine the succeed of DNA isolation and amplify the rbcL gene as a target gene for identification. Algae DNA was isolated by using innuPrep Plant DNA commercial kit, and the second one with a modified conventional Cetyl Trimetyl Ammonium Bromide (CTAB) method,  for the amplification process was using rbcL gene (ribulose-1,5-bisphosphate carboxylase / oxygenase large subunit) with two pairs of primers : rbcL 7F-753R and rbcL 577F-rbcSR. The results showed that the DNA of Gracilaria sp was succeed isolated by using CTAB method and it was denoted by the presence of DNA bands in agarose gel. Meanwhile DNA amplification for Gracilaria sp., and Sargassum sp., were succeed amplified with the appearance of DNA bands. But in algae Caulerpa sp., was only succeed on 1 pair of primary rbcL 7F and 7.Keywords : DNA, gene rbcL, algae Caulerpa sp., Sargassum sp., Gracilaria sp;AbstrakIsolasi DNA dan amplifikasi gen pada alga sangat dipengaruhi oleh beberapa faktor seperti karakter dan komponen pada dinding sel alga. Oleh karena itu proses isolasi DNA terkadang bisa berhasil pada satu jenis alga, namun tidak berhasil pada jenis alga lainnya. Oleh karena alasan tersebut, maka penelitian ini dilakukan untuk menentukan keberhasilan Isolasi DNA dan mengamplifikasi gen rbcL sebagai gen target identifikasi. Penelitian ini dilakukan dengan tahapan awal Isolasi DNA yang menggunakan kit komersil innuPrep Plant DNA Kit, dan metode konvensional Cetyl Trimetyl Ammonium Bromide (CTAB) yang telah dimodifikasi. Sedangkan untuk proses amplifikasi, menggunakan gen rbcL (ribulose-1,5-bisphosphate carboxylase/oxygenase large subunit) digunakan dua pasang primer yaitu rbcL 7F-753R dan rbcL 577F-rbcSR. Hasil isolasi DNA dari alga Gracilaria sp berhasil diisolasi menggunakan metode CTAB yang ditandai dengan adanya pita DNA pada gel agarose. Amplifikasi DNA pada alga Gracilaria sp., dan Sargassum sp., berhasil diamplifikasi dengan munculnya pita DNA. Namun pada alga Caulerpa sp. hanya berhasil pada 1 pasang primer rbcL 7F dan753R.Kata kunci : DNA, gen rbcL, alga Caulerpa sp., Sargassum sp., Gracilaria sp.
