Abstract

ABSTRAKSpons merupakan salah satu komponen terumbu karang yang berpotensi sebagai senyawa bioaktif namun belum banyak dimanfaatkan. Penelitian ini diarahkan untuk menperoleh beberapa ekstrak kasar spons dari perairan Salibabu Kepulauan Talaud, menentukan aktivitas antibakteri dari beberapa ekstrsk kasar spons serta membandingkan aktivitas antibakteri fraksi spons terhadap bakteri uji E. coli dan S. aureus. Hasil identifikasi spons ditemukan sebanyak tiga spesies yang terdiri dari: Siphonodictyon sp., Ircinia sp., Dysidea sp. Pengujian aktivitas antibakteri ekstrak kasar, fraksi air, metanol dan heksan menggunakan metode difusi agar dengan bakteri uji E. coli dan S. aureus. Ekstrak kasar siphonodictyon sp dan fraksi air dari ekstrak ini menunjukan aktivitas antibakteri tertinggi.Kata kunci : Spons, Antibakteri, E. coli, S. aureus

Abstract

Ecotourism as a form of tourism that emphasizes responsibility for nature conservation, provides economic benefits and maintains cultural needs for the local community. Mangrove forests with their uniqueness are natural resources that have the potential to be used as tourist attractions. The purpose of this study was to determine community structure, mangrove canopy cover and suitability of mangrove ecotourism land. This study used the line transect method for mangrove community structure, and hemispherical photography for the percentage of mangrove cover. The results of the study of the mangrove community structure were the highest density value, namely at station 1 on the transect 2 types of S.alba 0.16 ind / m2 relative 88.89% and the lowest density value in the type R. mucronata and R.apiculata 0.01 ind / m2 relatively 5.56% on transect 2 at station 1. The highest frequency type value at station 1 on the transect 1 type S. alba which is 1.00 relative 100% and the lowest frequency at station 1 transect 2 types R, piculate which is relative 0.50 33.33. The highest closing value of type on transect 2 standard 1 type S. alba 10.26 relative value 92.63%. Whereas the lowest type 3 transect closure is type R. piculate 0.49 the relative is 7.86%. The highest Important Value Index is at station 1 on transect 1, namely type S. alba 300 and lowest at station 1 on transect 2 types R. mucronata 30.55. The highest diversity value on transect 2, 0.43. Evenness index value (E) at station 1 on transect 1 is 0 and transect 2 is 0.39. While station 2 on transect 1 0 and transect 2 0.51. regarding the criteria for mangrove damage, at 2 stations included in the rare category with inya 50% canopy cover. The overall results of land suitability for coastal tourism in the Bunaken Island mangrove tourism category fall into the very appropriate category.Keywords: Mangrove, Ecotourism, Bunaken IslandEkowisata sebagai suatu bentuk wisata yang menekankan tanggung jawab terhadap kelestarian alam, memberikan manfaat secara ekonomi dan mempertahankan kebutuhan budaya bagi masyarakat setempat. Hutan mangrove dengan keunikan yang dimilikinya, merupakan sumberdaya alam yang sangat berpotensi untuk dijadikan sebagai tempat kunjungan wisata. Tujuan dari penelitian ini adalah untuk mengetahui struktur komunitas, tutupan kanopi mangrove dan kesesuaian lahan ekowisata mangrove. Penelitian ini menggunakan metode line transek terhadap struktur komunitas mangrove, dan hemispherical photography untuk persentase tutupan mangrove. Hasil penelitian struktur komonitas mangrove yaitu Nilai kerapatan tertinggi yaitu pada stasiun 1 di transek 2 jenis S.alba  0.16 ind/m2 relatifnya 88.89% dan nilai kerapatan terendah pada jenis R. mucronata dan R.apiculata 0.01 ind/m2 relatifnya 5.56% di transek 2 pada stasiun 1 .Nilai frekuensi jenis tertinggi pada stasiun 1 di transek 1 jenis S.alba yaitu 1.00 relatifnya 100% dan frekuensi terendah pada stasiun 1 transek 2 jenis R, piculate yaitu  0.50 relatifnya  33.33. Nilai penutupan jenis tertinggi pada transek 2 stasun 1 jenis S. alba 10.26 nilai relatifnya 92.63 %. Sedangkan penutupan jenis terendah transek 3 jenisR.  piculate  0.49 relatifnya 7.86%. Indeks Nilai Penting tertinggi yaitu pada stasiun 1 di transek 1 yaitu jenis S.alba 300 dan terendah pada stasiun 1 di transek 2 jenis R.mucronata 30.55.Nilai keanekaragaman tertinggi pada transek 2, 0.43. Nilai indeks kemerataan (E) pada stasiun 1 di transek 1 yaitu 0 dan transek 2 0,39.  Sedangkan stasiun 2 pada transek 1 0 dan transek 2 0.51. tentang kriteria kerusakan mangrove, pada ke 2 stasiun termasuk pada kategori jarang dengan tutupan kanopinya ≤ 50% . Hasil keseluruhan kesesuaian lahan untuk wisata pantai kategori wisata mangrove Pulau Bunaken masuk pada kategori sangat sesuai.Kata kunci: Mangrove, Ekowisata, Pulau Bunaken

