Abstract

Genetic Transformation and Expression of Sucrose Phosphate Synthase Mutant in Tomato Plant ABSTRACTSucrose phosphate synthase (SPS) is a key enzyme responsible for sucrose biosynthesis. In its regulation, SPS activity is modulated by an allosteric effector glucose-6-phosphate (G6P) suggested to have an ability to bind SPS N-terminus domain. To understand the role of N-terminus in regulating SPS, the SPS gene was mutated with the deletion of N-terminus domain (∆N-SPS). The ∆N-SPS gen was transformed into tomato plants with 5% transformation efficiency. Three transgenic tomato plant 4.20, 5.5.1, and 5.10 were obtained and confirmed by PCR analysis. Transgenic tomato expression was characterized by enzymatic analysis. Result showed that the G6P allosteric regulation in transgenic ∆N-SPS had lost and the SPS activity increased by 2-fold compared to non-transgenic plant. Untuk mengetahui peran N-terminus pada regulasi SPS, dilakukan mutasi SPS dengan penghilangan domain N-terminus (∆N-SPS). Gen ∆N-SPS diinsersi pada tanaman tomat melalui transformasi genetik dengan efisiensi transformasi 5%. Tiga tanaman transgenik tomat (event4.20; 5.5.1; dan 5.10) didapatkan dan positif terkonfirmasi melalui analisis PCR. Ekspresi mutan dikarakterisasi melalui analisis enzimatik. Hasil menunjukkan bahwa tanaman tomat transgenik ∆N-SPS tidak dipengaruhi regulasi alosterik G6P dan aktifitas SPS 2 kali lipat lebih tinggi daripada tanaman bukan transgenik. Abstract

Marigold flower (Tagetes erecta L.) is an ornamental flowering plant that belongs to the Astarecae family. Marigold flowers are used as ornamental plants in pots and as flower bouquets. The demand for marigold flowers is very high, so efforts are needed to quickly propagate marigold plants and produce good quality. Efforts to provide marigolds are carried out by increasing the production and quality of marigold plants. Tissue culture is an in vitro technique of plant propagation and is effective in producing large, uniform and disease-free plants. Giving ZPT 2,4-D and Kinetin can stimulate good callus growth and affect the success of callus induction. This study aims to determine the effect of giving 2,4-D and kinetin on the growth of marigold callus induction. The experiment used a randomized block design consisting of two factors, namely 2, 4-D with four concentration levels, namely 0 ppm, 0.5 ppm, 1 ppm and 2 ppm and kinetin with four concentration levels, namely 0 ppm, 0.5 ppm, 1 ppm and 2 ppm with three replicates. The results showed that the combination treatment without 2,4-D and 0.5 ppm kinetin gave the best response when callus appeared 11.67 HST and the treatment of 1 ppm 2,4-D and 2 ppm kinetin gave the best response on the percentage of successful callus induction marigolds.

Abstract

The purpose of this research is to determine the income from watermelon farming as seen from production in the Ringinanom sub-district, Nganjuk District, Nganjuk Regency. The research method used in this research is a quantitative method which is strengthened by descriptive qualitative methods by conducting R/C ratio analysis. Based on the results of the research described previously, the income from watermelon farming in Ringinanom Village, Nganjuk District, Nganjuk Regency was Rp. 2,132,000,. The results of the R/C Ratio analysis are 1.34, which means the farming business is worth pursuing. The BEP Product value is 480 Kg, and the BEP Price is IDR. 3,742

Abstract

This Community Service Activity aims to empower women entrepreneurs in Nganjuk Regency through training in processing banana heart and catfish into shredded fish. This program was initiated to improve the skills and economic income of women's groups, as well as to optimize the utilization of abundant but underutilized local resources. The criteria for women entrepreneurs who participate in this program are 1) women entrepreneurs who are members of the Nganjuk Regency UMKM, 2) have an interest in catfish processing and 3) women entrepreneurs who play an active role in developing the creative economy. The methods used include analysis of needs problems, technical training in raw material processing, business management, and shredded fish product marketing strategies, monitoring and evaluation. This activity was carried out at the Village Hall of Kauman Village, Nganjuk District, Nganjuk Regency. The results of this activity showed a significant increase in technical skills, business knowledge, and income of the women's groups involved. In addition, participants were able to learn business management and marketing strategies that helped them develop their businesses professionally and sustainably.. The success of this activity confirms that appropriate training and empowerment can produce sustainable positive changes and become a model that can be applied in other areas.

