Abstract

ABSTRACTInvertases (beta-D-fructofuranosidase, E.C. 3.2.1.26) are the keyenzymes involved in sucrose metabolism in sugarcane plants. They are highly correlated with sucrose and reducing sugar contentsduring plant growth. The sugarcane plants have two kindsof invertases, namely neutral invertase (NI) and acid invertase(AI). They have different function in sucrose accumulation. The research aimed to study the role of AI and NI in accumulationof reducing sugar during storage of sugarcane stems. Plantmaterials of 18-month-old field grown sugarcane of the commercialvariety R-579 (wet-land) and M 442-51 (dry-land) wereused. Three internodes were sampled to represent immature(internode 1-8/F1), maturing (internode 9-16/F2), and mature(internode 17-24/F3) stem tissues. All tissues were stored for0-9 days at room temperature (28-32oC) and each day, the samplewas extracted to determine invertase activity, total solubleprotein, and sugar contents. This observation was valid forinvertase activity expressed on a protein basis. At the initiateharvested (0-3 days), NI had a higher specific activity than AIin the sucrose-accumulating region of the sugarcane stems.Negative significant correlation was found between NI specificactivity and sucrose accumulation (r2 = 0.41, P < 0.05). AIshowed a higher specific activity after 4 days harvested and hadnegative correlation with sucrose accumulation (r2 = 0.40, P <0.05). These results showed that NI could be more responsiblein sucrose hydrolisis than AI at early storage of sugarcane stems.

Abstract

Sucrose transporter (SUT) is a protein that functions to translocate sucrose from source to sink. Protein activity is determined by the accumulation of sucrose in sugarcane stem. The level of sucrose accumulation is also influenced by the activity of  sucrose phosphat synthase (SPS) and neutral invertase (NI) in conjunction with the synthesis and hydrolysis of sucrose. This aim of this study is to determine the sucrose transporter protein expression in Genetically Modified Product (GMP) of sugarcane.Genetically Modified Product GMP sugarcane events 1, 2, 3, 4, 5, 18, 20 and non GMP were used as materials. The analysis was performed by Western Blot method to observe the presence of SUT protein expression, stem sucrose content by resorcinol method to observe the translocation of sucrose, the SPS enzyme activity and NI that play a role in determining the accumulation of sucrose. The results showed that (1) an increase in SUT protein content in GMP plants events 2 and 18 with a protein band that is thicker than the non GMP plants, (2) SPS and invertase activity at all events GMP crop plants to increase compared to non GMP. Sucrose content of the stem at all events GMP plant to increase with age segment. The older age segments, the higher the sucrose  content. Keywords: sugarcane (Saccharum officinarum L.), genetically modified product(GMP), sucrose transporter protein(SUT), sucrose

Abstract

The application of crashworthiness technology to a vehicle has a main objective of protecting the passenger or cargo from the effects of impact/collision. One of the strategies is by using impact energy absorbing (IEA) modules. Two of the alternatives of IEA modules are metallic tubes with internal inversion and controlled axial splitting mechanism. This paper presents both numerical and experimental approaches to understand the characteristics of the two mechanisms, before using them in design phase. LS-Dyna was used as the numerical simulation software for drop test case. The result of simulation is presented as the relationships between geometrical parameters and the crashworthiness parameters, e.g. impact energy and response force. Some cases are compared with quasi-static and drop test results.

Abstract

Abstract

Genetic Transformation and Expression of Sucrose Phosphate Synthase Mutant in Tomato Plant ABSTRACTSucrose phosphate synthase (SPS) is a key enzyme responsible for sucrose biosynthesis. In its regulation, SPS activity is modulated by an allosteric effector glucose-6-phosphate (G6P) suggested to have an ability to bind SPS N-terminus domain. To understand the role of N-terminus in regulating SPS, the SPS gene was mutated with the deletion of N-terminus domain (∆N-SPS). The ∆N-SPS gen was transformed into tomato plants with 5% transformation efficiency. Three transgenic tomato plant 4.20, 5.5.1, and 5.10 were obtained and confirmed by PCR analysis. Transgenic tomato expression was characterized by enzymatic analysis. Result showed that the G6P allosteric regulation in transgenic ∆N-SPS had lost and the SPS activity increased by 2-fold compared to non-transgenic plant. This showed that N-terminus domain-deleted SPS could be actively expressed in plant. Keywords: enzyme, genetic transformation, N-terminus domain deletion, sucrose phosphate synthase, tomato ABSTRAKSucrose phosphate synthase (SPS) merupakan enzim kunci yang bertanggung jawab dalam sintesis sukrosa. Abstract

Sucrose content is an important component of yield in sugarcane (Saccharum officinarum L.) juice and itssynthesis is determines by the activity of Sucrose phosphate synthase (SPS; EC. 2.4.1.14). This enzyme catalyzes thepenultimate reaction of the formation of sucrose from fructose-6-phosphate (F6P) and uridine-5-diphospho glucose(UDPG). Sucrose is synthesized on leaf and translocated into stalk at where it is accumulated to be accumulatedit. In contrast to SPS, acid invertase (AI) is an enzyme that hydrolyze sucrose to glucose and fructose. The abilityof sugarcane to accumulate the sucrose in stalk is determined by the differences between SPS and AI activities. This study was aimed know role of the internodes SPS and AI in sucrose accumulation in the internodes of somesugarcane varieties. SPS, AI and sucrose were extracted from internodes 1, 3 and 5 of 9 month old sugarcane. Theresults showed that sucrose content for all varieties increased concomitant with the rise in the age of internodes. The decrease in AI activity in internode caused the increase in sucrose content in internodes. Meanwhile, highersucrose content in the older internodes was not followed by higher in SPS activity. Generally, there was strongnegative correlation between sucrose content of internodes with SPS and AI activity. In conclusion, the ability ofsugarcane to accumulate sucrose at internodes was primary determined by AI activity, while SPS activity had asmall role on sucrose accumulation at internodes.
