Abstract

The quest for better and more effective treatments has encouraged the search for therapies derived from natural sources to obtain effective immune therapy, considering that several pandemics have arisen caused by viruses. Developing fucoidan from brown algae in drug encapsulation as an immunomodulator could be more promising. This study aimed to produce nanoencapsulation loaded with fucoidan-purified extract from brown algae and to evaluate the influence of nanoencapsulation formulation on the immunomodulatory activity of fucoidan. Fucoidan was obtained from brown algae and extracted by hot aqueous, followed by ethanol purification. Nanoencapsulation of fucoidan purified extract was prepared using the ionic gelation method. The carbon clearance method was carried out for the immunomodulatory activity test of the nanoencapsulation of fucoidan purified extract. Nanoencapsulation of fucoidan purified extract with the optimum composition of maltodextrin 9.9% and S-TPP 0.1% (1:5) resulted in particle size of 715.4 nm, zeta potential -0.1 mV, pH 7.54, transmittance 97.54%+0.08, and entrapment efficiency 89.94%+0.17. The carbon clearance test showed that the nanoencapsulation of fucoidan was a strong immunostimulant with a phagocytosis index of 1.65. The development of nanoencapsulation could increase the phagocytosis index of fucoidan purified extracts from brown algae. Further molecular studies are needed to demonstrate the molecular activity of this preparation as an immunomodulator.

Abstract

The village of Jagaraga, located in the Sawan district of Buleleng Regency, Bali Province, had significant potential in both culture and agriculture, particularly in rice farming. More than half of the village’s land area was rice fields, and over two-thirds of the villagers were rice farmers. However, ensuring healthy soils and crops through effective fertilizer solutions remained a major challenge. The aim of the community development initiative was to empower the village farmers to become innovative through collaborative learning and by inventing and commercializing a slow-release nano fertilizer. A participatory action and learning approach was used to engage the rice farming community. The Subak Babakan farmer organization and the local rice milling enterprise, PB Suwela Amertha, were involved in the empowerment activities, which included information sessions, training, technology application, guidance, evaluation, and sustainability planning. As a result, four formulas of slow-release organic nano fertilizers in pellet form were successfully developed. Additionally, a four-helix partnership involving academicians, the farmer community, enterprises, and the government was established to build a business platform for commercializing the innovation. Through this collaboration, academicians and community farmers successfully learned to address the primary challenges in rice agriculture innovatively while promoting sustainable practices to maintain the health of the land and the well-being of the villagers.

Abstract

Bali is one of clove (Syzygium aromaticum) producers in Indonesia. Clove essential oil is mainly produced from the leaves and flowers. Eugenol is the main component in the essential oil of clove. The objective of this research is to determine constituents and antioxidant activity of clove’s bud and leaf essential oils from Bali. The essential oils were isolated from clove’s bud and leaf samples by steam distillation with the yield of 12.90 and 2.63%. The constituents of the clove essential oils were analyzed by using gas chromatography-mass spectrometry (GC-MS). Thirty-six and twenty-nine constituents were identified based on GC-MS from the clove bud and leaf essential oils, respectively. Major classes of compounds are sesquiterpenes, phenyl propanoids, oxygenated sesquiterpenes, and esters. Different compositions in major constituents were found between both essential oils. Clove bud essential oil (CBEO) contained eugenol (65.29 %), trans-caryophyllene (20.06 %), and α-humulene (3.38 %). While, in clove’s leaf essential oil (CLEO), the composition was eugenol (64.47 %), trans-caryophyllene (27.19 %), and α-humulene (3.62 %). The clove essential oil and its main component show strong antioxidant activity. The antioxidant activity of CBEO, CLEO, and eugenol is 22.58, 29.19, and 17.53 μg/mL, respectively.
