Abstract

Background: Candida albicans is a common opportunistic pathogen that play a significantrole in Oral candidiasis and mucosal carcinogenesis. Several studies founD that Candida sp.associated with dysplastic alteration in the epithelium. Candida present in oral cavity withepithelial change is one of predisposing factor of candida infection and lead to atypia to dysplasia. Immunosupression condition is predisposition factor of oral candidal infection.Objective: To investigate the degree of epithelial dysplasia in tounge induced by Candidaalbicans in immunosuppred condition. Methods: This study was true experimental withposttest-only control group design. Sixteen healthy male Rattus novergicus (Wistar), 12 weeksand 200-250 g weight, were immunosuppressed through oral administration ofdexamethasone and tetracycline for 21 days and induced with C.albicans (ATCC-10231) 1Mc.Farland. The subjects were divided into 4 groups (n=4/group): healthy (HG),immunosuppressed (IG), Immunosupressed+Candida albicans (ICG), and nystatintreatment(NG). The subjects were treated for 14 days, later the rats were euthanized and theirtongue being biopsied. Epithelial dysplasia was subjected to HE examination, observed undera microscope (400x magnification) and statistically analyzed (Kruskall wallis, Mann withey, p<0.05). Result: Epithelial dysplasia of IG was the heaviest than other group. No eptithelialchange were found in group HG. Significant differences existed between HG with othergroups; IG with ICG; ICG with NG (p<0.05). No significant differences was present betweenIG and NG (p > 0.05). Conclusion: There was epithelial change in the tongue both inimmunosupressed condition with or without induction Candida albicans. Degree of epithelialdysplasia in tounge induced by Candida albicans in immunosupressed condition group wasthe heaviest than other group.

Abstract

Background: The denture base made of acrylic resin has a porosity and water absorbtion characteristic to cause the deposition of food waste, that problem was known to cause a growth of Candida albicans microorganisms. This problem can be solved by providing a coating (Surface coating) made of natural polymer (cellulose acrtate) of sea sea grass (Enhalus acoroides) mixed with hydrocoloid material so as to provide protection properties. Such protective properties can inhibit the bonding between the residual monomer results of acrylic resins and microorganisms. Objective: To analyze and compare the number of Candida albicans colonies on the acrylic resin plate that has surface coating and no surface coating. Method: Using post test only group design, using 12 samples divided into 2 groups. Group 1 is the control group (Heat cured acrylic plate contaminated with Candida albicans). Group 2 : Treatment group (Heat cured acrylic plate with surface coating made from sea grass Enhalus acoroides then contaminated with Cndida albicans). Calculating the number of colonies from Candida albicans was done by calculating the turbidity of SDB media (Saboraud Dextrose Broth) using UV-VIS Specrofotometer with 560 λ wave. Data analysis using an Independent T-Test. Results: Acrylic resin plates with surface coating showed significant differences in the number of Candida albicans colonies. The average results in the control group obtained an average number of colonies of 1,58 CFU/ml, while in the treatment group as much as 1,42 CFU/ml. Conclusion: The results show that surface coating can decrease the amount of Candida albicans colonies because it can close the microporosity, prevent a water absorption, and prevent the formation of the pellicle.

Abstract

Background: Diabetes mellitus (DM) is a metabolic disorders characterized by hyperglycemia caused by oxidative stress. Porphyromonas gingivalis (P.g) is a periodontopathogen caused periodontitis exacerbate blood glucose levels (BGL). Hyperbaric oxygen therapy (HBOT) combined with stichopus hermanii (SH) has a role as an antioxidant and antibacterial agent. Purpopse: To analysis effect of HBOT, SH powder and combination of both to the level of blood glucose diabetic Wistar rat induced P.g. Materials and Methods: This study was pre-test-post-test control group design, using 20 male Wistar rat were divided into 5 groups, Group1 (STZ), Group2 (STZ+P.g), Group3 (STZ+P.g+HBOT), Group4 (STZ+P.g+SH powder), Group5 (STZ+P.g+HBO+SH). STZ induction intravenous for 4 days in a row. P.g. induction was exposure 3 times for 4 days. After therapy (7 days), sample blood tail vein of rat were taken for measure BGL by glucometer. Result: Level of blood glucose K1(280.25±88.786)mg/dl, K2(372.25±135.662)mg/dl, K3(249.50±96.652)mg/dl, K4(172.00±82.077)mg/dl, K5(104.75±38.257)mg/dl. Conclusion: Induction of STZ increased the BGL but induction of P.g. cannot exacerbate BGL . HBOT combined with SH powder can reduce BGL in STZ and P.g. induction group.

Abstract

Background: Oral candidiasis is common opportunistic infection in oral cavity caused by the fungal pathogen Candida albicans and it can develop to malignant such as a candida leukoplakia if it’s affected by cigarette smoke exposure. Purpose: To analysis the effectivity of Stichopus hermanii supplementation of catalase activity in Wistar rats submandibular with oral candidiasis. Materials and Methods: This study was post-test-only control group design, using 35 male Wistar rats were divided into 5 groups, Group1 is negative control, Group2 is positive control, Group3 is treatment control with Stichopus hermanii 0,0225 mg/g BB powder, Group4 is treatment control with Stichopus hermanii 0,045 mg/g BB powder) Group5 is treatment control with Stichopus hermanii 0,09 mg/kg BB powder). Oral candida infection was achieved by spray and cotton swab rolled over all parts of the mouth. After treatment, rats were euthanized and submandibular were biopsied to make histopathology slide for measure activity of enzim catalase with spectrophotometer λ 280 nm. Data were analyzed using One Way ANOVA and Tukey LSD test. Results: There are significantly differences catalase activity between K1&K2, K2&K3, K4&K1, K2&K4, K2&K5. Conclusion: Suplementation of Stichopus hermanii effectiv of catalase activity in wistar rat’s submandibular with Candida albicans A infection.