Abstract

The objective of this research was to formulate a strategy for managing maritime ecotourism in the era of industrial revolution 4.0 in the city of Manado. The study was conducted at the coastal areas of the mainland of Manado City and Bunaken Island for 3 months, February - May 2019. The research method was a survey with descriptive analysis and SWOT. The results of the study obtained 4 strategy formulations (key success factors) in the context of managing maritime ecotourism in the industrial revolution era 4.0 in the city of Manado, namely: 1) Increasing law enforcement in the field of marine ecotourism, waste management on land and sea, 2) Awareness of the community environmental hygiene both on land and sea, 3) Empowering biodiversity resources of coral reefs, seagrass beds and mangroves in the park for developing marine maritime ecotourism through digital applications, and 4) managing and developing resorts, coastal culinary attractions, points of diving spots, and tourism  ports. The results of the SWOT curve show the condition of marine ecotourism in the city of Manado in quadrant 2, namely a situation where the threat to the development of marine ecotourism is more dominant than opportunity, but there are strengths of tourism organizations that can be relied on. Stakeholders are expected to be able to improve performance so that quadrant 2 conditions change to quadrant 1, which is to support an aggressive strategy: a very good situation because of the power that is used to seize profitable opportunities. In the era of industrial revolution 4.0, every stakeholder in the maritime ecotourism industry in the city of Manado was supposed to change the management system towards digital-based by making applicationsKeywords: Bunaken island, revolution industry 4.0, Manado city, marine ecotourism, strategyTujuan penelitian adalah untuk merumuskan strategi pengelolaan ekowisata bahari di era revolusi industri 4.0 di Kota Manado.  Penelitian dilakukan di wilayah pesisir daratan Kota Manado dan Pulau Bunaken selama 3 bulan, Februari - Mei 2019. Metode penelitian adalah survei dengan analisis deskriptif dan SWOT. Hasil penelitian diperoleh 4 rumusan strategi (faktor-faktor kunci keberhasilan) dalam rangka pengelolaan ekowisata bahari era revolusi industri 4.0 di Kota Manado, yaitu: 1) Meningkatkan penegakan hukum di bidang ekowisata bahari, pengelolaan sampah di darat maupun laut, 2) Menyadarkan masyarakat tentang kebersihan lingkungan baik di daratan maupun lautan, 3) Memberdayakan sumber daya keanekaragaman hayati terumbu karang, padang lamun dan mangrove di kawasan TNB untuk pengembangan ekowisata bahari melalui aplikasi digital, dan 4) Mengelola dan mengembangkan resort, tempat-tempat wisata kuliner pantai, titik-titik penyelaman, dan pelabuhan pariwisata.  Hasil kurva SWOT memperlihatkan kondisi ekowisata bahari Kota Manado berada dalam kuadran 2, yaitu situasi dimana ancaman terhadap pengembangan ekowisata bahari lebih dominan dibandingkan peluang, namun ada kekuatan organisasi kepariwisataan yang dapat diandalkan. Pemangku kepentingan diharapkan dapat meningkatkan kinerja agar kondisi kuadran 2 berubah menjadi kuadran 1, yaitu mendukung strategi agresif: situasi yang sangat baik karena adanya kekuatan yang dimanfaatkan untuk meraih peluang yang menguntungkan.  Dalam era revolusi industri 4.0, maka setiap pemangku kepentingan industri ekowisata bahari di Kota Manado sudah seharusnya merubah sistem pengelolaan yang ada ke arah berbasis digital dengan cara membuat aplikasi pengelolaan ekowisata bahari.Kata Kunci: Ekowisata bahari, revolusi industri 4.0, Kota Manado, Pulau Bunaken, strategi

