Abstract

This study aimed to select a proteolytic LAB with probiotic properties that can be applied to manufacture protein hydrolysate from fish heads. Tests on 20 isolates of LAB showed that nine isolates were proteolytic and non-pathogenic. A total of 5 isolates could grow well at 0.5% bile salt stress, and 3 of them could grow at pH 3. These three isolates had antagonistic ability against Salmonella bacteria, and one isolate was sensitive to the antibiotic tested. Molecular identification of the selected LAB isolates showed a 100% sequence similarity with Pediococcus pentosaceus with accession number MT515895.1. The LAB isolate has high proteolytic activity since it can increase the soluble fraction of fish meal powder from 32.10% to 88.38% in 48 hr. Production of protein hydrolysate using tuna waste was carried out for 30 days. Tuna waste protein hydrolysate had a medium antioxidant activity of 25.57 ± 0.93%. The hydrolyzed protein comprised 17 amino acids, including nine non-essential and eight essential amino acids, and is dominated by glutamic acid. Selected LAB isolate is potentially used in protein hydrolysate production, especially for flavor enhancers. Keywords: antagonistic, amino acid, Pediococcus pentosaceus

Abstract

The women empowerment program in Negeri Hulaliu aims to improve nutritional fulfillment and family economic welfare through diversification of fish products into meatballs. This activity was carried out as a response to the high potential of fish resources but the low utilization of processed fish products. In this program, mothers are trained to process fish into meatballs that are nutritious and have economic value compared to fresh fish. The methods used include participatory approach, technical training on fish processing, mentoring, evaluation and monitoring. The results of this program showed an increase in participants' skills in processing fish and family economic income through the sale of fish balls. In addition, this program also contributes to the increased consumption of animal protein, which indirectly supports the improvement of family nutrition and economy. Thus, diversification of fish products into meatballs is expected to be one of the solutions in improving nutritional and economic intake in Negeri Hulaliu.

Abstract

Bulu babi Diadema setosum relatif melimpah di Indonesia. Penelitian ini bertujuan untuk mengidentifikasi senyawa fitokimia dan kemampuan antibakteri ekstrak etanol cangkang dan duri bulu babi Diadema setosum terhadap isolat bakteri Escherichia coli, Staphylococcus aureus, dan Salmonella sp. Metode penelitian yang dilakukan meliputi ekstraksi cangkang dan duri bulu babi Diadema setosum dengan cara maserasi menggunakan etanol 96%, kemudian dilakukan skrining fitokimia dan uji aktivitas antibakteri dengan metode difusi sumur. Hasil penelitian menunjukkan bahwa skrining fitokimia ekstrak cangkang bulu babi (Diadema setosum) mengandung senyawa alkaloid, flavonoid, dan saponin. Senyawa yang ditemukan pada ekstrak duri bulu babi Diadema setosum antara lain alkaloid, flavonoid, dan tanin. Pengujian antibakteri dilakukan dengan menggunakan metode difusi. Hasil uji aktivitas antibakteri menunjukkan bahwa kedua sampel (cangkang dan duri) menghasilkan zona hambat terhadap isolat bakteri Escherichia coli, Staphylococcus aureus, dan Salmonella sp.
