Abstract

Genetic Transformation and Expression of Sucrose Phosphate Synthase Mutant in Tomato Plant ABSTRACTSucrose phosphate synthase (SPS) is a key enzyme responsible for sucrose biosynthesis. In its regulation, SPS activity is modulated by an allosteric effector glucose-6-phosphate (G6P) suggested to have an ability to bind SPS N-terminus domain. To understand the role of N-terminus in regulating SPS, the SPS gene was mutated with the deletion of N-terminus domain (∆N-SPS). The ∆N-SPS gen was transformed into tomato plants with 5% transformation efficiency. Three transgenic tomato plant 4.20, 5.5.1, and 5.10 were obtained and confirmed by PCR analysis. Transgenic tomato expression was characterized by enzymatic analysis. Result showed that the G6P allosteric regulation in transgenic ∆N-SPS had lost and the SPS activity increased by 2-fold compared to non-transgenic plant. This showed that N-terminus domain-deleted SPS could be actively expressed in plant. Keywords: enzyme, genetic transformation, N-terminus domain deletion, sucrose phosphate synthase, tomato ABSTRAKSucrose phosphate synthase (SPS) merupakan enzim kunci yang bertanggung jawab dalam sintesis sukrosa. Dalam regulasinya, aktifitas SPS dipengaruhi oleh alosterik efektor glukosa-6-fosfat (G6P) yang diduga dapat berikatan pada domain N-terminus SPS. Untuk mengetahui peran N-terminus pada regulasi SPS, dilakukan mutasi SPS dengan penghilangan domain N-terminus (∆N-SPS). Gen ∆N-SPS diinsersi pada tanaman tomat melalui transformasi genetik dengan efisiensi transformasi 5%. Tiga tanaman transgenik tomat (event4.20; 5.5.1; dan 5.10) didapatkan dan positif terkonfirmasi melalui analisis PCR. Ekspresi mutan dikarakterisasi melalui analisis enzimatik. Hasil menunjukkan bahwa tanaman tomat transgenik ∆N-SPS tidak dipengaruhi regulasi alosterik G6P dan aktifitas SPS 2 kali lipat lebih tinggi daripada tanaman bukan transgenik. Ini menunjukkan bahwa SPS dengan delesi domain N-terminus dapat terekspresi aktif pada tanaman.  Kata Kunci: delesi domain N-terminus, enzim, sucrose phosphate synthase, tomat, transformasi genetik

Abstract

In Vitro Somatic Embryogenesis and Plantlet Regeneration of Three Varieties of Alfalfa (Medicago sativa L.)ABSTRACTAlfalfa (Medicago sativa L.) is a valuable plant as a source of food for animal, forage, pharmaceutical, medicine, food supplement, and human consumption.  In vitro selection technology combined with induction or spontaneous mutagenesis has been effective in altering or isolating genetic variability for desirable characters.  Consequently, a reproducible in vitro propagation technique of that plant is mandatory. The aim of the research was to obtain information on the embryogenic callus induction, somatic embryogenesis, and plantlet regeneration of three varieties of alfalfa. The results showed that an optimum embryogenic callus induction (82%) was obtained on Murashige & Skoog (MS) basal medium containing 2 ppm 2,4-dichlorophenoxyacetic acid (2,4-D), 2 ppm kinetin and 2 ppm a-naphthaleneacetic acid (NAA). Those embryogenic calli could subsequently develop into somatic embryos, which germinated and regenerated into normal plantlets on R1 medium consisting of MS nutrients without the addition of plant growth regulator.Keywords: alfalfa, callus, embryogenic, plantlets, regeneration ABSTRAKAlfalfa (Medicago sativa) adalah tanaman berharga sebagai sumber makanan untuk hewan, yaitu hijauan pakan ternak, farmasi, obat-obatan, suplemen makanan dan konsumsi manusia. Teknologi seleksi in vitro yang dikombinasikan dengan induksi atau mutagenesis spontan telah terbukti efektif dalam mengubah atau mengisolasi variabilitas genetik untuk karakter yang diinginkan. Oleh sebab itu, keberhasilan teknik perbanyakan in vitro yang telah terbukti dapat direproduksi dari tanaman tersebut menjadi syarat yang harus terpenuhi. Tujuan dari penelitian ini adalah untuk mendapatkan informasi mengenai induksi kalus embriogenik, embriogenesis somatik dan regenerasi planlet dari tiga varietas alfalfa. Hasil penelitian menunjukkan bahwa induksi kalus embriogenik optimal (82%) didapat pada media Murashige & Skoog (MS) dengan  2 ppm 2,4-dichlorophenoxyacetic acid (2,4-D), 2 ppm kinetin dan 2 ppm a-naphthaleneacetic acid (NAA). Kalus embriogenik tersebut dapat membentuk embrio somatik, embrionya berkecambah dan beregenerasi membentuk planlet normal pada perlakuan media R1 yaitu nutrisi MS tanpa penambahan zat pengatur tumbuh.Kata Kunci: alfalfa, embriogenik, kalus, planlet, regenerasi

