Abstract

Carbon tetrachloride (CCl4) is a volatile liquid that can cause hepatotoxicity in metabolism, because it produces Reactive Oxygen Species (ROS) which can trigger inflammation and cell dysfunction. Gandaria fruit (Bouea macrophylla Griffith) is known to contain a number of antioxidant compounds that function as free radical scavengers. This study aims to determine the effect of administering ethanol extract of gandaria fruit (Bouea macrophylla Griffith.) on the morphology and histology of the spleen of white rats (Rattus norvegicus L.) induced by carbon tetrachloride (CCl4). This study used RAL using 25 rats divided into 5 groups, Negative controls was only given food and drink, Positive controls was given CCl4 1mL/kgBW Treatment P1, P2, P3, was given CCl4 1 mL/kgBW + gandaria fruit extract with doses of P1 (100 mg/kgBW), P2 (150 mg/kgBW), P3 (200 mg/kgBW). CCl4 administration was carried out every 3 days for 30 days and gandaria fruit extract was administered daily starting from day 8 to day 30.The stages in this study include qualitative phytochemical screening tests, antioxidant activity tests using the DPPH method, macroscopic analysis of the morphological condition of the spleen organ and histological examination of the spleen organ using the paraffination and HE staining methods. Data analysis used the one-way ANOVA test and continued with Duncan's advanced test. The results showed that administration of gandaria fruit ethanol extract did not affect the morphology of the spleen organ of white rats induced by carbon tetrachloride (CCl4) including the edge shape, color, surface, texture, weight, length and width. However, induction of carbon tetrachloride at a dose of 1 mL/kgBW can damage the histological condition of the spleen tissue including bleeding and necrosis and widening the diameter of the white pulp, the diameter of the germinal center and the marginal zone. Administration of gandaria fruit ethanol extract in the P3 treatment at a dose of 200 mg/kgBW was the most effective dose in repairing spleen organ tissue.

Abstract

This study aimed to evaluate the effect of gandaria fruit ethanol extract on IL-6 expression in the brain tissue of CCl₄-induced rats. A completely randomized experimental design was employed using 20 male white rats (Rattus norvegicus L.), which were assigned to five groups: normal control (KN), positive control (K+), treatment group 1 (P1) receiving 100 mg/kgBW of gandaria fruit ethanol extract, treatment group 2 (P2) receiving 150 mg/kgBW, and treatment group 3 (P3) receiving 200 mg/kgBW. IL-6 gene expression in brain tissue was analyzed using reverse transcription polymerase chain reaction (RT-PCR), followed by band intensity and relative density analyses. The relative density analysis revealed that the greatest reduction in IL-6 gene expression occurred in the P1 treatment group administered 100 mg/kgBW of gandaria fruit ethanol extract, with a fold change value of 2.5. These findings suggest that gandaria fruit ethanol extract may attenuate the inflammatory response induced by CCl₄ exposure through the suppression of IL-6 expression, with the 100 mg/kgBW dose exhibiting the most pronounced reduction based on relative density analysis.

Abstract

This study aimed to evaluate the effects of Mikania micrantha Kunth leaf extract on erythrocyte count, hemoglobin concentration, hematocrit, leukocyte count, and platelet count in male rats. A completely randomized design (CRD) was employed using 20 male rats randomly allocated into five experimental groups. The normal control group received no treatment for 30 days, whereas the positive control group was exposed to cigarette smoke from three cigarettes per day for 30 days. The treatment groups (P1, P2, and P3) were exposed to the same amount of cigarette smoke for 30 days and were administered Mikania micrantha Kunth leaf extract at doses of 100, 200, and 300 mg/kg body weight, respectively, for 23 days. The experimental procedures included phytochemical screening and measurement of hematological parameters, including erythrocyte count, hemoglobin concentration, hematocrit, leukocyte count, and platelet count, using a hematology analyzer. The data were analyzed using one-way analysis of variance (ANOVA), followed by Duncan’s multiple range test. The results showed that administration of Mikania micrantha Kunth leaf extract significantly affected erythrocyte count, hemoglobin concentration, and hematocrit (p < 0.001), as well as leukocyte count (p < 0.001) and platelet count (p = 0.001). Among the tested doses, 300 mg/kg body weight produced hematological parameter values that most closely approximated those of the normal control group.

Abstract

Smoking exerts detrimental effects on the body, notably by elevating oxidative stress, which, if not properly managed, may contribute to various pathophysiological conditions, including cardiovaskular. Boroko leaves are recognized as a traditional herbal remedy believed to possess therapeutic potential for multiple ailments; however, their application in healthcare requires scientific validation to confirm their efficacy. This study was conducted to analyze the phytochemical content of the ethanol extract of Boroko leaves (EDB) and to evaluate its effects on hematological parameters in cigarette smoke–exposed white rats. A randomized experimental design was employed, consisting of five treatment groups with five replications each: K1 (normal control), K2 (cigarette-exposed control), K3 (cigarette + EDB 100 mg/kg BW), K4 (cigarette + EDB 200 mg/kg BW), and K5 (cigarette + EDB 400 mg/kg BW). Antioksidan of EDB was evaluated using DPPH method and Hematological parameters were measured using a hematology analyzer, Data were analyzed using one-way ANOVA. The findings indicated that EDB administration was associated with modulation of erythrocyte and leukocyte counts in cigarette smoke-exposed rats. However, no significant differences were observed in hemoglobin, hematocrit, and platelet levels among the treatment groups.
