Abstract

The growth of Cantang grouper is influenced not only by genetic factors but also by the quality of feed, particularly the protein content, which is closely related to amino acid levels. This study aims to investigate the effect of adding commercial amino acids to feed on the growth of Cantang grouper. The research was conducted using an experimental method with a Randomized Block Design (RBD), comprising four treatments and three replicates of amino acid doses added to commercial feed: P0 (0%), P1 (1%), P2 (2%), and P3 (3%). The parameters observed included survival rate, weight growth rate, length growth rate, specific growth rate, and feed conversion ratio. The results showed that adding amino acids to commercial feed over a 21-day rearing period significantly affected all parameters (p<0.05). The highest survival rate, weight growth, length increase, and specific growth rate were found in the P2 treatment with the values of 95.00 ± 5.00%, 7.40 ± 0.24 g, 3.69 ± 0.75 cm, 9.83 ± 0.15%/day, respectively. In addition, the lowest feed conversion ratio was obtained in the P2 treatment (1.22 ± 0.03).

Abstract

Koi fish (Cyprinus rubrofuscus) is one of the aquaculture commodities that is highly marketable because of its unique color pattern and body shape. The prevalence of parasites and pathogenic agents in Koi fish among farmers poses challenges and leads to the occurrence of numerous illnesses. Water quality management is implemented in an effort to prevent several infectious microorganisms. The application of a recirculation system is used to enhance water quality and bolster the disease resistance of Koi fish. The objective of this study was to evaluate the application of RAS technology in the koi fish culture under different filter media on bacterial density. This study used four treatments, namely treatment A (RAS with a dacron filter and pumice), B (RAS with a dacron filter and sand), C (RAS with a sand and pumice filter), and K (control without a recirculation system). The parameter observed in this study was the density of bacteria in the koi fish rearing-media. These findings demonstrated that the utilization of various filters had an impact on bacterial density. Treatment C (RAS with sand and pumice filter) exhibited the greatest bacterial density with an average value exceeding 3.4 log CFU. mL-1 from the first week to the third week. The density of nitrifying bacteria in the koi fish production system is positively influenced by the high-quality maintenance practices that are maintained under optimal conditions.

Abstract

The drying process is often a problem in maintaining the stability of probiotics and the quality of microcapsules in the processing of synbiotic microencapsulation. This study aims to optimize the temperature and drying time to maintain the core material's stability and the microcapsules' quality. The research design in this study was a completely randomized design with two factorials, namely drying temperature (30°C, 35°C, and 40°C) and drying time (2, 2.5, and 3 hours). The results showed that the highest viability and efficiency values were obtained in the drying process at 40°C for 2 hours, which were 9.29 log CFU/mL and 98.10%. In addition, the water content and gel strength of the microcapsules obtained also showed optimal conditions. Based on the results obtained, it can be concluded that the drying process at 40°C for 2 hours is the best treatment, with the highest viability and EE values and low water content values.

Abstract

Tilapia (Oreochromis niloticus) is a freshwater fish that has high economic value and potential to be cultivated. The problem that often arises in tilapia cultivation is the lack of optimal use of the feed given. The purpose of this research was to find out blood profile of tilapia during culture period. The method used was a completely randomized design (CRD) with five treatments and every treatment with four replications. This study used 240 Tilapia with average weight of 15 g. The stocking density of the aquaria is 15 fish. Test feed in this study is the form of dry pellets consisting combination of GAA levels, protein content (25% dry matter bases), and source of protein (fish meal and non-fishmeal). During the cultured period of 70 days, fish fed by formula diet of 3% biomass per day. Sampling for blood profile tested of erythrocytes, leukocytes, and hematocrit is carried out after the research. The water quality of temperature, pH, and DO were daily monitoring. The combination treatment of guanidino acetic acid (GAA) levels with different protein sources (fishmeal and non-fishmeal) had a significant effect on total erythrocyte of tilapia, but did not have significant differences between treatments on total leukosyte and hematocrit. Treatment with 1.2 g/kg GAA, fish meal protein source was the best treatment in this research.

Abstract

Abstract. Mahariawan IMD, Kusuma WE, Yuniarti A, Beltran MAG, Hariati AM. 2021. Application of wheat flour (Triticum aestivum) on spore density and sporulation efficiency of Bacillus megaterium isolated from Litopenaeus vannamei gastrointestinal tract. Biodiversitas 22: 3709-3715. Bacillus megaterium is frequently used in fish farming, such as white shrimp (Litopenaeus vannamei) pond, which can produce spores with high stability in its implementation. Currently, spore production still requires the availability of high-cost carbon sources. The objective of this research was to evaluate the effect of different wheat flour doses on spore density and sporulation efficiency of B. megaterium BM1. In flasks, 50 mL of each test medium was treated with different doses of wheat (10, 20, 30 and 40 g. L-1, respectively) and glucose was used as a control. Each treatment was inoculated with B. megaterium BM1 (2.6 x 108 CFU. mL-1) and incubated in a shaker incubator (120 rpm) at 37 °C for 120 hours. The results showed that the highest vegetative cell concentration (17 x 108 CFU. mL-1), growth rate (0.8 hour-1) and spore (14.7 x 108 spores. mL-1) were found in the wheat flour dose of 30 g. L-1. Furthermore, the highest sporulation efficiency was achieved at 20 g. L-1 of wheat (91.30%) and germination should be done at a dose less than 40 g. L-1. The size of the spores was 1.35-1.39 µm. Thus, 30 g. L-1 of wheat flour is a potential dose to produce spore for probiotic candidates.

Abstract

Streptococcus agalactiae is a pathogenic bacteria in aquaculture, causing streptococcosis in fish and causing significant economic losses. The pathogenicity of this bacteria is related to its virulence factors, including its ability to form biofilms that increase antibiotic resistance and host immune responses. One alternative approach to controlling infection is to disrupt the bacterial quorum sensing (QS) communication system through a quorum quenching mechanism. This study aims to analyze the potential of octopus ink extract as a quorum quenching agent in inhibiting S. agalactiae biofilm formation in aquaculture. The method used was a systematic literature review by collecting and analyzing various scientific sources such as journals, books, and relevant articles. Data analysis was carried out using content analysis techniques by selecting the literature most relevant to the research topic. The results of the study indicate that octopus ink contains bioactive compounds, especially alkaloids, which have the potential to inhibit the QS system, suppress the expression of virulence factors, and inhibit bacterial biofilm formation. In addition, octopus ink extract also has the potential to improve the immune system of farmed fish. Thus, octopus ink extract has the potential to be developed as an environmentally friendly antibiofilm and immunostimulant agent. Further research is needed to identify specific active compounds and test their effectiveness and safety on a cultivation scale.
