Abstract

Diabetic nephropathy is one of the diabetes complications attacking kidney leading to kidney damage. Hyperglycemia accompanying DM causes the increase of Ad-vanced Glycation End-Product (AGE) and Receptor Advanced Glycation End-Product (RAGE) activity, then develop kidney damage and other diabetes compli-cations. The study aimed to investigate the effect of black soybean, purple sweet potato, or their combination on the expression of AGE, RAGE, and kidney necro-sis T2DM model rats. The rats were given with high-calorie diet for five weeks and then injected with a low dose of streptozotocin (30 m/kg Body Weight) in intraperitoneal. DM rats were divided into: normal, K- (T2DM), K+ (T2DM + glibenclamide 0.6 mg/kg body weight), P1 (T2DM + black soybean), P2 (T2DM + purple sweet potato), and P3-5 Combination 1-3 (T2DM + combination of black soybean and purple sweet potato in ratio of 1 : 3, 2 : 2, and 3 : 1). DM rats were then given the treatments for thirty days. The effect of black soybean, purple sweet potato, or the combination of both was evaluated through the expression of AGE, RAGE, and necrosis of renal tubules. The changes in renal tubules histological characteristics were evaluated using hematoxylin-eosin (HE) staining. Immuno-histochemistry analysis of renal tubules was to evaluate AGE-RAGE expression after the treatments. The research results indicated that there was a significant dif-ference from the combination of black soybean and purple sweet potato in reduc-ing AGE, RAGE, and renal tubules necrosis. The BSB and PSP combination ratio of 1:1 was able to improve renal tubules, decrease the expression of AGE and RAGE towards near normal. The combination of black soybean and purple sweet potato could be used as one of the alternatives to improve kidney damage in dia-betic nephropathy.

Abstract

Background: Obesity is a global epidemic caused by excessive body fat, which is increasing free fatty acids in the liver, causing oxidative stress and liver cell damage. Green tea yogurt (GTY) with encapsulated Lacticaseibacillus paracasei E1 (GTY-LpE1) might have a beneficial effect in reducing liver cell damage. This study was conducted determine GTY-LpE1 effect on superoxide dismutase (SOD) expression, malondialdehyde (MDA) expression and liver histopathology in high-fat high-fructose diet (HFFD) mice. Material and Methods: A completely randomized design (CRD) with 7 groups, including normal diet (ND) group, HFFD group, 1.3 mg/kg BW simvastatin (SIM)-administered HFFD group, 5 g/kg BW probiotic yoghurt (PY)-administered HFFD (PY), 2.5 g/kg BW GTY-administered HFFD (2.5 GTY), 5 g/kg BW GTY-administered HFFD (5 GTY), and 10 g/kg BW GTY-administered HFFD (10 GTY). The diet was given for 16 weeks, followed by oral administration of sim/yoghurt during the last 4 weeks. Mice were sacrificed and the liver was collected. SOD and MDA expression were analyzed by flow cytometry. Histopathology analysis was done by evaluating hematoxylin-eosin (HE) staining of the liver.Result: The percentage of necrotic cells were 34.55, 34.31, and 21.95%, when treated with 2.5, 5, and 10 g/kg BW with GTY-administered HFFD, respectively, these were lower than the ones in the HFFD group (69.49%). The percentage of MDA expression were 15.55, 18.69, and 22.42%, respectively, these were lower than the ones in the HFFD group as well. The percentage of SOD expression were 9.49, 7.85, and 11.11%, respectively, these were higher than the ones in the HFFD group (3.44%). Conclusion: GTY-LpE1 could decrease the number of necrotic cells in the HFFD mice livers and improve the hepatocyte damage by reducing MDA expression and enhancing SOD expression. GTY-LpE1 can be used as an alternative food to control obesity. Keywords: alginate, chitosan, encapsulation, green tea, probiotic

