Abstract

            The study aimed to increase the sperm quality of polled Bali bulls with the supplementation of bean sprout (Phaseolus radiatus L.) in feeding. This research was used two polled Bali bulls aged 5-6 years old and supplemented with bean sprout 1 kg/bull two times in a week for two months. Semen was collected using artificial vagina method. The evaluation macroscopic of semen was volume while the microscopic evaluation such as motility, viability, abnormality, plasma membrane integrity (PMI), acrosome integrity, and DNA fragmentation. The result of this research showed that semen volume and spermatozoa abnormality of fresh semen after feeding of bean sprouts were not significantly difference (P>0.05) compared to before feeding, but motility and viability were significantly higher (P<0.01). Motility and acrosome integrity of frozen semen were not significantly difference (P>0.05) while viability and PMI were significant higher (P<0.01), abnormality and DNA Fragmentation were significant lower (P<0.01) after feeding compared to before. The research revealed that the supplementation of bean sprouts could increase the motility and viability but had no effect on volume and abnormality of fresh semen. Meanwhile in frozen semen, it increased viability and PMI and decreased abnormality and DNA fragmentation but had no effect on motility and acrosome integrity.

Abstract

Advances in semen quality analysis technology are developing rapidly. Computer-based semen analysis technology provides a more objective assessment of spermatozoa motility by measuring various kinematic movements of spermatozoa cells. This study aimed to analyze differences in spermatozoa movement patterns using Computer-Assisted Semen Analysis (CASA) in various goat breeds. This study used frozen semen from four goat breeds: Boer, Ettawa crossbred (PE), Saanen, and Anglo-Nubian. Parameter observations were conducted on thawed frozen semen from two straw samples for each group, consisting of six replicates from each breed. The observed sperm kinetic movement characteristics were VAP, VCL, VSL, DAP, DCL, DSL, STR, LIN, WOB, ALH, and BCF. The results showed that sperm movements did not differ significantly (P0.05) in VAP, VCL, VSL, DAP, DCL, and WOB. There were significant differences in DSL, the highest in Saanen 21.993.01 m/second, the highest STR and LIN values in Boer 0.690.04% and 0.450.03%, the longest ALH Anglo Nubian 6.190.39 m, and the largest BCF Boer 25.561.87 Hz. This study concludes that differences in goat breeds affect the kinematic movement of spermatozoa as measured by CASA in the parameters of DSL, STR, LIN, ALH, and BCF.

Abstract

Sapi Donggala adalah sapi lokal asli Sulawesi Tengah. Sapi Donggala memiliki keunggulan lebih dibanding sapi lainnya, di antaranya daya tahan panas yang lebih baik, adaptasi lingkungan dan reproduksi yang baik yaitu tetap dapat bereproduksi meskipun dalam kondisi tubuh yang buruk (skor kondisi tubuh yang rendah). Pemeriksaan hematologis sangat penting untuk dilakukan dalam memantau dan menganalisis kesehatan dari suatu kondisi fisiologi ternak. Penelitian ini bertujuan untuk mengetahui kondisi hematologis darah dari induk ternak sapi donggala dengan skor kondisi tubuh yang berbeda. Induk Sapi Donggala yang dijadikan sebagai ternak percobaan adalah induk Sapi Donggala dari peternakan rakyat yang ada di Kabupaten Sigi, Desa Sibalaya. Berdasarkan hasil penelitian status kondisi tubuh (SKT) berpengaruh nyata terhadap parameter hematologis induk sapi Donggala. Induk sapi dengan SKT >2 memiliki jumlah leukosit, eritrosit, hemoglobin, dan hematokrit secara signifikan lebih tinggi dibandingkan induk sapi dengan SKT <2 (P<0,01), serta jumlah trombosit yang berbeda nyata (P<0,05). Hal ini menunjukkan bahwa semakin baik status kondisi tubuh, maka semakin optimal pula kesehatan darah dan fungsi fisiologis sapi Donggala, terutama dalam mendukung daya tahan tubuh, kapasitas angkut oksigen, dan potensi koagulasi.
