Abstract

Field survey on ascidian community was conducted along the coastal area of Malalayang Dua in order to find out species of ascidia, species abundance, and ascidian substrates. A survey method and quadrant transects were applied. Pictures were taken, while species and their substrates were sampled.  Species identification was based on morphological characteristics, while substrate type identification was based on ascidian species attachment.  The results shows that 21 ascidian species were found in the the coastal of Malalayang Dua.  Didemnum molle was the highest abundant species in the area, followed by Polycarpa aurata, Polycarpa sp.4. and Polycarpa sp.2.. Dead coral algaes (DCA) were found to be the most preferred  substrates by ascidians in the area. Keywords : ascidia, species, substrate, distribution, and abundance Survei lapangan terhadap komunitas ascidia dilakukan  di pesisir Malalayang Dua untuk mendapatkan data jenis, kelimpahan, dan substrat ascidia.  Metode yang digunakan yaitu metode survei jelajah dan transek kuadran. Identifikasi jenis ascidia dilakukan berdasarkan karakteristik morfologi.  Hasil penelitian ditemukan ada 21 jenis ascidia.  Substrat jenis death coral algae (DCA) merupakan substrat yang paling banyak ditempati ascidia. Kelimpahan ascidia tertinggi adalah Didemnum molle di pesisir Malalayang Dua, diikuti oleh Polycarpa aurata,   Polycarpa sp.4. dan Polycarpa sp.2. Death coral alga (DCA) ditemukan sebagai substrat yang paling disukai oleh ascidia di daerah itu. Kata Kunci : ascidia, spesies, substrat, distribusi, dan kelimpahan

Abstract

Phyllidiella nigra is an organism that is suspected to have secondary metabolites because their ability to develop its self defense system by camouflage and using chemical compounds derived from their nature diet as deterrent against their predators. The purpose of this study was to isolate symbiotic bacterial derived from P. nigra, extracted and followed by, the antibacterial assays against Escherichia coli and Bacillus megaterium as well as the anti-UV assay. The results showed that the five isolates tested had an antibacterial activity with the highest average inhibition zone against E. coli DSM 498 bacteria, isolate 1 (14.67 mm), isolate 5 (14 mm), and against B. Megaterium DSM 32T bacteria, isolate 3 (13.33 mm). The three isolates which had the highest inhibition zone and P. nigra extract were tested for anti-UV assay using a UV-Vis Spectrophotometer. The results obtained isolate 3 has absorption of UV-A with the UV absorbtion maximum at λ 340 nm and P. nigra extract has absorption on UV-B radiation with UV absorption maximum at λ 290 nm. Key words: Nudibranchia, Bacteria, Anti-bacteial, Anti-UV Phyllidiella nigra merupakan organisme yang diduga memiliki metabolit sekunder karena mampu mengembangkan sistem pertahanan dirinya dengan cara kamuflase dan menggunakan senyawa kimia sebagai racun yang didapat dari makanannya. Tujuan dari penelitian ini yaitu mendapatkan isolat bakteri yang bersimbiosis dengan P. nigra, mendapatkan ekstrak dari baktri simbion, dan menguji antibakteri dan anti-UV ekstrak etil aseta bakteri simbion dengan metode difusi agar terhadap bakteri Escherichia coli dan Bacillus megaterium. Hasil penelitian didapatkan kelima isolat yang diuji memiliki aktivitas antibakteri dengan rerata zona hambat tertinggi terhadap bakteri E. coli DSM 498 yaitu isolat 1 (14,67 mm), isolat 5 (14 mm), dan terhadap baktri B. megaterium DSM 32T yaitu isolat 3 (13,33 mm). Ketiga isolat yang memiliki zona hambat tertinggi dan ekstrak P. nigra diujikan anti-UV menggunakan alat UV-Vis Spektrofotometer. Hasil yang didapat isolat 3 memiliki serapan terhadap radiasi sinar UV-A dengan puncak tertinggi pada λ 340 nm dan ekstrak P. nigra memiliki serapan terhadap radiasi sinar UV-B dengan puncak tertinggi berada pada λ 290 nm. Abstract

This study aims to 1) identify the species of Nudibranchia in North Sulawesi marine waters, especially in Malalayang waters, Teep waters, Sangihe waters, Bunaken waters, Gangga waters and Lembeh straits, 2) find out species of Nudibranchia that is found in all sites, and 3) find out index community structure of Nudibranchia in all sites.  The result show that there were 11 species of Nudibranchia in Malalayang waters, 27 species in Lembeh Strait, 11 species in Teep waters, 57 species in Bunaken waters, 91 species in Bangka waters, and 20 species in Sangihe waters. Up to now, there were 139 species of Nudibranchia in North Sulawesi marine waters, where Species of Phyllidiella pustulosa, Phyllidia ocellata, Phyllidia varicose, Chromodoris annae, and Glossodoris cincta were found in all study locations. The species of Doriprismatica atromarginata was only found in the Malalayang waters.  A high Index Diversity was found in Bunaken and Bangka waters, and a middle Index Diversity was found in the waters of Lembeh Strait, Teep, and Sangihe. A stable community structure was found for all locations, while a low domination was categorized to all locations. Keywords: Nudibranchia, species, diversity, evenness, domination, and community structure.

Abstract

The purpose of this study was to determine the composition of nematocysts in the body tissue of the coral fungia, C. vaugani.  The location for the collection of C. vaugani colonies was on the coral reefs of the Malalayang coastal area, Manado. This research was carried out in September - December 2020. The observation of nematocysts of C. vaugani was carried out using an Olympus CX41 microscope with a magnification of x100 objective lens which connected to a computer monitor equipped with an optical view 7 application at the Fish Health Environment and Toxicology Laboratory, Faculty of Fisheries and Marine Science, UNSRAT.  The present results showed that C. vaugani had a composition of nematocysts, namely type I microbasic p-mastigophore (MpM-I) of 40.00%, small type II microbasic p-mastigophore (sMpM-II) of 31.11%, small holotrichous isorhizas (sHI) of 24.44 %, large type II microbasic p-mastigophore (lMpM-II) of 2.22%, and large holotrichous isorhizas (IHI) of 2.22%.  sMpM-II is the most dominant nematocyst type of C. vaugani and this type of nematocyst is thought to be used by coral for aggression and killing prey or predator if there is disturbance from the surrounding aquatic environment where the coral lives.  Further research is recommended to study the types and morphology of nematocyst from various species of coral fungia, including C. vaugani to understand the differences in their types and dimension of the nematocyst. Keywords: Composition, Cycloseris vaugani, Fungia, Manado, nematosit

Abstract

The diversity of crabs in the Malalayang One coast is thought to vary between research locations and limited information on biota that live on rocky, sandy and muddy coastal beaches. Sampling was carried out at low tide at night by capturing directly using a protected hand and for lighting with the help of a flashlight. The collected samples were taken to the Aquaculture Technology Laboratory, Faculty of Fisheries and Marine Sciences, UNSRAT to be identified and for diversity using the Shanon-winer mathematical formula. The results of crabs obtained at five coastal locations on the coast of Malalayang, Manado, with a total of 106 with 7 species, namely Metopograpsus latifrons, Sesarmops impressus, Ocypode kuhlii, Macrophthalmus mareotis depressus, Uca vocans dussumieri, Grapsus Longitarsis and Grapsus albolineatus.Keywords: Crab, Morphology, Diversity, Coastal Ecosystem.