Abstract

ABSTRACTBioactive compounds that are suspected of having anticancer activity are first tested for activity by means of a toxicity test. The purpose of this test is to obtain data on the ability of the activity of a bioactive compound to kill cells at small doses so as to obtain a lethal concentration or lethal data. These two measurements are often called LC50 or LD50, concentrations that can kill 50% of test animals. This study aims to test the cytotoxic activity of Thalassia hemprichii seagrass extract using the Brine Shrimp Lethality Test (BSLT) method.The results of the study showed that the mortality of Artemia salina larvae was influenced by the concentration of the test, meaning that the higher the concentration the more test animals died. Furthermore, the results of the probit analysis contained the toxicity activity of bio active compounds from seagrasses where the LC50 was 3.95 mg / l. A substance is declared to have the potential for cytotoxic activity if it has a value of LC50 at concentrations <1000 ppm for extracts and at concentrations <30 ppm for a compound. Keywords: Cytotoxic Activity, Thalassia hemprichii, Brine shrimp Lethality Test (BSLT)

Abstract

Bacteria are found to be highly associated with various marine organisms, such as sponges. Sponges are known as the product of bioactive compounds. However, some of the compounds produced by sponges are obtained by the simbion-bacteria. Therefore this situation enables sponges simbion bacteria to play a major role in producing bioactive compounds that have been isolated from the sponge. The purpose of this study is to isolate and determine the characteristics of morphology of spongy simbion bacteria resembling Cribochalina sp, which was taken from Malalayang waters, North Sulawesi. Sponge simbion bacteria grow on NB media. Initially, a free-breeding colony was being done before bacteria were accrued in the NA media by 2% sponge broth (patent SID201906301) by means of Strike Plate methods. Based on to this study we had isolate five spongy simbion bacteria that resembling the Cribochalina sp. All five of these isolations have different characteristics of morphology in terms of color, shape, size, and elevation.      Keywords : bacteria, isolation, simbionts, sponges

Abstract

The purpose of this study was to determine the type, dimensions and composition of nematocytes in Scleractinia, Acropora florida and Acropora divaricata.  This research was conducted in South Motandoi Beach, East Pinolosian District, South Bolaang Mongondow Regency, North Sulawesi for five (5) months; from February to June 2019.  The method of this research was observation using student-t statistical data analysis. Microbasic p-Mastigophore type-II (MpM-II) and Holotricous isorhiza (HI), while A. divaricata only has type-II Microbasic p-Mastigophore (MpM-II) nematocytes.  The nematocytes dimension showed an average capsule length of MpM II in A. florida of was 262.32 µm and A. divaricata was 125.15 µm, mean capsule width of MPM II in A. florida was 67.01 µm and A. divaricata was 31.95 µm, and the average length of mpM II on A. florida was 154.44 µm, and A. divaricata was 70.97 µm.  Nematocytes composition showed that MPM II in A. florida was 95.56% and HI was 4.44%, while the composition of MPM II nematocytes in A. divaricata was 100% and HI was 0%.  Student t-test showed that the ratio of 2 average values of capsule length, capsule width, and MPM II stem length from two populations of A. florida and A. divaricata were not significantly different (P> 0.05).   It can be concluded that both types of A. florida and A. divaricata corals have different types and composition of nematocytes however their size dimensions are not significantly different.  Further studies are important to examine genetic and environmental factors to find out how much the kinship relationship between the two types of those coral.Keywords: Acropora florida, Acropora divaricata, HI, Scleractinia coral, MpM II, nematocytes