Abstract

Comparison of Three RNA Extraction Kits for Transcriptome Analysis of Oil Palm (Elaeis guineensis Jacq.) ABSTRACTObtaining high-quality RNA is very important at an early stage of molecular biology research. To isolate RNA, high skill and caution are required in following laboratory procedures because RNA is easily degraded, especially samples from plant tissue culture. One of the parameters used to check the total RNA quality is RIN (RNA Integrity Number). The aim of this study was to obtain RNA extraction methods on oil palm leaves, callus and somatic embryos that were of good quality and high concentrations for transcriptomic analysis. RNA extraction was carried out using Plant RNA PureLink (Ambion), Genezol RNA Extraction (Geneaid) and RibospinTM Plant (Geneall) kit methods. The results showed that oil palm leaf, callus and somatic embryo RNA were successfully extracted using the RibospinTM (Geneall) kit. Based on the total RNA number of more than 4 μg and the RIN value of more than 7, the extracted RNA could be used in RNA sequencing for transcriptomic analysis. Keywords: callus, oil palm, RNA analysis, RNA quality, somatic embryo ABSTRAKMenghasilkan RNA berkualitas tinggi sangatlah penting pada tahap awal penelitian biologi molekuler. Untuk mengisolasi RNA diperlukan keterampilan dan kehati-hatian tinggi dalam mengikuti prosedur di laboratorium karena RNA lebih mudah terdegradasi, khususnya sampel hasil kultur jaringan tanaman. Salah satu parameter yang digunakan pada pengecekan kualitas RNA total adalah RIN (RNA Integrity Number). Penelitian bertujuan mendapatkan metode ekstraksi RNA pada daun, kalus dan embrio somatik kelapa sawit yang berkualitas baik dan memiliki konsentrasi tinggi untuk analisa transkriptomika.  Ekstraksi RNA dilakukan menggunakan metode kit Plant RNA PureLink (Ambion), Genezol RNA Extraction (Geneaid) dan RibospinTM Plant (Geneall). Hasil menunjukkan bahwa RNA daun, kalus dan embrio somatik kelapa sawit telah berhasil diekstraksi dengan menggunakan kit RibospinTM (Geneall). RNA hasil ekstraksi tersebut dapat digunakan untuk sekuensing RNA dengan tujuan analisis transkriptomika, dilihat dari jumlah total RNA yang lebih dari 4 μg dan nilai RIN lebih dari 7. Kata Kunci: analisis RNA, embrio somatic, kalus, kelapa sawit, kualitas RNA

