Abstract

Keanekaragaman hayati Indonesia mencakup temu mangga (Curcuma mangga Val.) yang ekstrak etanol daunnya berpotensi farmakologis namun menghadapi tantangan stabilitas dalam bentuk tradisional. Penelitian eksperimental laboratorium berpendekatan kuantitatif ini bertujuan memformulasikan sediaan mikroenkapsulasi metode gelasi ionik (konsentrasi 0,25%, 0,5%, 0,75%) dan mengevaluasi karakteristik fisik meliputi stabilitas, kadar air, dan daya lekat. Populasi daun temu mangga dari Medan (1.100 g simplisia, rendemen 12,2%) diambil sampel purposif menjadi empat formulasi memenuhi kriteria inklusi (bentuk bulat/oval, permukaan halus). Instrumen mencakup rotary evaporator, syringe 22G, oven pengering, dengan teknik analisis skrining fitokimia, cycling test, perhitungan kadar air, dan uji daya lekat mukosa lambung tikus. Hasil menunjukkan rendemen 12,2%, fitokimia positif (alkaloid, flavonoid, saponin, terpenoid, steroid, fenol), stabilitas organoleptik enam siklus, kadar air 0,75-1,34%, dan daya lekat hingga 120 menit. Simpulan menyatakan mikroenkapsulasi natrium alginat-kitosan menghasilkan sediaan herbal stabil ber-kadar air rendah dan mukoadhesi baik, potensial untuk pengembangan farmasi modern.

Abstract

Irreversible skin aging begins in early adulthood due to intrinsic factors such as genetics and cellular metabolism and extrinsic factors such as ultraviolet exposure, smoking, pollution, and lifestyle, which trigger reactive oxygen species (ROS) and collagen damage, leading to wrinkles and reduced skin elasticity. This study aimed to formulate an optimal anti-aging cream from ethanol extract of porang tubers (Amorphophallus muelleri Blume) using variations in the HLB value of Span 60-Tween 80 emulsifiers. This quantitative experimental study used three cream formulas (F1: HLB 4.95; F2: HLB 5.7; F3: HLB 6.8) prepared from 3 kg of porang tubers obtained from Sirapit, Langkat. Evaluations included organoleptic properties, homogeneity, pH, viscosity, spreadability, adhesiveness, cycling-test stability, irritation, preference, and antioxidant activity using the DPPH method. The results showed that F2 was the optimal formula, with medium viscosity, spreadability of 6.2 cm², pH 5.8, stability for six cycles, and an IC₅₀ value of approximately 114 μg/mL, indicating moderate antioxidant activity. Formula F2 therefore has potential as a topical antioxidant preparation for preventing premature skin aging based on locally sourced tropical material.

Abstract

Background: Kidney disease remains a major global health problem, one of which is associated with the use of nephrotoxic drugs such as gentamicin. Gentamicin induces renal damage through oxidative stress and inflammatory mechanisms. Muntingia calabura L. fruit contains bioactive compounds, including flavonoids and phenolic substances, which are believed to have nephroprotective properties. Objective: This study aimed to evaluate the nephroprotective effectiveness of Muntingia calabura fruit extract on renal histopathological changes and inflammatory markers in gentamicin-induced rats. Methods: This experimental laboratory study employed a post-test only control group design. A total of 25 male white rats were divided into five groups: negative control (0.5% CMC-Na), positive control (gentamicin 80 mg/kg BW), and three treatment groups receiving gentamicin 80 mg/kg BW combined with graded doses of Muntingia calabura fruit extract (50, 100, and 200 mg/kg BW) orally for 7 days. Renal histopathology was examined using hematoxylin-eosin (HE) staining, while serum interleukin-6 (IL-6) levels were measured using enzyme-linked immunosorbent assay (ELISA). Results: Administration of Muntingia calabura fruit extract significantly reduced IL-6 levels and improved renal histopathological features compared to the positive control group (p < 0.05). The nephroprotective effect increased in a dose-dependent manner, with the highest dose (200 mg/kg BW) showing renal conditions closest to normal. Conclusion: Muntingia calabura fruit extract has potential as a natural nephroprotective agent against gentamicin-induced nephrotoxicity through inhibition of inflammatory response as indicated by decreased IL-6 levels.

Abstract

Background: Gentamicin is a nephrotoxic antibiotic that induces kidney damage through oxidative stress and elevated TNF-α levels. Coffee fruit (Coffea arabica L.) contains bioactive compounds such as polyphenols and chlorogenic acid with potential nephroprotective effects. Objective: This study aimed to evaluate the nephroprotective efficacy of coffee fruit extract on renal morphological changes and TNF-α levels in gentamicin-induced rats. Methods: An experimental randomized group design was employed using 30 male Wistar rats divided into five groups: normal control, negative control (gentamicin 100 mg/kgBW), and three treatment groups receiving gentamicin plus coffee fruit extract at doses of 100, 200, and 400 mg/kgBW. Parameters assessed included renal histopathological changes (HE staining) and serum TNF-α levels (ELISA). Results: Administration of coffee fruit extract significantly improved renal tubular morphology and reduced TNF-α levels compared to the negative control group (p<0.05). The most protective effect was observed at a dose of 200 mg/kgBW. Conclusion: Coffee fruit extract exhibits nephroprotective activity against gentamicin-induced kidney injury, as evidenced by improved histological structure and reduced TNF-α levels, suggesting its potential as a natural agent for preventing nephrotoxicity.
