Abstract

Butterfly Pea Flower is one of the plants that has antioxidant activity. However, based on previous studies, the antioxidant content is highly dependent on the location and place of the BPF itself. Therefore, this study aims to measure the concentration of total flavonoids, phenolics, antioxidant and antibacterial activity in BPF so that this research can be further developed into certain products. Extraction using maceration method with 96% ethanol solvent (1:10). DPPH method for measuring antioxidant activity and the disc paper diffusion method for testing antibacterial activity. BPF contain flavonoids and total phenolics of 19.44 ± 0.060 mg Quercetin Equivalent (QE)/g and 36.37 ± 0.47 mg Gallic Acid Equivalent (GAE)/g, respectively. BPF also contains antioxidants with an IC50 index of 49.47 µg/mL and BPF extract has no antibacterial activity using the paper disc diffusion method. This antioxidant-rich BPF extract has a lot of promise for development into a cosmetic preparation with more research.  Keywords: Butterfly Pea Flower, Antioxidant, Antibacterial, Flavonoid, Phenolic.

Abstract

Free radicals are highly reactive molecules that can cause cellular damage and contribute to the development of various degenerative diseases. Butterfly pea flower (Clitoria ternatea L.) is a traditional medicinal plant rich in antioxidant compounds such as flavonoids, anthocyanins, and phenolics. This study aimed to evaluate the antioxidant activity of infusion and decoction preparations of butterfly pea flowers using the DPPH method. The research was conducted through laboratory experiments by measuring the IC?? value using UV-Vis spectrophotometry. The results showed that the infusion extract exhibited very strong antioxidant activity with an IC?? value of 31.25 ppm, while the decoction extract demonstrated moderate antioxidant activity with an IC?? value of 128.66 ppm. The positive control, Vitamin C, showed very strong antioxidant activity with an IC?? value of 14.04 ppm. Based on these findings, the infusion method is more effective at extracting antioxidant compounds than the decoction method. The infusion of butterfly pea flower has potential to be developed as a natural antioxidant source.

Abstract

Butterfly pea flower (Clitoria ternatea) is rich in anthocyanins with recognized antioxidant activity; however, its application in spray-based sunscreen formulations and its effect on photoprotective performance remain insufficiently explored. This study aimed to formulate and evaluate a sunscreen spray containing butterfly pea extract (BPE) in combination with ethylhexyl methoxycinnamate and to determine the optimal extract concentration based on antioxidant activity, SPF value, and formulation stability. Five formulations were prepared: F0 (without BPE), F1 (0% BPE + 0.8% BHT), and F2–F4 containing 1%, 2%, and 3% BPE, respectively. Evaluations included DPPH radical scavenging activity, SPF determination, antibacterial testing, and physical quality assessment. The antioxidant activity of BPE in spray formulations showed moderate activity, with IC₅₀ values of 90.53±1.0, 87.50±1.9, and 86.94±1.4 μg/mL for 1%, 2%, and 3% concentrations, respectively, compared to 49.90±0.9 μg/mL for the pure extract. Although the antioxidant activity in formulations was lower than the pure extract due to dilution effects, increasing BPE concentration significantly improved SPF values (27.36±2.7; 29.42±1.1; 31.98±0.9; p<0.05). No significant difference was observed between 2% and 3% concentrations in both antioxidant and SPF evaluations. Therefore, 2% BPE was considered the optimal concentration, offering comparable efficacy with improved material efficiency for potential industrial production. All formulations met acceptable physical quality parameters, while antibacterial activity was not observed. This study demonstrates the feasibility of incorporating C. ternatea extract into sunscreen spray systems and provides a scientific basis for developing plant-based antioxidant-enhanced photoprotective formulations.

Abstract

Background: Butterfly pea flower (Clitoria ternatea) contains high levels of anthocyanins with potent antioxidant activity, making it a promising natural ingredient for sunscreen formulations. However, the skin safety of sunscreen spray containing this extract must be evaluated before cosmetic application. Objective: This study aimed to evaluate the skin irritation potential of sunscreen spray formulations containing butterfly pea flower extract using the patch test method. Methods: A patch test was conducted on 15 male Wistar rats. The test materials included sunscreen spray formulations with butterfly pea flower extract at concentrations of 1% (F1), 2% (F2), and 3% (F3). A base formulation without extract (F0) served as a negative control, and sodium lauryl sulfate served as a positive control. Erythema and edema scores were observed at 24, 48, and 72 hours after application. Results: The mean erythema and edema scores for formulations F1, F2, and F3 were 0.0 ± 0, indicating no signs of skin irritation in all tested groups. The positive control group showed slight erythema (score 2.33 ± 0.5), confirming the validity of the test method. Conclusion: Sunscreen spray formulations containing butterfly pea flower extract at concentrations of 1%, 2%, and 3% did not induce skin irritation in male Wistar rats under the patch test conditions. These findings support the potential of butterfly pea flower extract as a safe natural ingredient for sunscreen products.