Abstract

The Effect of Basal Media and NAA on the In Vitro Induction of Oil Palm (Elaeis guineensis Jacq.) RootABSTRACTClonal propagation of oil palm plants using tissue culture technique results in a low percentage of rooted shoots. To increase the percentage of rooted shoots that are more uniform, the root induction method is supported by the use of basic media and the addition of growth regulators 1-naphthaleneacetic acid (NAA). This study aims to analyze the effect of a combination of base media and optimum NAA concentration in inducing the roots of oil palm shoots in vitro. This research used factorial completely randomized design (RAL) consisting of 2 factors. The first factor was the type of basic media, namely Murashige and Skoog (MS) and MS Modifications (MSM) media. The second factor was the concentration of NAA, namely 0; 0.05; 0.1; and 0.2 ppm. each treatment was repeated 10 times. The results showed that the use of MSM medium was better than that of MS, and the most optimum NAA concentration was 0.05 and 0.1 ppm, in inducing oil palm roots in vitro. In addition, the combination of MSM + NAA 0.1 ppm treatment produced the most optimum result in induction of oil palm roots in vitro.Keywords: basal media; NAA; palm oil; plantlet; root inductionABSTRAKPerbanyakan klonal tanaman kelapa sawit menggunakan teknik kultur jaringan menghasilkan persentase tunas berakar yang rendah. Dalam upaya meningkatkan persentase tunas berakar yang lebih seragam maka dilakukan metode induksi akar yang didukung oleh penggunaan media dasar dan penambahan zat pengatur tumbuh 1-naphthaleneacetic acid (NAA). Penelitian ini bertujuan untuk menganalisis pengaruh kombinasi media dasar dan konsentrasi NAA yang optimum dalam menginduksi akar tunas kelapa sawit secara in vitro. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) faktorial yang terdiri dari 2 faktor. Faktor pertama adalah jenis media dasar yang terdiri dari media Murashige and Skoog (MS) dan MS Modifikasi (MSM). Faktor kedua adalah konsentrasi NAA yang terdiri dari 0; 0,05; 0,1; dan 0,2 ppm. setiap perlakuan diulang sebanyak 10 kali. Hasil penelitian menunjukkan bahwa penggunaan media MSM lebih baik daripada MS, dan konsentrasi NAA yang paling optimum adalah 0,05 dan 0, 1 ppm dalam menginduksi akar kelapa sawit secara in vitro. Selain itu kombinasi perlakuan MSM+NAA 0,1 ppm memiliki hasil yang paling optimum dalam induksi akar kelapa sawit secara in vitro.

Abstract

Influence of Biofloatation Technique through Rhizopus sp. Fermentation on the Nutritional Quality of the Floating Fish FeedABSTRACTSolid fermentation using the mold Rhizopus has been used as an alternative method to improve the physical quality of fish feed, namely stability in water and floatability. Although the fermented fish feed produced had been shown previously to have better stability in water and floatability, the effect of the fermentation on the fish feed nutrition has not yet been known. This study aimed to determine the effect of solid fermentation using the mold Rhizopus sp. on the nutrient content of the fermented feed. In addition, the effect of adding tapioca as much as 0, 1, 2, 3, and 4 g to the dry weight loss and density of the fermented feed was investigated. The results showed that the fermented feed contained higher levels of ash and protein than that before fermentation. The addition of tapioca up to 4 g had no significat effect (p>0.01) on the dry weight loss of the fermented feed, but tended to increase its density compared to those without tapioca addition.Keywords: density, fermentation, fish feed, nutrition, Rhizopus ABSTRAKFermentasi padat menggunakan kapang Rhizopus telah digunakan sebagai metode alternatif untuk memperbaiki kualitas fisik pakan ikan, yakni stabilitas dalam air dan daya apung. Meskipun pakan ikan fermentasi yang dihasilkan sebelumnya sudah dibuktikan memiliki stabilitas dalam air dan daya apung yang lebih baik, namun pengaruh fermentasi terhadap nutrisi pakan ikan tersebut belumlah diketahui. Penelitian ini bertujuan mengetahui pengaruh fermentasi padat menggunakan kapang Rhizopus sp. terhadap kandungan nutrisi pakan fermentasi, dan pengaruh penambahan tapioka sebanyak 0, 1, 2, 3, dan 4 g terhadap kehilangan berat kering dan mass jenis pakan hasil fermentasi. Hasilnya menunjukkan bahwa pakan fermentasi mengandung kadar abu dan protein lebih tinggi dibandingkan sebelum difermentasi. Penambahan tapioka hingga 4 g tidak berpengaruh nyata (p>0,01) pada kehilangan berat kering pakan fermentasi, namun cenderung meningkatkan massa jenis dibandingkan tanpa penambahan tapioka.Kata kunci: fermentasi, massa jenis, nutrisi, pakan ikan, Rhizopus