Abstract

Kokedama merupakan teknik bercocok tanam pada tanaman hias yang memilik tipe perakaran serabut dengan menggunakan metode bungkus (wrap) tanpa menggunakan pot. Pembalutan cukup menggunakan media tanam seperti tanah, lumut atau sabut kelapa yang dibentuk seperti bola dan dilakukan pengikatan menggunakan tali rami. Pemberdayaan masyarakat melalui pelatihan pembuatan tanaman hias dengan teknik Kokedama di Lingkungan UPT PPSAA Kabupaten Nganjuk merupakan upaya yang bertujuan untuk meningkatkan kratifitas dan inisiasi wirausaha bagi lingkungan UPT PPSAA utamanya bagi para remaja dan pengurus yang berada di lingkungan tersebut. Pentingnya pengetauan dasar terkait kewirausahaan bagi remaja di lingkungan UPT PPSAA dapat dijadikan lifeskill yang nantinya dapat digunakan meningkatkan pendapatan secara ekonomi bagi remaja di lingkungan UPT PPSAA. Kegiatan pengabdian masyarakat ini dilakukan dalam bentuk sosialisasi dan melakukan pelatihan langsung dan sesi evaluasi bersama peserta pelatihan yang berlokasi di UPT PPSAA Kab. Nganjuk. Hasil dari adanya pengabdian masyarakat ini bertambahnya pengetahuan, wawasan dan keterampilan dari remaja dan pengurus dalam membuat tanaman hias kokedama.

Abstract

Rejoso Rejoso sub-district in Nganjuk district is known as a high-productivity shallot center. One of the important factors affecting crop yields is the irrigation system used. The increasing scarcity of water resources due to climate change and growing intersectoral demand has made irrigation a strategic issue in the national agricultural sector. This study aims to compare the effectiveness of conventional irrigation systems with sprinkle irrigation in shallot (Allium cepa L.) cultivation. The method used was a one-factor Completely Randomized Design (CRD) with five treatments, namely with a water volume of P1 = Manual watering, P2 = 75%, P3 = 100%, P4 = 150%, P5 = 200% of optimal needs. Parameters observed included plant height, number of leaves per clump, and tuber wet weight. Results showed that treatment P5 (200%) of water requirement and conventional watering gave the best growth and production results. The sprinkle irrigation system proved to be more efficient in water use, provided even water distribution, and reduced labor requirements. Thus, sprinkle irrigation can be an appropriate technology solution to support shallot cultivation as well as efficient and sustainable water management.

Abstract

Water is an important component in shallot cultivation activities, optimal water availability is very necessary in shallot cultivation activities, especially in the early stages of growth and bulb formation. Traditional irrigation methods tend to be wasteful and unpublished in air distribution which can result in decreased yields, especially in cultivation activities carried out in areas with limited air. Sprinkler irrigation appears as a potential solution to optimize air distribution through a spraying mechanism that allows for more even air distribution. The research design used was a one-factorial Completely Randomized Design with five replications. Irrigation carried out in this study was in the form of traditional control 100% manual irrigation needs (I0), sprinkler 75% manual irrigation volume (I1), sprinkler 100% manual irrigation volume (I2), sprinkler 150% manual irrigation volume (I3) and sprinkler 200% manual irrigation volume (I4) which were given once in the morning at 06.00 -07.00. This study aims to analyze the effect of the use of a sprinkler irrigation system on shallot productivity. The results showed that sprinkler irrigation with a volume of 200% of the manual method gave the best results on plant height, number of tillers, dry weight of the stump, and diameter of the shallot bulb. In contrast, the traditional irrigation method with 100% manual volume produced the lowest bulb weight loss. Sprinkler application with 200% manual irrigation volume can significantly increase shallot production.

Abstract

The development of Teaching Factory as an educational landscape Tourism is a strategic step to bring education as a synchronization of learning activities that refers to Merdeka Belajar Kampus Merdeka. Tourism is related to the beauty and relationship of the landscape between its elements. Perceptions and preferences for beauty are needed as a basis for developing a tourist area. The scenic beauty estimation (SBE) method will enable you to evaluate a landscape. Time is one of the landscape elements that will enhance the character of the landscape. The worst landscape is Landscape 19, which was the parking lot for the Department of Language, Communication, and Tourism. The perception of the respondent's memory that states that the Health Department is the landscape with the best value is based on the time when the picture was taken during the day, which is the perfect time in terms of lighting, shadows, and composition. Another thing is that the health department is an icon of Politeknik Negeri Jember.
