Abstract

Kerusakan ginjal berdampak pada proses eritropoiesis karena ginjal merupakan organ utama penghasil eritropoietin (EPO). Tanaman kumis kucing secara turun-temurun sudah digunakan untuk pengobatan gangguan saluran kemih, hipertensi, dan diabetes mellitus. Tanaman ini diketahui mengandung berbagai zat aktif yang bersifat nefroprotektif, seperti flavonoid, tannin, saponin, phenol, serta terpenoid. Penelitian ini bertujuan untuk melihat potensi tanaman kumis kucing untuk memperbaiki profil eritosit pada kondisi kerusakan ginjal yang diinduksi oleh etilen glikol. Sebanyak 35 ekor tikus galur Sprague Dawley jantan berumur 7 bulan dibagi ke dalam 7 kelompok perlakuan dengan pemberian etilen glikol dan atau ekstrak kumis kucing dengan lama permberian yang berbeda. Pemberian ekstrak daun kumis kucing mampu meningkatkan jumlah eritrosit pada tikus serta menekan penurunan jumlah eritrosit saat dan setelah pemberian etilen glikol. Selain itu, pemberian ekstrak kumis kucing juga mampu menurunkan jumlah leukosit serta rasio neutrofil: limfosit yang meningkat akibat induksi etilen glikol. Ekstrak kumis kucing memiliki potensi untuk memperbaiki proses eritropoiesis dan menekan peradangan pada kasus kerusakan ginjal.

Abstract

Singkong merupakan makanan pokok pengganti nasi dengan sumber karbohidrat tinggi. Singkong dapat digunakan sebagai bahan baku utama pembuatan lanting. Penelitian ini mengkaji potensi dan persepsi masyarakat terhadap produk makanan tradisional lanting berbahan dasar singkong di Kecamatan Cibeber Kabupaten Lebak-Banten. Metode penelitian yang digunakan adalah deskriptif kualitatif, pengumpulan data dilakukan melalui wawancara kepada produsen dan distributor serta penyebaran kuesioner kepada 42 responden konsumen. Hasil penelitian menunjukkan bahwa lanting populer di kalangan masyarakat, sebanyak 78,6% responden menyatakan menyukai produk ini dan 9,5% sangat menyukai. Kemudahan akses terhadap lanting juga dinilai tinggi oleh sebagian besar responden, meskipun ada pula yang mengalami kesulitan karena distribusi yang tidak merata. Potensi pasar Lanting yang sangat besar didukung oleh cita rasa khas dan manfaat singkong bagi kesehatan. Meskipun distribusi dan kapasitas produksi masih menjadi tantangan, inovasi dalam pemasaran dan pengemasan serta peningkatan kerja sama distribusi dapat meningkatkan keberhasilan produk di pasar yang kompetitif. Penelitian ini menyimpulkan bahwa lanting memiliki nilai ekonomi bagi produsen lokal dan berkontribusi terhadap diversifikasi pangan dan ketahanan pangan nasional.

Abstract

Chemistry is a branch of science that studies various natural phenomena, including the properties, changes in matter, structure, and the energy that accompanies them. The study of chemistry is interconnected with its concepts, making it continuous. The aim of this research is to analyze misconceptions about acid-base materials using a four-tier multiple-choice diagnostic test instrument. The research method used is descriptive qualitative, aiming to analyze student misconceptions. The subjects of this research were class XI students of SMK KIMIA PGRI SERANG CITY. To collect data, observation techniques, four-tier diagnostic tests, interviews, and documentation were used. The results of this research show that 40% of students have misconceptions about acid-base materials, 13% of students understand the concept, and 47% of students don't understand the concept. More students experience misconceptions compared to students who understand the concept. Based on the percentage results, the level of misconceptions for class XI students at SMK KIMIA PGRI Serang City is classified as medium. According to the results of interviews with students, student misconceptions are caused by two factors: internal factors (coming from students) and external factors (coming from learning media such as books and teachers). The factor that causes students' misconceptions is that schools emphasize practice rather than theory, where practice is 60% and theory is 40%.

Abstract

AbstractToxoplasma gondii is an obligate intracellular parasite, causing toxoplasmosis in almost all warm-blooded animals and humans worldwide. Toxoplasmosis is a zoonotic disease of serious public health concern. Host cell invasion by T. gondii tachyzoites has process involving the sequential secretion of Excreted-Secreted Antigens (ESAs). T. gondi ESAs could be a valuable candidate for the diagnosis of toxoplasmosis. Techniques to more accurately detection of T. gondii recently developed biotechnological methods that are currently being used, conventional and real time Polymerase Chain Reaction (RT-PCR). RT-PCR is more widely used because it is more sensitive and specific. The aims of this study were to optimize the Sybr Green RT-PCR in different region gene based on Excreted-Secreted Antigens (ESAs), tachyzoite surface antigen and bradhyzoite antige, then adapt the conventional PCR program to real-time PCR for detection Toxoplasma gondii. Optimization is necessary to get optimal condition of PCR to get the best results. T. gondii RH strains derived from liquid nitrogen and DNA extracted by DNAzol. The genetic marker used GRA1#1, GRA1#2, GRA7#1, GRA7#2, ROP1, MIC3, SAG1 and BAG1. The results of the optimization of multiple primer genes can adapt and be used optimal in RT-PCR by using the same cycle program simultaneously in one run. Overall, RT-PCR for the detection of T. gondii DNA demonstrated excellent agreement with conventional PCR. RT-PCR with melting curve analysis is rapid and simple that facilitates high throughput analysis to detect T. gondii. The optimal conditions obtained from the optimization results can facilitate further research to detect T. gondii.Keywords: Biotechnology molecular, Detection, excretory-secretory antigen, toxoplasmosis