Abstract

Phytochemical Screening and Analysis of Traditional Herbal Medicines of Bima DistrictBima people use medicinal plants for traditional medicine which is local wisdom that must be preserved. Thus, the phytochemical content of these plants needs to be studied. This study aims to determine the secondary metabolite content of traditional medicinal plants in order to enrich biotechnology and pharmacological data. Thus, the data can be used as a reference in healing diseases and managing various medicinal plants based on community welfare and environmental sustainability for the people of Bima regency. The method used is a detailed interview and a field survey followed by phytochemical screening to determine the content of compounds in plants. From the results of this study, as many as 17 types of medicinal plants have been used as traditional medicine. The plant parts used are leaves, stems, flowers, roots, rhizomes, fruit, gum or lenders with 33% weed habitus followed by herbs (29%), trees (29%), and shrubs (9%). The compounds identified in these medicinal plants are flavonoids, alkaloids, steroids, terpenoids, saponins, and tannins.Keywords: Bima regency; local wisdom; medicinal plants; phytochemicals; secondary metabolitesABSTRAKMasyarakat Bima memanfaatkan tumbuhan obat untuk pengobatan tradisional yang merupakan kearifan lokal yang harus dipertahankan, sehingga kandungan fitokimia dari tumbuhan-tumbuhan ini perlu diteliti. Penelitian ini bertujuan untuk mengetahui kandungan metabolit sekunder tumbuhan obat tradisional guna memperkaya data bioteknologi dan farmakologi. Dengan demikian, data tersebut dapat dijadikan acuan dalam penyembuhan penyakit dan pengelolaan berbagai tumbuhan obat berbasis kesejahteraan masyarakat dan kelestarian lingkungan bagi masyarakat kabupaten Bima. Metode yang digunakan adalah wawancara secara rinci dan survei lapangan dilanjutkan dengan skrining fitokimia untuk mengetahui kandungan senyawa dalam tumbuhan. Dari hasil penelitian ini diketahui sebanyak 17 jenis tumbuhan obat yang telah dimanfaatkan sebagai obat tradisional. Bagian tumbuhan yang digunakan berupa daun, batang, bunga, akar, rimpang, buah, getah atau lender dengan jenis habitus 33% gulma diikuti oleh herbal (29%), pohon (29%), dan perdu (9%). Sedangkan senyawa yang teridentifikasi dalam tumbuhan obat tersebut yaitu flavonoid, alkaloid, steroid, terpenoid, saponin, dan tannin.

Abstract

The Effect of Ionic Silver Nanoparticles Concentrations on the Inhibition of Lactobacillus casei and pH of MilkABSTRACTTechnological developments support the creation of various nanoparticles that can be utilized, one of which is silver nanoparticles. This study aims to determine the effect of ionic silver nanoparticle concentration on Lactobacillus casei growth inhibition, pH change and curdling of UHT milk. Variation in the concentration of ionic silver nanoparticles of 5, 10, 15, and 20 ppm was tested for their characteristics through UV-VIS spectrophotometry, showing the wave peaks at 413–430 nm, and through the PSA test resulting in the Z-average of 87.2 nm. Ionic silver nanoparticles were tested against L. casei using the Kirby-Bauer diffusion method for 72 hours. The test results showed a change in the clear zone that tended to increase for the concentration of silver nanoparticles 5, 10, 15 and 20 ppm with the gradient values of 0,0032; 0,0280; 0,0395; dan 0,0317, respectively. Tests for pH and UHT milk curdling were carried out for 78 hours and showed that ionic silver nanoparticles tended to be more able to maintain milk pH ±30 hours and curdling ±24 hours longer than milk that was not treated with ionic silver nanoparticles.Keywords: electrolysis; Lactobacillus casei; milk; nanoparticles; silver ABSTRAKPerkembangan teknologi mendukung terciptanya berbagai nanopartikel yang dapat dimanfaatkan, salah satunya nanopartikel perak. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi nanopartikel perak ionik terhadap daya hambat Lactobacillus casei, kadar pH, dan penggumpalan pada susu UHT. Variasi konsentrasi nanopartikel perak ionik 5, 10, 15, dan 20 ppm diuji karakteristiknya melalui spektrofotometri UV-VIS dengan hasil puncak gelombang serapan berada pada kisaran 413–430 nm dan uji PSA dengan hasil Z-average berada pada 87,2 nm. Sampel larutan nanopartikel perak ionik diujikan terhadap bakteri L. casei menggunakan metode difusi Kirby-Bauer selama 72 jam. Hasil uji menunjukkan terjadinya perubahan zona bening yang cenderung meningkat untuk konsentrasi nanopartikel perak 5, 10, 15 dan 20 ppm dengan masing-masing nilai gradiennya 0,0032; 0,0280; 0,0395 dan 0,0317. Pengaruhnya terhadap kadar pH dan penggumpalan susu UHT dilakukan selama 78 jam dan menunjukkan bahwa nanopartikel perak ionik cenderung lebih mampu mempertahankan pH susu ±30 jam dan penggumpalan ±24 jam lebih lama dibanding susu yang tidak diberikan perlakuan nanopartikel perak ionik.
