Abstract

Hot springs are a common source of thermophyles which produce thermostable enzymes. The objective of this study was to isolate and identify thermostable amylase-producing bacteria from a local geothermal spring. An amylase–producing bacterium strain was isolated from this hot spring which excreted amylase after being grown on starch agar screening plates at 37 °C. It was identified as Bacillus megaterium using the method of 16S ribosomal DNA. The organism is a rod-shape and is a spore-forming  bacterium.  Maximum amylase production was achieved after incubation in the production media for 72 hours. Preliminary analysis of the secreted amylase showed that the enzyme could bind to DEAE-Sepharose matrix and was discharged by eluting with 0.5 M NaCl. The partially purified enzyme was stable up to 75 °C, showing that this enzyme might have potential application in the starch-processing industry.

Abstract

Cellulose is included in the main polysaccharide group that is widely available in nature. Cellulose is a straight-chain glucose polymer that is bound by β-1,4 glucoside bonds. Cellulase is an enzyme complex consisting of several enzymes that work in stages or together in breaking down cellulose into glucose, by hydrolyzing the β-1,4 glucoside bond in cellulose. Cellulase enzymes have been used in various industries. Industrial applications require enzymes that are stable at high temperatures. Various types of thermostable enzymes can be isolated from thermophilic bacteria, cellulase enzymes are no exception. This study aims to isolate and identify thermophilic bacteria that produce cellulase enzymes from Sila hot springs. The results of isolation and screening of cellulase-producing bacteria from hot water samples obtained by 7 isolates that have the ability to degrade cellulose can produce clear zones on LB-CMC agar selective medium, but CR2 isolates showed the largest zone of 1.5 cm. CR2 isolates were Gram-positive bacteria and were identified as having close relations with Bacillus aryabhattai strain B8W22 using the 16S rRNA method.Keywords: Cellulose; Cellulose enzyme; Thermostable enzyme; Thermophilic bacteria

Abstract

Birds are an important component in a forest ecosystem. Birds play a role in pollination, seed dispersers, pest control, and even their droppings provide nutrients for the soil. After the earthquake in Ambon and surrounding areas in September 2019, many people fled to the forest. This research was conducted in Waai village to inventory the species of birds in the forest (HA) and forests that were converted into post-earthquake refuge (HP) using the list of bird species and natural sciences. The results show that 8 species of birds were found at the HA location while 3 species of birds were found at the HP location. The highest species abundance was Passer montanus at both locations. The lowest abundance value on HA is Todirhamphus chloris and Ducula bicolor, while on HP is Leptocoma aspasia. Diversity index analysis shows 1.67 index on HA and 0.86 on HP.

Abstract

Pendahuluan. Yoghurt merupakan produk hasil fermentasi yang mengandung asam laktat, dengan tambahan buah sebagai sumber prebiotiknya. Sirsak (Anonna muricata L.) mengandung serat pangan dan protein yang memberikan energi bagi proses metabolisme dalam yoghurt sehingga meningkatkan jumlah bakteri asam laktat dan menghasilkan lebih banyak senyawa antibakteri. Penambahan wine pala pada yoghurt buah sirsak dapat memberikan cita rasa pada yoghurt yang mengandung zat aktif yaitu alkaloid yang bekerja sebagai antioksidan primer. Tujuan. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dan aktivitas antioksidan dari yoghurt sari buah sirsak dengan penambahan wine pala. Metode. Metode yang digunakan adalah metode hitung cawan (Total Plate Count) untuk pengujian total BAL (Bakteri Asam Laktat), metode sumuran (Well diffusion) untuk uji aktivitas antibakteri, dan metode DPPH (2,2-difenil-1-pikrilhidrazil) untuk uji aktivitas antioksidan. Hasil. Hasil menunjukkan total BAL yoghurt sari buah sirsak yang terbaik pada konsentrasi 25% (9×107). Nilai pH dari yoghurt sari buah sirsak berkisar antara 3-4. Hasil pengujian antibakteri menunjukan terbentuk zona bening di konsentrasi 75% pada Eschericia coli ATCC 8739 (2,67 mm) dan Staphylococcus aureus ATCC 6538 (8,33 mm). Aktivitas antioksidan yang terbaik pada konsentrasi 25% (53,59%). Data yang diperoleh menunjukkan total pH, total bakteri asam laktat, dan aktivitas antioksidan, yoghurt sari buah sirsak dengan penambahan wine pala yang baik adalah konsentrasi 25%. Kesimpulan. Yoghurt konsentrasi sirsak 75% dengan penambahan wine pala memiliki aktivitas antibakteri yang baik terhadap E.coli dan S. aureus, sedangkan konsentrasi 25% menunjukkan nilai total BAL, pH, dan aktivitas antioksidan yang paling baik.