Abstract

Macroalgae are chlorophyll plants, consisting of thallus and can live in polluted waters. The purpose of the study was to analyze the lead metal (Pb) in the water column and thallus red algae Kappaphycus alvarezii which was cultivated in a certain period of time in the waters of Totok Bay. The cultivation method used is the bottom method. The algae weight is measured every two weeks. Water quality parameters and lead metal analysis (PB) are carried out in the Baristan laboratory. The results obtained are algae can grow according to the time period. Water quality can affect algae growth. Lead metal content in water is detected high enough as well as in thallus algae in the 3rd week. Keywords : Kappaphycus alvarezii, Lead metal, Water quality, Totok Bay Makroalga merupakan tumbuhan yang berklorofil dan tubuhnya berupa thallus serta mampu hidup pada perairan tersemar. Penelitian ini dilakukan dengan tujuan untuk menganalisis kandungan logam timbal (Pb) dalam air dan thallus alga merah Kappaphycus alvarezii yang dibudidayakan dalam periode waktu tertentu di perairan Teluk Totok. Metode budidaya pada yang digunakan adalah metode lepas dasar (bottom method). Pengukuran berat alga dilakukan setiap dua minggu. Beberapa parameter kualitas air dan analisis logam timbal (Pb) dilakukan di Laboratorium Baristan. Hasil yang diperoleh yaitu alga dapat tumbuh sesuai dengan periode waktu. Kualitas air dapat mempengaruhi pertumbuhan alga.Kandungan logam timbale pada air terdeteksi cukup tinggi demikian juga pada thallus alga di minggu ke-3. Kata kunci : Kappaphycus alvarezii, Logam Timbal, Kualitas air, Teluk Totok

Abstract

Ascidians are sesile marine invertebrates that have bioactive compounds such as antibacterial and anti-UV. The purpose of this study is to determine the antibacterial activity against Escherichia coli and Staphylococcus aureus bacteria and to test the anti-UV activity of the ascidian water fraction. Antibacterial activity test was carried out using the diffusion method (disc diffusion Kirby & Bauer) and the water fraction of ascidian that showing antibacterial activity tested in a UV spectrophotometer to see the anti-UV activity. Results of the study were obtained 4 types of ascidians extracted from partitioned into water fraction, n-hexane fraction, methanol fraction. All three fractions were tested for antibacterial activity and the result showed that there were antibacterial activities of ascidian extract Clavelina sp. against both test bacteria with inhibition of S. aureus and E. coli with strong categories. Ascidian Phlebobranch sp. showed the presence of antibacterial activity with inhibition of S. aureus and E. coli with very strong categories (16,6 mm). extract Eudistoma sp. showed the presence of antibacterial activity with inhibition S. aureus and E. coli in the medium category (9 mm). The water fraction found active in antibacterial testing is then tested using a UV spectrophotometer for anti UV testing, the result show that water fraction of the four ascidians can absorb UV-B (290-320nm) and UV-A ( 320-400nm).Keywords : Ascidian, Antibacterial, Partition, Anti- UV.

Abstract

Sponge organisms produce bioactive compounds that are toxic as a means of self-defense. The compound is known to have the potential as an antibacterial and anti-UV which can absorb sunlight with the potential to be used as a material for making sunscreen. The purpose of this study was to obtain antibacterial activity against Escherichia coli and Staphylococcus aureus bacteria and anti-UV activity from crude extracts and sponges fractions. The antibacterial test done by agar diffusion method (Kirby and Bauer diffusion disc) and the crude extract and the active fraction of antibacterial compounds were tested in a UV spectrophotometer to see its anti-UV activity. As a result, 4 species of sponges were extracted and partitioned into water fractions, methanol fractions, and n-hexane fractions. All samples were tested for antibacterial activity and the results showed antibacterial activity against S. aureus by Plakortis sp. in crude extract (9 mm) and water fraction (8.6 mm), Agelas sp. in crude extract (7 mm) and in E. coli bacteria shown by Plakortis sp. in crude extract (12.6 mm) and water fraction (9 mm), Liosina sp. in the water fraction (7.6 mm), Haliclona sp. in the water fraction (8 mm) and Agelassp. in crude extract (10.3 mm). Crude extracts and water fractions were tested using a UV spectrophotometer for anti-UV testing, the results showed that crude extract and all the water fractions of four species sponge could absorb UV-B (λ 290-320 nm) and UV-A (λ 320-400 nm).Keywords: sponges, antibacterial, partition, anti-UV

Abstract

This study is intended to find out the fish that are cultured into the mangrove area of Tasik Ria beach, Mokupa, Minahasa Regency. Fish is obtained through gill jarring at high tide. The fish that were identified were Archamia fucata, Lutjanus fulvus, Lutjanus eherenbergii, Lethrinus ornatus, Scolopsis lineate, Sargocentron diadema, Epinephelus merra and Scorpaenopsis oxycephala. The fish are generally carnivorous, and are nocturnal.Key words: Tasik Ria Beach, the fishes, carnivorous, mangroves
