Abstract

Red rice bran is a by-product produced from rice milling in Jatiluwih village, Tabanan, Bali which has secondary metabolites that have the potential to inhibit bacterial activity. The purpose of this study was to determine the potential of 96% ethanol extract of red rice bran to inhibit bacterial growth Escherichia coli. The research method started with the extraction process by maceration method using 96% ethanol solvent. The viscous extract obtained was then subjected to phytochemical screening which showed the presence of compounds flavonoid, alkaloid, and tannin. Antibacterial activity test using disc diffusion method paper disk, measured by looking at the clear zone formed and analyzed using the mean test. The antibacterial activity test results of red rice bran extract at concentrations of 20% (8.63 mm) and 25% (9.93 mm) were categorized as moderate, concentration 30% (10.33 mm) was categorized as strong inhibition. Positive control in this study using chloramphenicol 250 mg which resulted in an inhibition zone of 21.42 mm in the strong inhibition zone category. Comparison of the antibacterial activity of the 96% ethanol extract of red rice bran showed significant difference with p value <0.05.

Abstract

Red rice bran contains various secondary metabolite compounds. The phenolic compound content in red rice bran has high antioxidant activity which has the potential to be used in solid soap preparations to protect the skin from free radicals. The research aimed to formulate red rice bran extract into a solid soap preparation that has antioxidant activity. Extraction was carried out by maceration method using 96% ethanol solvent. The viscous extract obtained was subjected to phytochemical screening. Solid soap formulation with the addition of various extract concentrations of 1.5%, 2% and 2.5%. Antioxidant activity testing was carried out using the DPPH method. The research result showed that red rice bran extract had very strong antioxidant activity with an IC50 value of 18,164 ppm, then it can be formulated into soap preparations that have antioxidant activity. The highest antioxidant activity in solid soap preparations varied in concentration of 2.5% red rice bran extract (F3) with an IC50 value of 526.80 ppm, 2% concentration (F2) with an IC 50 value of 664.28 ppm, and the lowest in variations concentration of 1.5% (F1) with an IC 50 value of 5042.20 ppm. The physical quality characteristics of solid soap preparations from red rice bran extract (organoleptic characteristics, pH, free alkali, free fatty acids and foam height) met the requirement in accordance with SNI quality requirements for solid soap preparations.

Abstract

Red Rice Bran (Oryza nivara) is one of the natural ingredients that contains flavonoids, tannins, alkaloids, and terpenoids that are able to provide antibacterial activity. The purpose of this study is to determine the antibacterial activity of solid soap preparations of 96% ethanol extract of brown rice bran by cold process method against Staphylococcus aureus bacteria. The extraction process is carried out by the maceration extraction method, namely by using 96% ethanol solvent. The antibacterial activity test of the extract was carried out using Staphylococcus aureus test bacteria with variations in the concentration of ethanol extract of brown rice bran which were 1.5%, 2.5%, and 3.5%. The antibacterial activity test of solid soap of brown rice bran extract was carried out using Staphylococcus aureus test bacteria with F0, F1, F2 and F3 formulations. The method used for the antibacterial activity test was diffusion with Kirby-baurer disc paper discs using Mueller Hilton agar media. The results of the antibacterial activity test of solid soap preparations of 96% ethanol extract of brown rice bran by cold process method have antibacterial activity with moderate inhibitory power against Staphylococcus aureus bacteria. The inhibition comes from the soap base, the addition of extracts to the 2%, 2.5%, 3% concentration variations does not provide an increase in inhibition against the growth of Staphylococcus aureus bacteria.