Abstract

Abstract

Background: Obesity-induced inflammation causes hematopoietic stress, disrupting bone marrow homeostasis. Green tea yogurt supplemented with Lacticaseibacillus paracasei E1 microcapsules (GTYP) is a promising way to overcome obesity due to its high antioxidant and anti-inflammatory activity. However, GTYP effects on blood production, specifically erythrocytes and B cells, remain unexplored.Materials and methods: Male Balb/C mice were fed either a high-fat fructose diet or a normal diet for 12 weeks. Microencapsulation was done by double coating of alginate-chitosan. There were seven groups in this study: normal diet (ND), high-fat fructose diet (HFFD), HFFD with 1.3 mg/kgBW Simvastatin (T1), HFFD with 5 g/kgBW plain yogurt (T2), HFFD with 2.5 g/kgBW GTYP (T3), HFFD with 5 g/kgBW GTYP (T4), and HFFD with 10 g/kgBW GTYP (T5). Erythrocyte counts from the peripheral blood were taken weekly. After 28 days of treatment, mice were sacrificed, bone marrow (BM) and lymphocytes were isolated. The cells of Ter119+, Ter119+CD59+, and B220+SDF-1+ were measured using flow cytometry. Results: HFFD not only reduces the peripheral erythrocyte count (2.15×109 cell/mL) but also affects the hematopoietic system, depleting Ter119+ (11.76%), TER119+CD59+ (0.050%), and B220+SDF-1+ (0.465%). Mice receiving 5 g/KgBW GTYP improved erythrocyte count (9.95×109 cells/mL). The parameters of erythrocyte and B cell development showed more remarkable improvement with GTYP treatment than simvastatin and plain yogurt (p<0.05). Molecular docking also indicated a great inhibitory effect of EGCG (-7.7) for the CXCR4 receptor. Conclusion: GTYP can potentially increase erythrocyte count and B cell development, particularly under obese conditions.   Keywords: anti-obesity, B lymphopoiesis, erythrocyte count, green tea yogurt, probiotics

Abstract

This research was conducted based on the Regulation of the Minister of Education and Culture Number 23 of 2015 concerning character development which is implemented in the School Literacy Movement. The purpose of this research is to describe the application of literacy in Islamic Religious Subject, explain information related to supporting factors, and inhibiting factors for the application of literacy at SMAN 4 Palangka Raya. This research is a descriptive qualitative research and the research subjects are teacher and students at SMAN 4 Palangka Raya. The results showed that the application of literacy in SMAN 4 Palangka Raya reached the second stage, namely the reading and writing stages. The application of literacy in SMAN 4 Palangka Raya is not in accordance with the guidelines in the GLN and GLNB manuals. This is because the books that are often used in literacy during PAI subjects are textbooks, whereas the appropriate books should be non-textbooks. The supporting factor for the application of literacy is the ability of teachers to utilize technology and learning media. The servant factors for the application of literacy are the lack of mentoring, the lack of availability of non-text books and the lack of student persistence and interest.

Abstract

An unhealthy lifestyle can cause changes in the body's metabolism, leading to obesity. The development of obesity is supported by a disturbance in gut microbiota balance that triggers visceral fat deposition in organs such as the testes. Excess fat deposition triggers inflammation, dysfunction, and high ROS production that can damage testicular tissue. Yoghurt, a fermented milk product fortified with green tea, is high in antioxidants that can help reduce excess ROS. Adding encapsulated probiotics in yoghurt can stabilize the gut microbiota in obesity so that dysbiosis can be resolved. This study was conducted to determine the potential of green tea-probiotic yoghurt (GTY) on testicular tissue repair in mice fed a high-fat and fructose diet (HFFD). The research procedure includes feeding HFFD for 3 months, calculating the Lee index, lactic acid bacteria preparation, microencapsulated probiotics, yoghurt preparation, treatment, data collection, including relative weight of testes, the diameter of seminiferous tubules (DST), the epithelium thickness (ET), the number of Leydig cells (LC), and the number of spermatogenic cells. The mice groups were divided into normal (P0), HFFD (P1), HFFD + simvastatin 1.3 mg/Kg BW (P2), HFFD + plain yoghurt 5 g/Kg BW (P3), HFFD + GTY 2.5 g/Kg BW (P4), HFFD + GTY 5 g/Kg BW (P5), and HFFD + GTY 10 g/Kg BW (P6). The results showed that green tea infusion yoghurt with encapsulated probiotics could improve the structure of testicular tissue in mice after HFFD administration. The most effective dose is green tea yoghurt 5 g/Kg BW.