Abstract

Coconut is a plant and natural potential that is excellent in Latuhalat Village. Coconut has so many benefits that it has earned the nickname the tree of life because every part of it can be used for the benefit of society. Coconut processing in Latuhalat Village is carried out on a small and traditional scale, namely processing it into coconut cooking oil. This processed product will leave processed waste (processed waste) known as "taiminya". This processed residue is usually not used anymore, then thrown away or used as animal feed. The Pattimura University PKM Team presents innovation and development of taiminya into "taiminya confectionery" and "taiminya chili sauce" products. This innovative product is expected to increase the income of the people of Latuhalat Village

Abstract

Drugs are one of society's diseases that are growing very rapidly and are difficult to eradicate. The main target of the largest distribution and use is dominated by the younger generation while the younger generation is the backbone and driver of change for the Indonesian nation. Formation of morals and ethics from an early age is the first step as an effort to prevent drug use, so that in real work lectures in Amahusu Village students run a program namely socialization of the dangers of drug use for the millennial generation. The purpose of this program is to shape the character of students by introducing caused by drug use. This activity was carried out on November 15 2022 located at SMP Negeri 11 Ambon. The implementation method uses lecture and sharing methods. With this activity, it is hoped that students will not only know the dangers but can fight drugs for the sake of creating a healthy life, a harmonious family and a bright future.

Abstract

Sustainable agriculture systems utilizing PGPR (Plant Growth-Promoting Rhizobacteria) have the potential to increase due to their environmentally friendly and practical nature, to replace the increasing use of chemical-based fertilizers and pesticides. Peanut (Arachis hypogea L) local variety Maybrat is a plant that has the potential to be isolated by bacteria in its rhizosphere area to be used as a biostimulant candidate. This study aimed to determine the type of rhizobacteria in the rhizosphere area associated with local varieties of peanuts in the Maybrat Regency and obtain superior rhizobacteria for biostimulants (PGPR). A total of 8 isolates were successfully isolated from peanut rhizosphere and based on physiological characteristics showed that KM2 isolates produced the highest IAA of 2,625 mg l-1, and KM8 isolates produced the highest GA3 concentration of 4.19 mg l-1. KM7 isolates produce salicylates with the highest concentration of 0.788 mg l-1, while KM3 isolates produce the highest type of catechols of 1.89 mg l-1. The KM8 isolate is capable of dissolving the highest phosphate of 12.46 mg l-1. Cyanide production tests showed that all eight bacterial isolates did not have the ability to produce cyanide. Based on physiological characteristics, it shows that these 8 isolates have the potential to be biostimulants and biofertilizers for plants.

Abstract

Disaster mitigation is an effort to reduce casualties and provide a proper and good understanding to the public regarding natural disasters. This activity aims to provide early insight and knowledge regarding natural disasters. The method of implementing this community service uses the socialization method with Rutong State Elementary School students as participants. This socialization material focuses on mitigating the tsunami and earthquake disasters, a major threat to Rutong Village, located in a coastal area.

Abstract

Consumerism has become a problem in many communities nowadays, especially among teenagers. Growing a culture of saving from an early stage can solve this problem. Therefore, socialization with students about the importance of a culture of saving from an early age is essential to shape their behaviour to be more frugal and teach them to manage their finances. The socialization aims to educate students on the importance of saving early on and motivate them to set aside part of their income for savings. The result of the socialization is to grow students' enthusiasm for saving and increase knowledge about saving, such as the definition, the benefits, and the methods; it is highly expected that the seventh-grade students of SMP Negeri 8 Ambon can implement the saving habit in their lives.

Abstract

Environmental care activities carried out in the form of reforestation in Negeri Kilang are also an implementation of the Pattimura University KKN student program, the first batch of Class L, aimed at increasing the knowledge of the Negeri Kilang community about the role and function of plants, as well as creating the character of the Negeri Kilang community who care about the environment. Carrying out activities is a form of assistance in the field as well as outreach to village communities regarding reforestation efforts as a form of protecting and preserving the environment. The results of this tree planting activity are the planting of clove, mahogany, ironwood and durian tree seedlings in reforestation locations and in hamlets/community gardens which can be utilized according to their intended purpose to make land use more efficient, apart from optimizing ecological functions, as well as providing economic value for the people of the Negeri Kilang.
